Aim: Produce biodegradable nanoparticles to target antigen-presenting cells and evaluate their potential to be used as a vaccine delivery system. Materials & methods: Untargeted PEGylated PLGA-based nanoparticles and mannose-grafted nanoparticles were formulated and physicochemically characterized. Immortalized and primary antigen-presenting cells were used to study nanoparticle internalization patterns. The endocytic pathways and intracellular trafficking followed by nanoparticles were also investigated. Results & discussion: Nanoparticles displayed mannose residues available for binding at the nanoparticle surface. Different nanoparticle internalization patterns by immortalized and primary antigen presenting cells were verified. Macropinocytosis, clathrin-mediated endocytosis, caveolin- and lipid raft-dependent endocytosis are involved in nanoparticles internalization. Nanoparticles demonstrate both endo-lysosomal and cytosolic localizations and a tendency to accumulate nearby the endoplasmic reticulum. Conclusion & future perspective: The developed nanoparticles might drive antigens to be presented through MHC class I and II molecules to both CD8+ and CD4+ T cells, favoring a complete and coordinated immune response. This paper draws on vulnerability analysis as it emerged as a complement to classical risk analysis, and it aims at exploring its ability of nurturing risk and vulnerability governance actions. An analysis of the literature on vulnerability analysis allows us to formulate a threefold critique: first, vulnerability analysis has been treated separately in the natural and the technological hazards fields. This separation prevents vulnerability to unleash the full range of its potential, as it constraints appraisals into artificial categories and thus already closes down the outcomes of the analysis. Second, vulnerability analysis focused on assessment tools that are mainly quantitative, whereas qualitative appraisal is a key to assessing vulnerability in a comprehensive way and to informing policy-making. Third, a systematic literature review of case studies reporting on participatory approaches to vulnerability analysis allows us to argue that participation has been important to address the above, but it remains too closed down in its approach and would benefit from embracing a more open, encompassing perspective. Therefore, we suggest rethinking vulnerability analysis as one part of a dynamic process between opening-up and closing-down strategies, in order to support a vulnerability governance framework. Shift-share analysis is a decomposition technique widely used in regional studies to quantify an industry-mix effect and a competitive effect on the growth of regional employment (or any other relevant variable) relative to the national average. This technique has always been subject to criticism for its lack of theoretical basis. This paper presents a critical assessment of the methods suggested by Dunn (1960) and by Esteban-Marquillas (1972) and proposes a new shift-share method, which separates out the two effects unambiguously. By way of illustration, we provide an application to manufacturing employment in the Belgian provinces between 1995 and 2007. [less ▲]

L’usage des chiffres dans l’action publique (ou la « quantification du politique ») mobilise deux registres en tension : premièrement celui de la performance, où « le chiffre est conçu comme un acteur contribuant à encadrer l’action en restreignant les options soumises à délibération et les choix ouverts à la décision collective », et deuxièmement la démocratie, perçue comme « un facteur d’extension du débat public et d’accroissement des sphères d’exercice de la responsabilité politique des citoyens ». Ogien relève le renforcement de cette tension, tout en soulignant l’absence de démocratisation de la mesure elle-même. Afin de remédier à cet écueil, l’auteur propose que les citoyens, rarement pris en considération dans l’élaboration et la définition des termes de la mesure, soient parties prenantes « du système du chiffre qui sert à rendre compte de leur existence et de leur destinée ». Cette contribution analyse les modalités concrètes de cette tension dans le cadre de l’évaluation d’une politique régionale mise en place au niveau local, les plans de cohésion sociale (PCS) en Région wallonne (Belgique), en mobilisant les données recueillies lors de l’évaluation des PCS terminée en mars 2013 . En effet, un enjeu important des PCS était la mise en place d’un processus d’évaluation qui se voulait « participatif» . La participation citoyenne étant une notion centrale de la philosophie PCS, un des objectifs de la Direction interdépartementale de la cohésion sociale (DiCS – administration régionale en charge du programme) était de la promouvoir à travers toutes les étapes de mise en œuvre de cette politique, y compris pour l’évaluation d’impact, inspirée en cela par les propositions méthodologiques du Conseil de l’Europe. Dans un premier temps, le bureau régional de statistique avait développé un indicateur composite « d’accès aux droits sociaux fondamentaux » afin d’objectiver la clé de répartition des ressources entre les territoires locaux. Ce n’est que dans un second temps, en fin de programme, que la DiCS était sommée de produire une « évaluation participative » mesurant l’impact des actions réalisées à travers les PCS sur la cohésion sociale et le bien-être. L’article propose de confronter ces deux démarches d’évaluation : celle qui aboutit à un indicateur quantitatif fondé sur une logique statistique et l’approche participative favorisant l’expression des parties prenantes. A full Bayesian approach based on ODE-penalized B-splines and penalized Gaussian mixture is proposed to jointly estimate ODE-parameters, state function and error distribution from the observation of some state functions involved in systems of affine differential equations. Simulations inspired by pharmacokinetic studies show that the proposed method provides comparable results to the method based on the standard ODE-penalized B-spline approach (i.e. with the Gaussian error distribution assumption) and outperforms the standard ODE-penalized B-splines when the distribution is not Gaussian. This methodology is illustrated on the Theophylline dataset. [less ▲]

An ordered mesoporous carbon (OMC) was functionalized with ammonium or chelating phosphine ligands. In both cases, the functionalization procedure started by oxidation by nitric acid treatment, followed by activation of surface carboxylic acid groups with thionyl chloride, then formation of amide bonds with diamines. The pendant amine groups were then either quaternized or further reacted with phosphine. The introduced functions were used as anchors for molecular mixed-metal clusters. These organometallic grafted species could then be thermally transformed into hetero-metallic nanoparticles (NP) embedded within the mesoporous framework. The NP/OMC nanocomposites could find application in hydrogenation, heterogeneous catalysis or as electrodes in fuel cells. [less ▲]
