[more ▼] The Io footprint (IFP) is a set of auroral spots and an extended tail resulting from the strong interaction between Io and the Jovian magnetosphere. For the first time, we present measurements of the brightness and precipitated power for each individual spot, using the image database gathered from 1997 to 2009 with the Hubble Space Telescope in the Far-UV domain. We show that the relative brightness of the spots varies with the System III longitude of Io. Moreover, our novel measurement method based on 3D simulations of the auroral features allows to derive the precipitated energy fluxes from images on which the emission region is observed at a slant angle. Peak values as high as 2 W/m² are observed for the main spot, probably triggering a localized and sudden heating of the atmosphere. Additionally, strong brightness differences are observed from one hemisphere to another. This result indicates that the location of Io in the plasma torus is not the only parameter to control the brightness, but that the magnetic field asymmetries also play a key role. Finally, we present new data confirming that significant variations of the spots' brightness on timescales of 2-4 minutes are ubiquitous, which suggests a relationship with intermittent double layers close to Jovian surface. [more ▼] Background In order to replace petrochemicals by bio-based lignin products in a lot of high value-added applications, a formic/acetic acid treatment was adapted to beech wood (Fagus sylvatica L.) for lignin extraction. Results Beech wood particles were delignified at atmospheric pressure by a formic acid/acetic acid/water mixture. Cooking time and temperature were optimized for delignification, pulp yield and 2-furfural concentration. Response surface design analysis revealed that delignification yield increased with cooking time and temperature. Conclusion The multi-criteria optimization of delignification was used to find the ideal cooking conditions (5h07 min, 104.2°C) which could be satisfactory for the maximization of delignification (70.5%) and pulp yield (58.7%) and, to a lesser extent, for the minimization of 2-furfural production. Treatment conditions were found to influence the chemical structure of extracted lignins. Cooking time and temperature influenced inversely lignin molecular weights. [more ▼] Dans les diverses réformes conçues et implémentées en vue de moderniser l’administration, l’accent est mis sur les marchés avec un focus particulier sur les citoyens désormais clients (Korunka et al., 2007 ; Thomas, 2012). Alors que les approches initiales du NPM se centraient sur l’efficience, l’applicabilité et la pertinence de techniques managériales importées du secteur privé dans les organisations publiques (Ferlie et al., 1996), les réformes plus récentes tendent à inclure d’autres objectifs tels que la gestion de la qualité, la transparence et le centrage client (Meier & O’Toole, 2007 ; Lapsley, 2008). Du Gay (1996), pour sa part, affirme que le NPM est un projet identitaire, dans la mesure où il vise à redéfinir la main d’œuvre (en termes de profils et compétences recherchées, ciblant la figure du travailleur-entrepreneur en lieu et place du fonctionnaire). Bon nombre d’études interrogent également les paradoxes et les tensions intrinsèques du modèle NPM (Lynn, 1998; Aucoin, 1990; Hood, 2000 ; Goldfish & Wallis, 2010), les difficultés liées à sa mise en œuvre (Mathiasen, 1999 ; Dunleavy et al., 2006 ; Jones & Cole, 2009) ou à l’évaluation de ses outputs concrets pour l’administration (Pollitt, 1993; Hodge, 1999). De façon générale, la littérature met en lumière les effets que ces réformes ont produit sur les différentes facettes de l’administration : ses modes de fonctionnement et ses structures (Gualmini, 2008), son management, ses stratégie et pratiques de GRH (Pichault, 2007), ses missions et son identité organisationnelle (Horton, 2006) mais aussi sur l’ethos (Maesschalk, 2004 ; Fortier, 2010) et la motivation des fonctionnaires (Vandenabeele et Hondeghem, 2007), entre autres éléments. Plusieurs auteurs mentionnent les débats et questionnements identitaires qui se produisent lorsque l’organisation fait face à des situations de bouleversement profond (Albert et Whetten, 1985 ; Dutton et al., 1994 ; Alvesson et Empson, 2008). Dans un processus de réforme, l’organisation se remet en question et s’interroge sur ce qu’elle est, ce qu’elle devient, ce qu’elle veut être, propulsant l’identité organisationnelle au premier plan des préoccupations (Skalen, 2004 ; Meyer & Hammerschmid, 2006). Dans un contexte où l’identité organisationnelle est en proie à tel bouleversement, comment ses membres en font-ils l’expérience ? Ce questionnement, à la base de cet article, se pose en termes d’identification organisationnelle, unissant dans une même équation deux termes centraux : l’identité organisationnelle et les perceptions qu’ont les membres du contexte en évolution. La question que nous adressons est la suivante : dans un contexte d’organisation publique en mutation, quelles sont les dynamiques identitaires à l’oeuvre en regard de la perception des évolutions du contexte des individus ? A partir de l'analyse de la réforme Copernic (introduction du NPM dans l'administration fédérale belge) réalisée en 2005-2006, nous avons élaboré de manière inductive une typologie des logiques et profils identitaires, issue de l’analyse d’entretiens qualitatifs (Rondeaux, 2007, 2008, 2010). Sur base de celle-ci, nous avons mené une enquête par questionnaires auprès d’une administration régionale belge, engagée dans un processus de modernisation, en vue de vérifier l’existence et la pertinence de cette typologie, au moyen d’une démarche quantitative. Réalisée en 2010, cette enquête illustre en particulier trois aspects : 1. les représentations associées au travail au sein de l’administration en regard de son évolution, et le degré d’identification à l’institution dans ce contexte ; 2. la perception de la modernisation et le rôle de la hiérarchie dans cette perspective ; 3. les identités représentées au sein de l’institution (logiques institutionnalisée et alternatives, profils en congruence ou en dissonance). Nous montrons notamment que la majorité des positions est marquée par la dissonance, principalement relative à la valeur ajoutée de la modernisation et à sa mise en œuvre concrète. However, there is still a controversy on the origin of steroids in these animals. In this study, free and esterified testosterone concentrations were measured in the hermaphroditic aquatic gastropod Lymnaea stagnalis exposed to molecules known for their androgenic (testosterone and tributyltin), anti-androgenic (cyproterone-acetate) and estrogenic (chlordecone) properties by reference to their mode of action in vertebrates. In parallel, snail oviposition and fecundity were followed over the 21-day exposure period. Testosterone exposure resulted in increased esterified testosterone levels whereas free testosterone concentrations remained stable. In contrast, cyproterone-acetate increased significantly the free form of testosterone with no changes in the esterified form whereas chlordecone showed a tendency to reduce, though not significantly, esterified testosterone concentrations without changes in free testosterone levels. Finally, tributyltin did not alter testosterone homeostasis. The production of egg-clutches and eggs was significantly reduced only in the snails exposed to the highest concentrations of chlordecone (19.6 µg/L) and tributyltin (94.2 ng Sn/L). Overall, this study demonstrates that uptake of testosterone from the exposure medium occurs in L. stagnalis. Moreover, it shows that cyproterone-acetate and, to a lesser extent, chlordecone can alter endogenous testosterone levels in this freshwater snail. However, relationship between hormonal changes and snail reproduction has not been established. The compressor was tested with oil injection mass flow fractions of up to 45%. At an evaporating temperature of -10C and condensing temperature of 30C, the overall isentropic efficiency was up to 70% at the highest oil injection rate. Overall, over the testing envelope investigated, there are no significantly negative effects experienced for the compressor and the compressor isentropic efficiency and refrigerant mass flow rate improve monotonically as the oil injection rate is increased. Contrary to earlier assumptions, it is now clear that both PTKs and PTPs are highly specific, non-redundant, and tightly regulated enzymes. Hematopoietic cells express particularly high numbers of PTKs and PTPs, and aberrant function of these proteins have been linked to many hematopoietic disorders. While PTK inhibitors are among FDA approved drugs for the treatment of leukemia and other cancers, efforts to develop therapeutics that target specific PTPs are still in its infancy. Here, we describe methods on how to evaluate effects of PTP inhibitors on T cell receptor signaling. Moreover, we provide a comprehensive strategy for compound prioritization, applicable to any drug discovery project involving T cells. We present a testing funnel that starts with relatively high-throughput luciferase reporter assays, followed by immunoblot, calcium flux, flow cytometry, and proliferation assays, continues with cytokine bead arrays, and finishes with specificity assays that involve RNA interference. We provide protocols for experiments in the Jurkat T cell line, but more importantly give detailed instructions, paired with numerous tips, on how to prepare and work with primary human T cells.
