Abstract :
[en] Developmental orthopaedic diseases and specially osteochondrosis (OC), are one of the main causes of subsequent joint disease. The principal aims of this study are to determine the current prevalence of OC in Belgium, to investigate the impact of the environment in OC evolution and the research of possible biomarkers for OC diagnosis and prediction through transcriptomic and metabolic profiles. 38.37% of the Belgian Sport yearlings presented OC lesions. The most commonly affected joint was the fetlock followed by the hock and stifle. Palmar/plantar fragmentations in the margin of the proximal phalanx were not included in the final evaluation of OC prevalence. Concerning the effect of the environment in the evolution of OC, foals fed with concentrates show a higher probability of the development of OC lesions (p=0.06), while foals not receiving concentrates, show a higher probability of healing from existing lesions (p=0.001). The effect of housing conditions was not statistically significant. As environment is able to modify the evolution of the disease, diagnostic and predictive biomarkers where researched with two main procedures transcriptomic analysis and ELISA technique. This later technique showed that sclerostin concentrations (a glycoprotein related to the bone formation) where higher in OC affected foals of 6 month of age, but not in 18 month ones. That test showed that there was not relation between sclerostin concentrations and the type of environment. In the transcriptomic analysis OC affected foals showed a different expression of several genes when compared to a control group and this expression was influenced by age and affected joint. Differential expression of genes according to the joint was also studied and based on the profiles of expression of some genes, the localization of the disease can be determined. Besides, transcriptomics allowed to study the transcript profile of foals in order to predict the presence of the disease. The Odds ratio showed a relationship between the OC status assessed by a radiographic protocol at 18 months of age and the OC status given by transcriptomic tools at 6 months of age. In conclusion, this research may open the door to an OC predictive tool since OC prevalence is still quite high.
[fr] Les maladies orthopédiques du développement et spécialement l’ostéochondrose (OC), sont une des premières causes des futures maladies articulaires du cheval. Le principal objectif de cette étude est d’établir la prévalence actuelle d’OC en Belgique, de connaître l’impact de l’environnement dans l’évolution de la maladie, et de rechercher des biomarqueurs possibles comme moyen diagnostique et prédictif à partir des profils métaboliques et transcriptomiques. 38.37% des poulains de Sport Belge présentaient des lésions d’OC. L’articulation la plus touchée était le boulet, suivi du jarret et du grasset. Les fragmentations palmaires/plantaires dans la partie proximale de la première phalange n’ont pas été inclus dans la prévalence totale. Concernant l’effet de l’environnement sur l’évolution de l’OC, les poulains nourris avec des concentrés avaient une probabilité plus élevée de développer de l’OC (p=0.06) et les poulains nourris avec du foin avaient une probabilité significative de guérir des lésion existantes (p=0.001). L’effet de l’hébergement n’était pas significatif. Comme l’environnement est capable de modifier l’évolution de la maladie, des marqueurs diagnostiques et prédictifs ont été recherchés avec des techniques transcriptomiques et test ELISA. Cette dernière a montré que les poulains atteints d’OC ont des concentrations plus élevées de sclérostine (une glycoprotéine liée au métabolisme osseux) mais pas sur des poulains de plus de 18 mois. Aucune relation a été observée entre les concentrations de sclérostine et le type d’environnement. Les analyses transcriptomiques ont montré que les poulains atteints d’OC montrent une expression différente de certains gènes par rapport au groupe contrôle. Cette expression était influencée par l’âge et l’articulation atteinte. Avec cette technique la localisation articulaire de la maladie peut être déterminée. De plus, la transcriptomique a permis d’étudier le profil d’expression des gènes pour la prédiction de la maladie. Les Odds Ratio ont démontré une relation entre le profil prédictif à 6 mois et le résultat du protocole radiographique de dépistage à 18 mois. En conclusion, cette recherche pourrait ouvrir la porte à un outil prédictive de l’OC pourvu que la prévalence d’OC reste toujours très élevée.
