Abstract :
[en] Equine bone gamma-carboxyglutamic acid containing protein - also called osteocalcin - has been isolated for the first time till homogeneity. After acid demineralisation and acid extraction, the purification procedure was based on reverse phase chromatography, gel filtration and ion exchange chromatography. The amino acid sequence of the primary structure has been determined to be YLDHWLGA(HYP) APYPDPL(GLA)PRR(GLA)VC (GLA)LNPDCDELADHIGF-AYRRFYGPV. The protein has a calculated molecular weight of 5,732 kDa and a calculated isoelectric point of 4,45. It contains 49 amino acids, from which 3 are gamma-carboxyglutamic acid residues at position 17, 21 and 24 and has a fully hydroxylated proline in position nine and cysteine in position 23 and 29. Equine osteocalcin shows a strong homology with bovine (98%), ovine (96%) and human (94%) osteocalcin
[fr] L’ostéocalcine, protéine osseuse contenant de l'acide gamma-carboxyglutamique, a été isolée jusqu'à homogénéité pour la première fois dans l'espèce équine. Après déminéralisation acide et extraction acide, l'ostéocalcine a été purifiée par chromatographie en phase greffée par chaîne apolaire en C18, chromatographie de filtration sur gel et chromatographie échangeuse d'ions. La séquence en acides aminés de l'ostéocalcine équine a été déterminée, elle correspond à YLDHWLGA(HYP) APYPDPL(GLA) PRR(GLA)VC(GLA)LNPDCDELAD HIGFQEAYRRFYGPV. La masse moléculaire et le point isoélectrique de la protéine ont été calculés de manière théorique à partir de la séquence transcrite, ils correspondent respectivement à 5,732 kDa et à 4,45. La protéine contient 3 résidus acides gamma-glutamiques en position 17, 21 et 24. La séquence révèle également la présence de hydroxyproline en position 9 et de cystéine en positions 23 et 29. L’homologie de séquence avec l'ostéocalcine bovine (98 %), ovine (96 %) et humaine (94 %) est élevée
