Abstract :
[en] The general framework of this study is the discovery of prebiotics and more specifically the production of oligosaccharides by enzymatic hydrolysis of locust bean gum. These, though not much studied, could present a rich prebiotics potential. The aim of this work was the purification of beta-mannanases located in germinating carob seeds as well as the validation of a AEC-PAD method applicable to the analysis of locust bean gum hydrolysates. Therefore, it has permitted to establish a concentration and purification procedure of beta-mannanases and to determine a suitable preservation method of enzymatic extracts. An original method to evaluate the enzymatic activity by viscosity measurements has also been developed and permits the study of the evolution of enzymatic activities during germination. The validation method of characterization and quantification of (galacto)-manno-oligosaccharides constitutes the last part studied. Seven performance criteria calculated for each of the nine standards were used to validate the method. It has then been used to analyze locust bean gum hydrolysates. The results of this work are the first of a new research theme. They offer many opportunities and future applications.
[fr] Le cadre général de cette étude est la découverte de prébiotiques et plus spécifiquement la production d’oligosaccharides par hydrolyse enzymatique de la gomme de caroube. Ceux-ci, encore peu étudiés, pourraient présenter un riche potentiel en prébiotiques. Ce travail avait pour objectifs la purification de beta-mannanases situées dans des graines de caroube en germination, ainsi que la validation d’une méthode AEC-PAD applicable à l’analyse des hydrolysats de gomme de caroube. Dès lors, il a permis d’établir un protocole de concentration et de purification des beta-mannanases et de déterminer une technique de conservation adéquate des extraits enzymatiques. Une méthode originale d’évaluation de l’activité enzymatique par mesure de la viscosité a également été mise au point et permet notamment l’étude de l’évolution des activités enzymatiques au cours de la germination. La validation d’une méthode de caractérisation et de quantification des (galacto)-manno-oligosaccharides constitue le dernier volet étudié. Sept critères de performance calculés pour chacun des neufs standards ont permis de valider la méthode. Celle-ci a été utilisée pour analyser des hydrolysats de gomme de caroube. Les résultats de ce travail sont les premiers d’une nouvelle thématique de recherche. Ils laissent entrevoir de nombreuses possibilités et applications futures.
