Hydroponics has the main advantage of providing easy access to the root system and is therefore commonly used for gene expression analyses in molecular studies of the model plant Arabidopsis thaliana. However, the impact of root substrate on plant growth remains poorly documented. Here we show that hydroponics accelerates both shoot growth and developmental phases as compared with culture on soil. In order to identify molecular changes in the roots that could account for these medium effects, a transcriptomic comparison was performed by microarray analysis. This experiment revealed that more than 20% of the genes were differentially expressed in hydroponics vs soil. Among them, the flowering time gene FLOWERING LOCUS C and two clades of microRNA targeted genes. To further assess the role of these genes in roots, artificial microRNAs were designed for root specific expression in transgenic Arabidopsis plants. Their advantages as hosts for proteins production include correct post-translational modifications, low cost of maintenance and no risk of contamination by human pathogens. Targeting heterologous proteins to the extracellular space is required for the correct folding of complex proteins and makes harvesting and purification easier. However, the quantity and the quality of recombinant proteins have been proved to be reduced by the action of endogenous co-secreted proteases. In this study, we characterized root-secreted proteases in the model plant Arabidopsis thaliana, at the activity and expression levels. Their activity was analyzed by in vitro degradation of a target protein (Bovine Serum Albumine, BSA) in a range of pH and in the presence of several proteases inhibitors. Serine proteases were identified as the major protease class involved in the degradation of BSA under all tested conditions. As a first step towards the identification of the key players, the expression level of selected members of this class was analyzed by quantitative RT-PCR in roots and leaves. Les éleveurs dont les troupeaux ont des moyennes de production égales ou supérieures à 8.000 kg tirent un meilleur parti de l’environnement (effet positif) que ceux dont les troupeaux ont des moyennes de production inférieures à 7.000 Kg. Only a few objects were discovered, and many non-detections reported and statistically interpreted. The field is now in need of a technological breakthrough. Aim. Our aim is to open a new search space with first-generation systems such as NACO at the Very Large Telescope, by providing ground-breaking inner working angle (IWA) capabilities in the L' band. The L' band is a sweet spot for high contrast coronagraphy since the planet-to-star brightness ratio is favorable, while the Strehl ratio is naturally higher. Methods. An annular groove phase mask (AGPM) vector vortex coronagraph optimized for the L' band, made from diamond subwavelength gratings was manufactured and qualified in the lab. The AGPM enables high contrast imaging at very small IWA, potentially being the key to unexplored discovery space. Results. Here we present the installation and successful on-sky tests of an L'-band AGPM coronagraph on NACO. Using angular differential imaging, which is well suited to the rotational symmetry of the AGPM, we demonstrated a \Delta L' > 7.5 mag contrast from an IWA ~ 0".09 onwards, during average seeing conditions, and for total integration times of a few hundred seconds. Quelles sont les raisons de ce rapprochement rapide ? Quels sont les acteurs des relations belgo-allemandes après 1945 ? Est-ce qu’on peut parler d’une « réconciliation » au niveau sociétal ? Quelle était la position de la Belgique face à la réunification de 1990 ? Les réponses à ces questions ne se retrouvent pas seulement dans la « grande Histoire » des hommes d’État et diplomates, mais également dans l’histoire régionale de l’espace frontalier belgo-allemand.
