Abstract :
[en] Since several years, research efforts have been focused towards improving the reproductive potential of domestic animals. The authors present a review of different technics in particular the superovulation and priming, embryo micromanipulation and clonage. The technic of superovulation allows an increase of the total number fo the offspring by an increase of the ovocytes total number. Different FSH injections at the end of the cycle, preceded by small FSH quantity at the beginning of the cycle, procure a great number of mature follicles available for reproduction. Micromanipulation allows a multiplicaton by sectioning the embryo in two or more segments. Isolated blastmeres can also be used but it was observed that those cells very rapidly loose their differentiation potential. Clonage allows the reproduction of an animal from only one of his own cells. The nucleus of this cell is extracted and reimplanted in an enucleated ovocyte. Meanwhile from a practical point of vue, this technic is, at present, only possible using a stem cell and not using a mature somatic cell.
[fr] Depuis plusieurs années, de nombreux travaux de recherche ont pour but d'améliorer le potentiel reproducteur des animaux domestiques. Les auteurs réalisent une revue de ces différentes méthodes, particulièrement: la superovulation et le priming, la micromanipulation et le clonage. La superovulation permet l'accroissement du nmbre de descendants en augmentant le nombre d'ovocytes disponibles lors de chacun des cycles. Par une série d'injections de FSH en fin de cycle, précédée en début de cycle par des injections sensibilisantes (priming), on obtient un accroissement du nombre de follicules venant à maturation et pouvant être exploités pour la reproduction. La micromanipulation permet une multiplication des embryons en les sectionnant en deux ou plusieurs fragments qui réimplantés donneront naissance à plusieurs foetus. Des blastomères isolés aux stades précoces peuvent également être utilisés, cependant, on constate très rapidement une perte du pouvoir de différenciation. Le clonage permet lui de reproduire un individu à partir d'une seule de ses cellules dont le noyau sera prélevé et inséré dans un ovocyte au préalable énucléé. Cependant, en pratique, si le transfert de noyaux à partir de cellules somatiques de l'organisme adulte paraît aujourd'hui chose très difficile, le transfert de noyaux de cellules embryonnaires est parfaitement réalisable.
[nl] Sedert verscheidene jaren wordt onderzoek gedaan naar het verbeteren van de voortplantingsresultaten bij de huisdieren. Er wordt een overzicht gegeven van de verschillende methods die hierbij worden toegepast, meer bepaald de superovulie en priming, de micromanipulatie en het cloneren van embryo's. Superovulatie laat toe om meer nakomlingen te bekomen door het verhogen van het aantal ovocyten dat beschikbaar komt bij elke cyclus. Door een reeks injecties van FSH op het einde van de cyclus, voorafgegaan door sensibiliserende injecties (primint) in het begin van de cyclus, verhoogt het aantal follikels dat tot rijping komt en dat dus beschikbaar komt voor de voortplanting. Micromanipulatie laat toe om meer embryo's te bekimen door splitsen in 2 of meer fragmenten die, na overplanten, ontwikkelen tot meerdere foeti. Geisoleerde blastomeren kunnen eveneens gebruikt worden doch deze cellen verliezen zeer snel de mogelijkheid tot differentiate. Onder cloneren verstaat men de voortplanting van een individu uitgaande van een van zijn eigen cellen, waarvan de kern wordt getransplanteerd in een vooraf geenucleerede eicel. Op dit ogenblik is kerntranplantatie uitgaande van somatische cellen van volwassen individuen praktisch moeilijk haalbaar, daar waar de transplantatie van kernen van embryonale cellen perfekt mogelijk is.
