[less ▲]

Steam explosion is a thermo-mechanicochemical pretreatment which allows the breakdown of lignocellulosic structural components by the action of heating, formation of organic acids during the process, and shearing forces resulting in the expansion of the moisture. Two distinct stages compose the steam-explosion process: vapocracking and explosive decompression which include modification of the material components: hydrolysis of hemicellulosic components (mono- and oligosaccharides released), modification of the chemical structure of lignin, and modification of the cellulose crystallinity index, etc. These effects allow the opening of lignocellulosic structures and influence the enzymatic hydrolysis yield of the material. In the present study, four RIA systems (RIA-1 to -4) based on two antisera raised against ovine pregnancyassociated glycoproteins (ovPAGs), combined with an ovine or a bovine PAG tracer were used to measure PAG concentrations in plasma and milk samples of dairy ewes. Blood and milk samples were collected on different days of gestation: 0, 18, 20, 22, 25, 28, 32, 42, and 49. From day 20 onward, the PAG in plasma could be detected in all pregnant ewes using the four RIA systems. By using milk, except for RIA-1, the other systems showed a sensitivity of 100% from day 28 of gestation onward. In plasma, PAG concentrations were higher in multiple than in single pregnancies, while no clear relationship was observed in milk. In conclusion, milk is a good alternative to plasma for early pregnancy diagnosis in sheep from day 28 to day 42. [less ▲]

Objective: To date, only a small number of epidemiological studies on myelofibrosis have been performed. The current study aimed to characterize the myelofibrosis patient population in Belgium according to predefined disease parameters (diagnosis, risk categories, hemoglobin ,10 g/dl, spleen size, constitutional symptoms, platelet count, myeloblast count), with a view to obtaining a deeper understanding of the proportion of patients that may benefit from the novel myelofibrosis therapeutic strategies. Methods: A survey was used to collect data on prevalence and disease parameters on all myelofibrosis patients seen at each of 18 participating hematologic centers in 2011. Aggregated data from all centers were used for analysis. Analyses were descriptive and quantitative. Results: A total of 250 patients with myelofibrosis were captured; of these, 136 (54%) were male and 153 (61%) were over 65 years old. One hundred sixty-five (66%) of myelofibrosis patients had primary myelofibrosis and 85 (34%) had secondary myelofibrosis. One hundred ninety-three myelofibrosis patients (77%) had a palpable spleen. About a third of patients (34%) suffered from constitutional symptoms. Two hundred twenty-two (89%) myelofibrosis patients had platelet count§50 000/ml and 201 (80%) had platelet count §100 000/ml. Of 250 patients, 85 (34%) had a myeloblast count §1%. Six (2%) patients had undergone a splenectomy. Thirteen (5.2%) patients had undergone radiotherapy for splenomegaly. Conclusions: The results of this survey provide insight into the characteristics of the Belgian myelofibrosis population. They also suggest that a large proportion of these patients could stand to benefit from the therapies currently under development. [less ▲]

Gathering user feedback on provisional design concepts early in the design process has the potential to reduce time-to-market and create more satisfying products. Among the parameters that shape user response to a product, this paper investigates how design experts use sketches, physical prototypes, and computer-aided design (CAD) to generate and represent ideas, as well as how these tools are linked to design attributes and multiple measures of design quality. Eighteen expert designers individually addressed a two-hour design task using only sketches, foam prototypes, or CAD. It was found that prototyped designs were generated more quickly than those created using sketches or CAD. Analysis of 350 crowdsourced responses to the resulting designs showed that those created as prototypes were perceived as more novel, more aesthetically pleasing, and more comfortable to use. It was also found that designs perceived as more novel tended to fare poorly on all other measured qualities. [less ▲]

DUSP3 is a small dual-specificity protein phosphatase with an unknown physiological function. We report that DUSP3 is strongly expressed in human and mouse monocytes and macrophages and that its deficiency in mice promotes tolerance to lipopolysaccharide (LPS)-induced endotoxin shock and to polymicrobial septic shock following cecal ligation and puncture. By using adoptive transfer experiments, we demonstrate that resistance to endotoxin is macrophage-dependent and transferable and that this protection is associated with a striking increase of M2-like macrophages in DUSP3-/- mice in both the LPS and cecal ligation and puncture models. We show that the altered response of DUSP3-/- mice to sepsis is reflected in decreased TNF production and impaired ERK1/2 activation. Our results demonstrate that DUSP3 plays a key and non-redundant role as a regulator of innate immune responses by mechanisms involving the control of ERK1/2 activation, TNF secretion and macrophage polarization. A systematic study on the presence of bottlenose dolphins (Tursiops truncatus) in the Río Negro Estuary (RNE), Patagonia Argentina, was carried out between the months March – July from 2008 to 2011. Data on the dolphin’s activity patterns were gathered via an Ad Libitum focal group sampling mode accompanying dorsal fin images taken for identification and re-identification of individuals. A total effort of 188 h resulted in 58 h of positive observation of 124 dolphin groups (sightings per unit effort (SPUE) = 0.66 groups/h). Data analysis showed two main activity states for the observed groups, travelling (65%) and foraging (26%). The remaining 9% of the groups were involved in other activity states. The photo-identification effort, which started opportunistically in 2006, resulted in a catalogue of 21 individual dolphins, with a total mean re-identification rate of 9 days (max = 24 days). When comparing these pictures to the existing catalogue of Bahía San Antonio (approximately 200 km west from the study area) dorsal fins of 20 individuals could be positively matched and most (n = 17) could be subsequently re-identified in both areas, indicating their long distance movements along the North Patagonian coast during the austral autumn months. This season coincides with the lowest amount of feeding activity observed in Bahía San Antonio. This study suggests that bottlenose dolphins enter RNE, mainly during autumn, to forage. It appears that the search for food resources may be the trigger for their movement patterns along the North Patagonian coast during this season, at least for certain individuals. More research is needed to accurately confirm this hypothesis. [less ▲]

The flight of barnacle geese at airspeeds representing high-speed migrating flight is investigated using experiments and simulations. The experimental part of the work involved the filming of three barnacle geese (Branta Leucopsis) flying at different airspeeds in a wind tunnel. The video footage was analysed in order to extract the wing kinematics. Additional information, such as wing geometry and camber was obtained from a 3D scan of a dried wing. An unsteady vortex lattice method was used to simulate the aerodynamics of the measured flapping motion. The simulations were used in order to successfully reproduce the measured body motion and thus obtain estimates of the aerodynamic forces acting on the wings. It was found that the mean of the wing pitch angle variation with time has the most significant effect on lift while the difference in the durations of the upstroke and downstroke has the major effect on thrust. The power consumed by the aerodynamic forces was also estimated; it was found that increases in aerodynamic power correspond very closely to climbing motion and vice versa. Root-mean-square values of the power range from 100 W to 240 W. Finally, it was observed that tandem flying can be very expensive for the trailing bird. [less ▲]
