Background: We recently characterized a specific inorganic triphosphatase (PPPase) from Nitrosomonas europaea. This enzyme belongs to the CYTH superfamily of proteins. Many bacterial members of this family are annotated as predicted adenylate cyclases, because one of the founding members is CyaB adenylate cyclase from A. hydrophila. The aim of the present study is to determine whether other members of the CYTH protein family also have a PPPase activity, if there are PPPase activities in animal tissues and what enzymes are responsible for these activities. Methodology/Principal Findings: Recombinant enzymes were expressed and purified as GST- or His-tagged fusion proteins and the enzyme activities were determined by measuring the release of inorganic phosphate. We show that the hitherto uncharacterized E. coli CYTH protein ygiF is a specific PPPase, but it contributes only marginally to the total PPPase activity in this organism, where the main enzyme responsible for hydrolysis of inorganic triphosphate (PPPi) is inorganic pyrophosphatase. We further show that CyaB hydrolyzes PPPi but this activity is low compared to its adenylate cyclase activity. Finally we demonstrate a high PPPase activity in mammalian and quail tissue, particularly in the brain. We show that this activity is mainly due to Prune, an exopolyphosphatase overexpressed in metastatic tumors where it promotes cell motility. Conclusions and General Significance: We show for the first time that PPPase activities are widespread in bacteria and animals. We identified the enzymes responsible for these activities but we were unable to detect significant amounts of PPPi in E. coli or brain extracts using ion chromatography and capillary electrophoresis. The role of these enzymes may be to hydrolyze PPPi, which could be cytotoxic because of its high affinity for Ca2+, thereby interfering with Ca2+ signaling. [less ▲]

The renewed interest in donation after cardio-circulatory death (DCD) started in the 1990s following the limited success of the transplant community to expand the donation after brain-death (DBD) organ supply and following the request of potential DCD families. Since then, DCD organ procurement and transplantation activities have rapidly expanded, particularly for non-vital organs, like kidneys. In liver transplantation (LT), DCD donors are a valuable organ source that helps to decrease the mortality rate on the waiting lists and to increase the availability of organs for transplantation despite a higher risk of early graft dysfunction, more frequent vascular and ischemia-type biliary lesions, higher rates of re-listing and re-transplantation and lower graft survival, which are obviously due to the inevitable warm ischemia occurring during the declaration of death and organ retrieval process. Experimental strategies intervening in both donors and recipients at different phases of the transplantation process have focused on the attenuation of ischemia-reperfusion injury and already gained encouraging results, and some of them have found their way from pre-clinical success into clinical reality. The future of DCD-LT is promising. Concerted efforts should concentrate on the identification of suitable donors (probably Maastricht category III DCD donors), better donor and recipient matching (high risk donors to low risk recipients), use of advanced organ preservation techniques (oxygenated hypothermic machine perfusion, normothermic machine perfusion, venous systemic oxygen persufflation), and pharmacological modulation (probably a multi-factorial biologic modulation strategy) so that DCD liver allografts could be safely utilized and attain equivalent results as DBD-LT. We discuss the case of two patients who developped delayed malignant peripheral nerve sheath tumor (MPNST) following radiotherapy. Case report: the first patient is a sixty year-old woman with a 2 years history of right cervicobrachial pain and weakness. The neurological examination depicted severe weakness, atrophy and loss of sensation in the right C5 and C6 territories. A subclavicular hardened and enlarged lymph node was noted. Her past medical history was notable for a Hodgkin’s disease (HD) treated with radiation therapy (>40Gy) 35 years earlier. Brachial plexus MRI revealed a tumoral mass arising from the right brachial plexus. Biopsy of the subclavicular mass revealed a poorly differentiated malignant tumour consisting of spindle cells showing moderate polymorphism and a high mitotic index. Immunohistochemistry showed positivity for the S-100 protein, the CD56 and for the epithelial membrane antigen (EMA) and a diagnosis of MPNST of the brachial plexus was proposed. The second case is a 36 year-old man referred for a history of right sciatic neuralgia that appeared 3 years earlier. The medical history of the patient was notable for a right seminoma, treated by orchidectomy and prophylactic radiotherapy (24 Gy) 5 years earlier. The neurologic examination revealed right L5 and S1 radicular territories involvement, and the CT of the pelvis demonstrated a nodular mass at the level of the greater sciatic foramen. A surgical biopsy was performed and the neuropathological findings were consistent with a diagnosis of low-grade MPSNT. Discussion: MPNSTs are rare tumors accounting for 3 to 10% of all tissues sarcomas. Half of the cases described are sporadic, while the other half tend to appear in patients suffering from tumor prone conditions, such as neurofibromatosis type 1. Although secondary neoplasms are known complications of radiotherapy, descriptions of peripheral nerve sheath tumors (PNST) are scarce. The exact pathophysiology of radiation-induced PNSTs remains unclear but vascular alterations, direct damages to axon or Schwann cell and nerve compression by soft tissue fibrosis are thought to play a role. Although surgical removal sometimes followed by chemotherapy is the mainstay of MPNSTs, they usually carry a poor prognosis. Our 2 cases emphasize that the possibility of radiation-induced MPNST has to be kept in mind when investigating a localized neuropathy in a previously irradiated area. [less ▲]

Health is a particularly important concern in the today’s society as expenses linked to health problems are continuously increasing. Actions aiming to limit the phenomenon are needed and one of the priorities consists to fight against the increasing prevalence of obesity. In this field, the emphasis has been set on nutrition and sedentariness. It clearly appears that school and physical education are key sectors that should be directly involved in the approach that should be implemented (Young & William, 1989; WHO, 2001). At school, any project that is focused on any aspect of the health would require the involvement of all actors: (1) the school principal who is in charge of the development of the school’s project and to orient the strategic decisions, and; (2) all other actors of the school life such the pedagogical staff but also the students and their parents as well as the medical structures if they exist. When it is question of promotion of a long life physically active lifestyle, the Scientific Statement from the American Heart Association Council (Pate et al., 2006) underlines five main dimensions that should be considered in order to determine the emphasis that one particular school provides: (1) physical education; (2) active transportation; (3) life environment (recesses, spaces); (4) sport at school, and; (5) physical activity in the classroom. We are considering that the presence of physical activity in the written educational school policy represents a sixth determining dimension that should be accessed in that perspective. In the analysis of 54 out of the 514 French speaking schools of Belgium, we showed that the mean score reached 9/20 with PA policy being the worst aspect (4.80/20) and facilities being the best (12.3/20). It seemed that the role that schools could play today in PA promotion is quite limited. These findings have been confirmed in another study based on 1997 Cale’s model of active school (Van Hoye, Motter & Cloes, 2009). On the other hand, a third study underlined that it was possible to improve the involvement of one primary school in PA promotion through one action research (Cloes & Dequenne, 2009). In the latter study, OPHEA (2009) model of active school was considered. Finally, other examples of initiatives aiming to increase school in the development of an active lifestyle will be described (L’école bouge, 2006). As stated by Tappe & Burgeson (2004), physical education should be considered as the corner stone of PA promotion at school. In fact, the physical education teacher is definitely the only teacher whose specific preparation allows developing theoretical and practical concepts related to physical activity and its effects on health. Beyond the fight against sedentariness, PE teachers are able to contribute effectively to the mastery of many other competences involved in the development of physically educated persons (NASPE, 1992). Nevertheless, in order to meet the requirements, it is needed that PE respects two current concepts: literacy and accountability. It means that PE has to develop attitudes, knowledge and competences that would be used lifelong by the students, on one hand; to bring concrete modifications in youths, on the other hand. Several principles should be applied to encourage students to adopt an active lifestyle: (1) being a model; (2) updating ones’ knowledge; (3) knowing ones’ students; (4) planning; (5) acting; (6) controlling. Theoretical foundations and examples will be provided in the presentation. Based on Cloes, Del Zotto and Motter (2009), several categories of specific actions are proposed by PE teachers in order to promote an active life style in their students. These actions aim to motivate students to be active, increase time on task or attendance or to encourage students to open their mind to a more positive attitude towards PA. As they mentioned, appreciating PE might be a first step in the direction of a lifelong active lifestyle In conclusion, we consider that PE teachers need to become aware about the change in the definition of their role. In 40 years, they passed from teaching gymnastics to teaching sports while the society asks them today to become agents of the development of lifelong PA competences. It means that, during initial or continuous preparation, PE teachers’ educators need to develop original activities that can help practitioners to develop their own actions on the field. According to the current development of the research on teacher education, this underlines the importance of collaborative research as well as the development of communities of practice. Strategies are existing and should be shared. Use of modern tools of communication represents a promising way to reach success. It should also be determining that professionals and researchers would learn to work together more closely. [less ▲]

OBJECTIVE: Over the last decade, the Cambridge Neuropsychological Test Automated Battery (CANTAB), which comprises visuo-spatial tasks, has been utilized in cognitive studies of schizophrenia. A clear approach concerning the usage of CANTAB for the appraisal of neurocognitive dysfunction in schizophrenia is currently lacking. METHOD: In this paper, we have first reviewed the overall applications of CANTAB and then evaluated methodological strengths and weaknesses of CANTAB as a neurocognitive battery for schizophrenia. We carried out a systematic search and assessment of studies where CANTAB was utilized to measure cognitive function in schizophrenia. We have also attempted to quantify the available data and perform a meta-analysis, but this approach turned out to be still premature. RESULTS: CANTAB has enabled researchers to highlight significant deficits affecting broad cognitive domains in schizophrenia, such as working memory, decision-making, attention, executive functions and visual memory. So far, the most consistent deficit observed with CANTAB testing has been attentional set-shifting, suggestive of fronto-striatal dysfunctions. In addition, preliminary evidence points towards the potential use of CANTAB to identify cognitive predictors of psychosocial functioning, to describe the relationships between symptoms and cognition, and to measure the impact of pharmacological agents on cognitive functioning. CONCLUSION: CANTAB has been used successfully to highlight the range of visuo-spatial cognitive deficits in schizophrenia, producing similar results to those obtained with some traditional neuropsychological tests. Further studies validating the use of CANTAB following the standard set by Measurement And Treatment Research to Improve Cognition in Schizophrenia (MATRICS) are warranted. [less ▲]

Synthesis of Bi2-xPbxSr2Ca2Cu3O10-y (x = 0 and x = 0.3) has been performed using the glass recrystallization route technique. After splat quenching, the samples were annealed at 820, 850 or 870-degrees-C for 96 or 144 h (with one intermediary grinding). High-resolution polarization micrographs and electrical (and magnetic susceptibility) measurements have been taken. Various growth mechanisms can thus be checked, and the physical properties are studied in relation to the microstructure. It is found that the 2223 phase does not crystallize directly from the amorphous one but results from a dissolution-reprecipitation process. The stability of the 2212 phase is emphasized. The 2201 phase precipitates at high temperature and rapidly reacts with the melt to form the 2223 phase. The kinetics of the lead-substituted sample seems to be controlled by the Ca2PbO4 phase. which acts as a flux. [less ▲]
