Title: Technical and clinical evaluation of the VITROS Immunodiagnostic Products 25-OH Vitamin D Total Assay - comparison with marketed automated immunoassays and a liquid chromatography - tandem mass spectrometry method

Author, co-author: CAVALIER, Etienne; ROUSSELLE, Olivier; FERRANTE, Nunzio; CARLISI, Ignazia; LE GOFF, Caroline; Souberbielle, Jean-Claude

Abstract: Background: The study was conducted to evaluate the
technical and clinical performance of the VITROS ® Immunodiagnostic
Products 25-OH Vitamin D Total Assay, and
compare it with the performance of five marketed automated
assays and a liquid chromatography/mass spectrometry
reference method (LC-MS/MS).
Methods: Three hundred patient serum samples were used
to compare the correlation of the VITROS ® 25-OH Vitamin
D Total Assay with both the other immunoassays and the
LC-MS/MS method, using Passing-Bablok regression and
Bland-Altman analyses. Concordance of the diagnosis
of vitamin D status was calculated to test the agreement
between the different assays. In addition, samples containing
vitamin D2 were used to test the assay ’ s ability to
detect the D2 form of the vitamin.
Results and conclusions: These results from the VITROS ®
25-OH Vitamin D Total Assay generally correlated well
with those from most of the marketed immunoassays.
Cross-reactivity of the D2 form was calculated as being
close to 100%. Additionally, we found substantial variability
in performance amongst the various assays, which
suggests the need for optimisation and recalibration of
commercial methods.
Author, co-author: Nisolle, Michelle; DELBECQUE, Katty; PERRIER d'HAUTERIVE, Sophie; FIRQUET, Anne; CHANTRAINE, Frédéric

Abstract: OBJECTIVE: To present our experience in hysteroscopic removal of abnormally invasive placenta (AIP) residuals using bipolar energy. DESIGN: Case series. SETTING: University hospital. POPULATION: Sixteen patients with AIP residuals after 17 pregnancies. METHODS: Cases were identified by ultrasound, treated with hysteroscopic bipolar electrosurgery and oral contraceptives, and followed up by ultrasound or hysteroscopy. Nine subsequent pregnancies were described. MAIN OUTCOME MEASURES AND RESULTS: Complete removal of AIP residuals was achieved by hysteroscopic bipolar electrosurgery in all cases except one. No peroperative complications occurred. AIP residual recurred in one patient after a subsequent pregnancy and was successfully treated using the same procedure. CONCLUSIONS: AIP residual is a rare condition. Management by hysteroscopic resection using bipolar energy is save and feasible.
Author, co-author: Coudyzer, Pauline; Lemoine, Pascale; Jordan, Benedicte F.; Gallez, Bernard; Galant, Christine; Nisolle, Michelle; Courtoy, Pierre J.; Henriet, Patrick; Marbaix, Etienne

Abstract: Menstrual endometrial breakdown induced by estradiol and progesterone withdrawal is regularly attributed to vasospasm of spiral arteries causing ischemia and hypoxia. We investigated whether hypoxia actually occurred in an in vivo model of menstruation. Three complementary approaches were used to look for signs of hypoxia in fragments of human functionalis xenografted to ovariectomized immunodeficient mice bearing pellets-releasing estradiol and progesterone, and then deprived of ovarian steroids. Hormone withdrawal 21 d after grafting induced menstrual breakdown and MMP expression within 4 d. Local partial oxygen pressure (pO2) was measured by electron paramagnetic resonance using implanted lithium phtalocyanine crystals. In mice with hormone maintenance until sacrifice, pO2 was low one week after grafting (14.8+/-3.4 mmHg) but increased twofold from the second week when tissue was largely revascularized. After 3 wk, pO2 was not modified by hormone withdrawal but was slightly increased on hormone reimpregnation 4 d after removal (34.7+/-6.1 mmHg) by comparison with hormone maintenance (27.1+/-8.6 mmHg). These results were confirmed using fluorescence quenching-based OxyLite measurements. In a further search for signs of hypoxia, we did not find significant HIF1-alpha immunostaining, nor pimonidazole adducts after hormone withdrawal. We conclude that hypoxia is not needed to trigger menstrual-like tissue breakdown or repair in human endometrial xenograft.-Coudyzer, P., Lemoine, P., Jordan, B.
Author, co-author: Marini Djang'Eing'A, Roland; Fillet, Marianne; Vancauwenberghe, Roy; Crommen, Jacques; Frederich, Michel; Hubert, Philippe

Persistent Organochlorine Pollutants with Endocrine Activity and Blood Steroid Hormone Levels in Middle- Aged Men

Title: Persistent Organochlorine Pollutants with Endocrine Activity and Blood Steroid Hormone Levels in Middle- Aged Men

Author, co-author: Emeville, Elise; Giton, Frank; Giusti, Arnaud; Oliva, Alejandro; Fiet, Jean; Thomé, Jean-Pierre; Blanchet, Pascal; Multigner, Luc

Abstract: Abstract
Background: Studies relating long-term exposure to persistent organochlorine pollutants (POPs) with endocrine activities
(endocrine disrupting chemicals) on circulating levels of steroid hormones have been limited to a small number of
hormones and reported conflicting results.
Objective: We examined the relationship between serum concentrations of dehydroepiandrosterone, dehydroepiandrosterone
sulphate, androstenedione, androstenediol, testosterone, free and bioavailable testosterone, dihydrotestosterone,
estrone, estrone sulphate, estradiol, sex-hormone binding globulin, follicle-stimulating hormone, and luteinizing hormone
as a function of level of exposure to three POPs known to interfere with hormone-regulated processes in different way:
dichlorodiphenyl dichloroethene (DDE), polychlorinated biphenyl (PCB) congener 153, and chlordecone.
Methods: We collected fasting, morning serum samples from 277 healthy, non obese, middle-aged men from the French
West Indies. Steroid hormones were determined by gas chromatography-mass spectrometry, except for dehydroepiandrosterone
sulphate, which was determined by immunological assay, as were the concentrations of sex-hormone binding
globulin, follicle-stimulating hormone and luteinizing hormone. Associations were assessed by multiple linear regression
analysis, controlling for confounding factors, in a backward elimination procedure, in multiple bootstrap samples.
Results: DDE exposure was negatively associated to dihydrotestosterone level and positively associated to luteinizing
hormone level. PCB 153 was positively associated to androstenedione and estrone levels. No association was found for
chlordecone.
Conclusions: These results suggested that the endocrine response pattern, estimated by determining blood levels of
steroid hormones, varies depending on the POPs studied, possibly reflecting differences in the modes of action generally
attributed to these compounds. It remains to be investigated whether this response pattern is predictive of the subsequent
occurrence of disease.

Revisited rules of papilla level adjacent to single-tooth dental implants according recent implant designs. A retrospective study.

Title: Revisited rules of papilla level adjacent to single-tooth dental implants according recent implant designs. A retrospective study.

Author, co-author: LECLOUX, Geoffrey; Beaujan, Laurie; ROMPEN, Eric; LAMBERT, France

Abstract: Background:
According to Choquet and Tarnow principles (2001), a papilla is always present if the distance from the base of the contact point between two crowns to the crest of the bone is 5 mm or less. The arrival of new designs in implantology suggested more tissue preservation and so a possible evolution for the aesthetic treatments.
Aim:
This study was designed to determine whether the distance from the base of the contact point to the crest of the bone would correlate with the presence or absence of the interproximal papillae adjacent to single-tooth implants using the recent implant designs.
Methods:
A clinical, photographical and radiographical retrospective evaluation of the papilla level around single dental implants and their adjacent teeth was performed in the anterior maxilla in 34 patients restored with 39 implants. These implants were loaded at least 6 months and present a modified profile (Implants : NobelConcept and NobelActive, Nobel Biocare ; Bone Level, Straumann. Abutments : Platform-switching and curvy). Sixty-three papillae were available for evaluation. The Jemt index, the vertical distance between the base of the contact point and the bone crest, the distance from the papilla level to the contact point were measured. The measurements were rounded off to the nearest 0,001 millimeter and used with calibration computer programs (ImageJ) to be analyzed. The statistical analyses were realized with SAS program (SAS Institute, Cary, NC, USA).
Results:
When the measurements from the contact point to the crest of bone was 5 mm or less, the papilla was present 100% of the time. When the distance was 6 mm, the papilla was present 96% of the time. And when the distance was 7 and 8 mm, the papilla was present 57,14% and 50% of the time.
Conclusions and clinical implications:
These results show the influence of the bone crest on the presence or absence of papillae between implants and adjacent teeth. The data also show a positive influence for recent designs of implant and abutment systems. This designs shift the actual critical bone distance under the contact point from 5mm to 6mm and could improve the aesthetic outcome.
