Abstract :
[en] Background Plasma transforming growth factor β1 (TGFβ1) levels are elevated in patients with lung cancer. As TGFβ1 is mainly found in platelets and as nonmalignant pulmonary diseases (NMPD) are frequently associated with lung cancer, we investigated the potential contribution of platelet degranulation and/or of a concomitant NMPD to the increased plasma levels of TGFβ1 reported in patients with lung cancer. Materials and Methods Blood samples were collected in duplicate from 30 healthy subjects, 14 patients suffering from NMPD and 37 patients with lung cancer. The platelet count was determined and the samples were processed to obtain plasma. One sample was collected in EDTA (EDTA plasma) and the other in a mixture inhibiting platelet degranulation (PIM plasma). TGFβ1 concentrations and β-thromboglobulin (βTG) levels, an index of platelet degranulation, were measured in both plasma samples. Results TGFβ1 and βTG plasma levels measured in PIM plasma were lower than those obtained in EDTA plasma. With respect to PIM plasma, both TGFβ1 and βTG levels were higher in patients with lung cancer than those with NMPD and in healthy individuals. In patients with NMPD, only TGFβ1 levels were increased as compared to healthy controls, βTG levels being similar. Conclusion Methods for collecting and processing blood samples are critical in determining reliable circulating TGFβ1 levels. Increased TGFβ1 plasma levels observed in patients with lung cancer are related, at least partly, to concomitant NMPD and also to platelet degranulation as proved by increased βTG levels.

Comme TGFβ 1 est principalement trouvée dans les plaquettes et les maladies non cancéreuses pulmonaires (NMPD) sont fréquemment associés au cancer du poumon, nous avons enquêté sur la contribution potentielle de la dégranulation des plaquettes et / ou d'une administration concomitante NMPD à l'augmentation des taux plasmatiques de TGFβ 1 signalé chez les patients présentant cancer du poumon. Matériels et Méthodes Des échantillons de sang ont été prélevés en double à partir de 30 sujets sains, 14 patients souffrant de NMPD et 37 patients atteints de cancer du poumon. The platelet count was determined and the samples were processed to obtain plasma. La numération plaquettaire a été déterminée et les échantillons ont été traités pour obtenir le plasma. One sample was collected in EDTA (EDTA plasma) and the other in a mixture inhibiting platelet degranulation (PIM plasma). Un échantillon a été prélevé dans l'EDTA (plasma EDTA) et l'autre dans un mélange d'inhiber la dégranulation des plaquettes (plasma PIM). TGFβ 1 concentrations and β-thromboglobulin (βTG) levels, an index of platelet degranulation, were measured in both plasma samples. Résultats TGFβ 1 et les niveaux plasmatiques βTG mesurée dans le plasma PIM étaient inférieurs à ceux obtenus dans le plasma EDTA. With respect to PIM plasma, both TGFβ 1 and βTG levels were higher in patients with lung cancer than those with NMPD and in healthy individuals. En ce qui concerne le plasma PIM, les deux TGFβ 1 et les niveaux βTG étaient plus élevés chez les patients atteints de cancer du poumon que ceux qui ont NMPD et chez les individus sains. In patients with NMPD, only TGFβ 1 levels were increased as compared to healthy controls, βTG levels being similar. Chez les patients avec NMPD, seuls TGFβ 1 étaient en augmentation par rapport aux contrôles sains, βTG niveaux étant similaires. Conclusion Methods for collecting and processing blood samples are critical in determining reliable circulating TGFβ 1 levels. Conclusion méthodes de collecte et de traitement des échantillons de sang sont essentiels pour déterminer TGFβ fiables circulent niveaux 1. Increased TGFβ 1 plasma levels observed in patients with lung cancer are related, at least partly, to concomitant NMPD and also to platelet degranulation as proved by increased βTG levels. Augmentation du TGFβ 1 niveaux plasmatiques observées chez les patients atteints de cancer du poumon sont liés, au moins en partie, d'une cardiopathie NMPD et aussi de la dégranulation des plaquettes ainsi que le prouvent des niveaux accrus βTG.
