While Multi-Variate Pattern Analysis techniques based on machine learning have now been regularly applied to neuroimaging data, decoding brain activity is usually performed in highly controlled experimental paradigms. In more realistic conditions, the number, sequence and duration of mental states are unpredictably generated by the individual, resulting in complex and imbalanced fMRI data sets. Moreover, in the case of spontaneous brain activity, the mental states can not be linked to any external or internal stimulation, which makes it a highly difficult condition to decode. This study tests the classification of brain activity, acquired on 14 volunteers using fMRI, during mental imagery, a condition in which the number and duration of mental events were not externally imposed but self-generated. Application of the obtained model on rest sessions allowed classifying spontaneous brain activity linked to the task which, overall, correlated with their behavioural performance to the task. Plant traits are particularly important in determining plant community structure. However, how can one identify which traits are the most important in driving community assembly? Here we propose a method 1) to quantify the direction and strength of trait selection during community assembly and 2) to obtain parsimonious lists of traits that can predict species relative bundances in plant communities. We tested our method using floristic data from 32 plots experiencing different treatments (fertilisation and grazing) in southern France. Twelve functional traits were measured on 68 species. We determined the direction and strength of selection on these 12 traits using a metric derived from a maximum entropy model (i.e. lambda). We then determined our parsimonious list of traits using a backward selection of traits based on these lambda values (for all treatments and in each treatment separately). We finally compared our method to two other methods: one based on iterative RLQ and the other based on an entropy-based forward selection of traits. We found major differences in the direction and strength of selection across the 12 traits and treatments. From the 12 traits, plant vegetative and reproductive heights, leaf dry matter content leaf nitrogen content, specific leaf area, and leaf phosphorus content were particularly important for predicting species relative abundances when considering all treatments together. Our method yielded results similar to those produced by the entropy-based approach but differed from those produced by the iterative RLQ, whose selected traits could not significantly predict species relative abundances. Together these results suggest that the assembly of these communities is primarily driven by a small number of key functional traits. We argue that our method provides an objective way of determining a parsimonious list of traits that together accurately predict community structure and which, despite its complementarities with entropy-based method, offers significant advantages. The influence of dimensions on the mechanical behavior of f.c.c. single crystals with dimensions larger than a few micrometers has been the topic of many experimental investigations and controversies during the 1970s and this question is still open. The objective of this article is to shed new light on this point by performing finite element simulations thanks to a strain gradient crystal plasticity model. Based on the model identification for nickel, several single crystal samples with various thicknesses and orientations were tested numerically in tension. The effect of dimensions was then analyzed considering the spatial distribution of dislocation densities. Near loading boundaries, dislocation density gradients appeared perpendicular to the Burgers vector direction of the primary activated slip system which modified the mechanical behavior. These gradients are discussed in terms of boundary conditions, crystal orientation and dislocation interactions with surfaces. [less ▲]

Mastitis remains the most important health issue in dairy cattle. Improved methods to identify cows developing subclinical mastitis would benefit farmers. We herein describe a novel method to determine the somatic cell counts (SCC) of individual cows by bulk genotyping a sample of milk from the milk tank with panels of genome-wide single nucleotide polymorphisms (SNP). We developed a simple linear model to estimate the contribution of individual cows to the genomic DNA present in the tank milk from 1) the known genotypes of individual cows for the interrogated SNP and 2) the ratio of SNP alleles in the tank milk. Using simulations, we estimate that 3,000, 50,000, and 700,000 SNP are sufficient to accurately (R(2)>0.98) estimate individual SCC in tanks containing milk from 25, 100, and 500 cows, respectively. Using actual data, we demonstrate that the SCC of 21 cows can be estimated with a coefficient of determination of 0.60 using approximately 9,000 SNP. The proposed method increases the value of the proposition of SNP genotyping individual cows for genomic selection purposes. [less ▲]

First human retrovirus discovered, HTLV-1 infects approximately twenty million individuals worldwide. HTLV-1 is the causative agent of different diseases among which the most important are the adult T-cell leukemia (ATL) and a neurodegenerative disorder called HAM/TSP (Human associated myelopathy/ Tropical spastic paraparesis). We are interested in the mechanisms of transformation by the viral Tax oncoprotein. Recently, we showed that Tax interacts with the minichromosome maintenance MCM2-7 helicase and binds to origins of DNA replication (Boxus et al, 2012 Blood 119:151). Thereby, Tax modulates the spatiotemporal program of origin activation during the S phase of cell cycle. In fact, Tax accelerates S phase progression by inducing early firing of late replication origins. Concomitantly, Tax-MCM2-7 interplay also modulates viral transcription. Together, our data demonstrates that interaction between Tax and MCM2-7 is involved in replication reprogramming and viral transcription. [less ▲]
