Excessive neutrophil stimulation and reactive oxygen species (ROS) production are involved in numerous human or horse pathologies. The modulation of the neutrophil NADPH oxidase (NOX) has a great therapeutic potential since this enzyme produces superoxide anion whose most of the other ROS derive. The measurement of NOX activity by cell-free systems is often used to test potential inhibitors of the enzyme. A major drawback of this technique is the possible interferences between inhibitors and the probe, ferricytochrome c, used to measure the activity. We designed the "EquiNox2", a new pharmacological tool, to determine the direct interaction of potential inhibitors with equine phagocytic NOX and their effect on the enzyme activity or assembly. This method consists in binding the membrane fractions of neutrophils containing flavocytochrome b558 or the entire complex, reconstituted in vitro from membrane and cytosolic fractions of PMNs, onto the wells of a microplate followed by incubation with potential inhibitors or drugs. After incubation, the excess of the drug is simply eliminated or washed prior measuring the activity of the reconstituted complex. This latter step avoid the risk of interference between the inhibitor and the revelation solution and can distinguish if inhibitors, strongly bound or not, could interfere with the assembly of the enzymatic complex or with its activity. The EquiNox2 was validated using diphenyliodonium chloride and Gp91ds-tat, two well-known inhibitors largely described for human NADPH oxidase. The present technique was used to study and understand better the effect of curcumin and its water-soluble derivative, NDS27, on the assembly and activity of NOX. We demonstrated that curcumin and NDS27 can strongly bind to the enzyme and prevents its assembly making these molecules good candidates for the treatment of horse or human pathologies implying an excessive activation of neutrophils. Je discute ici le projet diltheyen d'une "fondation psychologique des sciences de l'esprit". Je suggère que ce projet peut être rapproché d’un certain programme épistémologique baptisé "empirisme psychologique". L’hypothèse qui se trouve à la base de l’empirisme psychologique est la suivante : l’expérience interne, psychologique, est la source des concepts au moyen desquels nous pouvons penser et connaître le monde. Après avoir brièvement reconstruit le programme diltheyen en ce sens, j'examine deux objections qui ont été soulevées contre lui: l'objection néokantienne (Windelband, Rickert) et l'objection psychologique (Ebbinghaus, Stumpf). La seconde, selon moi, met en évidence une difficulté réelle du projet de Dilthey: la "connexion structurelle de la vie" dans sa totalité peut difficilement être considérée comme "donnée" dans l'expérience interne, ainsi que l'affirme Dilthey. Je soutient donc que le programme de l'empirisme psychologique peut être poursuivi à condition de le débarrasser de cette thèse problématique. [less ▲]

Cette communication vise à étudier la participation des femmes migrantes marocaines à la vie sociale du « pays d’accueil » ; participation qui se réalise grâce aux activités de la vie quotidienne. En particulier, la performance publique des pratiques culinaires, observée dans une ville de la province de Milan, permet aux femmes considérées d’exercer une agency à l’intérieur des espaces institutionnellement concédés à l’expression des migrants. D’un côté, la consommation des « recettes de l’Autre » de la part de la population majoritaire détermine une incorporation de la diversité afin de la contrôler. De l’autre côté, la préparation de ces recettes consent aux migrantes d’exprimer leur subjectivité (individuelle et collective) de façon active et en présentant des aspects qui vont au-delà de la culture alimentaire. Une participation sociale fondée sur le partage de nourriture donne la possibilité aux femmes d’assumer un rôle actif dans la communauté locale, tout en affirmant ses propres spécificités. Des sujets tels que la liberté religieuse sont abordés, et certains droits peuvent être réclamés par des individus qui ont ainsi acquis une position politiquement importante au sein de la société locale. High-level, long-lived radioactive waste remains hazardous for periods that go well beyond our human conception of time (many thousands of years). Because active safety measures are considered unreliable, unjustifiable and simply impossible over such long time spans, experts worldwide recommend geological disposal as the preferred strategy for long-term radioactive waste management, to a large extent due to its promise of delivering ‘passive safety’. Passive safety refers to the repository being safe ‘by itself’, i.e. unmediated by human actors and actions. Safety is thus approached technically and delineated as an intrinsic property of the disposal system. As such, the notion of 'passive safety' entails a system simplification that allows for approaching safety in a more calculable and predictable manner than would be the case for 'active safety'. In this article, we describe and analyze the ambiguity of this seemingly straightforward approach to safety. Drawing on constructivist insights from safety science and science and technology studies, we propose a more integrated sociotechnical vision that transcends the active versus passive safety division. The notion of 'oversight', as it is currently starting to surface in international radioactive waste management discourses, will be used as a sensitizing concept, offering potential to elaborate such an integrated vision. Aims. The main-sequence B-type star zeta Cassiopeiae is known as a N-rich star with a magnetic field discovered with the Musicos spectropolarimeter. We model the magnetic field of the star by means of 82 new spectropolarimetric observations of higher precision to investigate the field strength, topology, and effect. Methods. We gathered data with the Narval spectropolarimeter installed at Télescope Bernard Lyot (TBL; Pic du Midi, France) and applied the least-squares deconvolution technique to measure the circular polarisation of the light emitted from zeta Cas. We used a dipole oblique rotator model to determine the field configuration by fitting the longitudinal field measurements and by synthesizing the measured Stokes V profiles. We also made use of the Zeeman-Doppler imaging technique to map the stellar surface and to deduce the difference in rotation rate between the pole and equator. Results. zeta Cas exhibits a polar field strength B_pol of 100-150 G, which is the weakest polar field observed so far in a massive main-sequence star. Surface differential rotation is ruled out by our observations and the field of zeta Cas is strong enough to enforce rigid internal rotation in the radiative zone according to theory. Thus, the star rotates as a solid body in the envelope. Conclusions. We therefore exclude rotationally-induced mixing as the cause of the surface N-enrichment. We discuss that the transport of chemicals from the core to the surface by internal gravity waves is the most plausible explanation for the nitrogen overabundance at the surface of zeta Cas.
