For the hydrological modeling of forested watersheds, the understanding of the forest hydrodynamic is essential. This study focusses on the hydrology of a Belgian forested plot with high stoniness and steep slope. The soil water content is monitored at several positions on the toposequence and at different depth. Therapeutic perspectives targeting angiogenesis in cancer stimulated an intense investigation of the mechanisms triggering and governing angiogenic processes. Several publications have highlighted the importance of typical dual-specificity phosphatases (DSPs) or MKPs in endothelial cells and their role in controlling different biological functions implicated in angiogenesis such as migration, proliferation, apoptosis, tubulogenesis and cell adhesion. However, among atypical DSPs, the only one investigated in angiogenesis was DUSP3. We recently identified this DSP as new key player in endothelial cells and angiogenesis. In this chapter we provide with detailed protocols and models used to investigate the role of DUSP3 in endothelial cells and angiogenesis. We start the chapter with an overview of the role of several DSPs in angiogenesis. We continue with providing a full description of a highly efficient transfection protocol to deplete DUSP3 using small interfering RNA (siRNA) in the primary Human Umbilical Vein Endothelial Cells (HUVEC). We next describe the major assays used to investigate different processes involved in angiogenesis such as tube formation assay, proliferation assay and spheroids sprouting assay. We finish the chapter by validating our results in DUSP3-knockout mice using in vivo angiogenesis assays such as Matrigel plug and Lewis lung carcinoma cell subcutaneous xenograft model followed by anti-CD31 immunofluorescence and ex vivo aortic ring assay. All methods described can be adapted to other phosphatases and signaling molecules. [less ▲]

Platelets are small blood cells derived from cytoplasmic fragments of megakaryocytes and play an essential role in thrombosis and haemostasis. Platelet activation depends on the rapid phosphorylation and dephosphorylation of key signaling molecules, and a number of kinases and phosphatases have been identified as major regulators of platelet function. However, the investigation of novel signaling proteins has suffered from technical limitations due to the anucleate nature of platelets and their very limited levels of mRNA and de novo protein synthesis. In the past, experimental methods were restricted to the generation of genetically modified mice and the development of specific antibodies. More recently, novel (phospho)proteomic technologies and pharmacological approaches using specific small-molecule inhibitors have added additional capabilities to investigate specific platelet proteins. In this chapter, we report methods for using genetic and pharmacological approaches to investigate the function of platelet signaling proteins. While the described experiments focus on the role of the dual-specificity phosphatase 3 (DUSP3) in platelet signaling, the presented methods are applicable to any signaling enzyme. Specifically, we describe a testing strategy that includes 1) aggregation and secretion experiments with mouse and human platelets, 2) immunoprecipitation and immunoblot assays to study platelet signaling events, 3) detailed protocols to use selected animal models in order to investigate thrombosis and haemostasis in vivo, and 4) strategies for utilizing pharmacological inhibitors on human platelets. Image de la menace par excellence, le Grand Méchant Loup est un adversaire dont on ne compte plus les apparitions, notamment dans les fictions destinées au jeune public. Dans Les Trois Petits Cochons, qui s’impose comme cas d’école parmi ces récits où surgit l’animal redouté, le loup devient l’ennemi du cochon, une créature traditionnellement associée à l’enfance. Ce tandem, qui repose sur un principe général d’opposition, brasse avec lui une série de représentations qui transparaissent en filigrane. Le détour opéré par l’histoire culturelle du cochon et quelques-uns de ses prolongements cinématographiques permet de dessiner progressivement les contours de la place occupée par le loup dans ce conte. Background: The study was conducted to evaluate the analytical and clinical performance of the VIDAS® 25-OH Vitamin D Total assay. The clinical performance of the assay was compared with four other immunoassays against the results of two different liquid chromatography/ mass spectrometry methods (LC-MS/MS) standardized to NIST reference materials. Methods: VIDAS® 25-OH Vitamin D Total assay precision, linearity, detection limits and sample matrix comparison were assessed following CLSI guidelines. For method comparison, a total of 150 serum samples ranging from 7 to 92 ng/mL were analyzed by all the methods. Correlation was studied using Passing-Bablok regression and Bland- Altman analysis. The concordance correlation coefficient (CCC) was calculated to evaluate agreement between immunoassays and the reference LC-MS/MS method. In addition, samples containing endogenous 25(OH)D2 were used to assess each immunoassay’s ability to detect this analyte. Pregnancy and hemodialysis samples were used to the study the effect of vitamin D binding protein (DBP) concentration over VIDAS® assay performance. Results: The VIDAS® 25-OH Vitamin D Total assay showed excellent correlation to the LC-MS/MS results (y = 1.01x+0.22 ng/mL, r = 0.93), as obtained from two different sites and distinct LC-MS/MS methods. The limit of quantification was determined at 8.1 ng/mL. Cross-reactivity for 25(OH)D2 was over 80%. At concentrations of 10.5, 26 and 65.1 ng/mL, within-run CVs were 7.9%, 3.6% and 1.7%, while total CVs (between runs, calibrations, lots and instruments) were 16.0%, 4.5% and 2.8%. The VIDAS® performance was not influenced by altered DBP levels, though under-recovery of 25(OH)D as compared to LC-MS/ MS was observed for hemodialysis samples. Conclusions: The VIDAS® 25-OH Vitamin D Total assay is therefore considered suitable for assessment of vitamin D status in clinical routine. Les études contemporaines autour des cultures alimentaires, menées au départ de différentes perspectives disciplinaires, mettent en évidence le dynamisme interne et interrelationnel qui caractérise les gastronomies, plutôt qu’une « pureté » et un « immobilisme » originels (La Cecla, 1998). Cependant, lorsque l’analyse est portée sur le rapport entre alimentation et migration, une essentialisation des pratiques est souvent produite, plaçant ces dernières dans l’un ou l’autre de deux pôles opposés : le modèle alimentaire associé au pays d’origine et celui majoritairement diffusé dans le pays d’accueil. Une évaluation de ces habitudes ainsi que d’une éventuelle résistance au changement, permettrait d’établir la disposition des migrants à « s’intégrer » à la société locale (Bastenier et Dassetto, 1993 ; De Lesdain, 2002 ; Scardella et al., 2003). Dans ce contexte discursif, où l’inclusion des individus dans certains groupes (la communauté majoritaire, la collectivité « saine » etc.) se fait par l’adhésion aux normes locales, les familles issues de la migration performent leur quotidien alimentaire. Bricoler avec ces normes surdéterminées leur permet d’exprimer une créativité qui se met en œuvre à plusieurs niveaux : celui de l’approvisionnement, celui de la préparation des repas, celui de la transmission des savoir-faire entre les générations. Ces dynamiques seront explorées au travers de l’analyse des données collectées lors d’un travail de terrain mené pendant dix-huit mois au milieu d’un groupe de femmes marocaines résidant dans la province de Milan en Italie. Observations et entretiens ont été conduits à la fois dans les maisons et dans les espaces publics. Ils portaient sur les (micro-)pratiques alimentaires entendues comme facteurs centraux dans la définition du Soi, suivant une approche praxéologique à la subjectivation (Warnier, 2001). Grâce à cette analyse, il sera possible de constater qu’un modèle alimentaire composite se redéfinit au niveau familial, bien qu’il ne soit pas exempté de l’effet de dynamiques culturelles, sociales, économiques et politiques plus amples et apparemment structurantes. On assiste en fait à une diversification et innovation des pratiques. D’un côté, avec leurs déplacements pour faire leurs achats, femmes et hommes dessinent des chorégraphies complexes à l’intérieur de la ville, et ils engagent aussi une circulation transnationale de biens alimentaires. D’un autre côté, mères et filles puisent de savoirs et savoir-faire différents et s’appuient sur une pluralité d’outils (mémoire incorporée, bouche à oreille, livres de recettes, internet) pour faire face à leurs exigences alimentaires. Ainsi, les familles continuent d’effectuer un choix actif en matière d’alimentation. [less ▲]

En 1928, des étudiants de l’Académie des Beaux-Arts de Liège fondent la revue d’architecture et d’urbanisme L’Équerre. S’inscrivant bien que tardivement dans la continuité des avant-gardes internationales, le mensuel revendique une vision transversale de l’architecture où la réflexion plastique occupe une place centrale. Dès le départ, elle s’associe à Anthologie, revue imprégnée des théories futuristes et qui diffuse les artistes de l’avant-garde locale. D’abord par le biais de l’édition et des expositions, plasticiens et architectes travaillent ensuite ensemble dans le cadre de projets ambitieux financés par les pouvoirs publics. Cette contribution revient aux racines de cette collaboration en définissant les liens théoriques ou personnels unissant les deux revues. En se concentrant sur les infrastructures publiques, elle comment les artistes et les architectes ont répondu ensemble aux exigences concrètes de la commande en les replaçant dans le contexte social, politique et économique d’une ville belge de seconde importance à la veille de la Seconde Guerre mondiale. The determination of comprehensive activity-travel patterns is important in the context of agent-based micro-simulation modelling. This paper presents an improved method based on profile Hidden Markov Models (pHMMs) able to include information related to the agents’ residential locations. As proposed in the framework of Liu et al. (2015), pHMMs only characterize activity-travel patterns from the activity sequences perspective. In this context, information related to the primary activity locations (e.g. home, work) is not available and, as a result, it cannot be extracted from the pHMMs themselves. With respect to this limitation, we propose to apply the framework of Liu et al. (2015) with an extension to include characterization of residential locations. Following the established guidelines, the activity sequences and their related residential locations are extracted from the activity-travel diaries in order to estimate the regularity of the activities as well as their sequential order. Subsequently, within each residential activity, we include a categorization at an aggregate level (provinces). The methodology is powerful as it characterizes any length of sequence, allowing the generation of unlimited agent plans with information about residential location. Regarding data collection, the activity-travel diaries are provided by the Belgian Household Daily Travel Survey (2010). The results obtained after the simulations indicate a good match between the predicted and observed residential locations at both the national and provincial levels. [less ▲]
