&lt;br/&gt;Résumé: Introduction: The injuries of the suspensory ligament (SL) are important causes of lameness and financial losses in the equine industry. Ultrasound examination permitted to visualize some parts of the SL. The significance of “abnormal” findings is however not sufficiently known. Until now, few studies described the relationship between the ultrasonographic appearance and the exact morphology in histological sections. The aim of this study is to develop good techniques for cutting and staining the SL and to improve knowledge about the normal morphology of the SL.&#xD;
Methods: In this study, the SL of eight &lt;sound&gt; horses were collected. The body of the SL was divided in 3 thirds and sampling was realised within each third and between the thirds. The samples were embedded in paraffin or in Tissue-Tek for cryosections. The sections were stained with hematoxylin/eosin or Masson's trichrome. For 3 SL, ultrasounds were performed before sampling. The digital tip was maintained in physiological position owing to a press.&#xD;
Results: Most of the paraffin sections were shredded because of the hardness of the tissue. Cryosection revealed a better preservation of tissues. Only some freezing artifacts (holes) appeared on a few sections.  Muscles fibers surrounded by adipose tissue containing blood vessels were present mainly in the proximal and medium third of the SL whereas they were not found in the distal third. The remaining structure look like a tendon and was composed of collagen fibers, stained in green with the Masson's trichrome coloration.&#xD;
Conclusions: This study permitted to develop cutting and staining techniques for the SL and helped to map the adipose, muscular and tendinous parts within the SL. It lays down the bases of subsequent studies that will concern ultrasonographically examined digital tips of sound and pathological horses of different breeds and ages.</description>

&lt;br/&gt;Résumé: The third interosseous muscle (suspensory ligament, TIOM) is composed of connective tissue (CT) with a variable proportion of muscle (MT) and adipose tissue (AT). The aim of our study is to quantify the CT, MT and AT within the body and the branches of right thoracic and pelvic limbs TIOM in sound horses to determine whether there are differences in CT, MT and AT between age, sex, limbs and levels. Right limbs from 11 sound horses were collected. Samples from 6 levels of the TIOM were embedded in paraffin or in Tissue-Tek®. Most of the paraffin sections were shredded. Using the cryosection, some artefacts appeared. Cryoprotection was carried out, which produced the best results. Hematoxylin–phloxine–saffron and Hematoxylin–eosin gave a good contrast of colours between the tissues observed allowing the use of an image analysis programme to calculate percentage of each tissue within the TIOM. The percentage of MT and AT decreased significantly (P &lt; 0.0001), whereas the percentage of CT increased significantly (P &lt; 0.0001) with age and when descending from the proximal to the distal level of the TIOM. The percentage of MT was significantly higher (P &lt; 0.0001) in females than males, while the percentage of CT was significantly higher (P &lt; 0.0001) in males than females. The percentage of AT was significantly higher (P = 0.0278) in pelvic limbs than in thoracic limbs. These results confirm the variation in tissue composition within the TIOM of sound horses.</description>

&lt;br/&gt;Résumé: Dogue de Bordeaux (DDB) dogs are predisposed to a juvenile glomerulonephropathy (JGN), usually leading to an end-stage chronic kidney disease (CKD), mostly before 2 years of age. Since its first description a few years ago, the number of DDB dogs diagnosed with JGN has drastically increased in Europe. Furthermore, some DDB dogs might develop clinical signs far later than initially thought. As the mode of inheritance of this condition has not yet been elucidated, an early diagnosis of JGN is of central importance for breeders and owners.&#xD;
To ease screening of affected dogs, we first wanted to improve our knowledge of this ancient French breed by describing characteristics of healthy DDB dogs. Given available information, we indeed suspected that some biological specificity existed in this breed, which could affect diagnostic processes and lead to erroneous clinical decisions. We thus determined, in a first study, breed-specific biochemical and hematological reference intervals (RI). For most of the 29 measured biochemical analytes, using the manufacturer’s generic canine RI would not have had a major clinical impact, however breed-specific RI established for 6 of them were relevantly different. Limits of the RI for total proteins, total cholesterol, aspartate aminotransferase, alanine aminotransferase and lipase were higher, while they were markedly lower for total thyroxin. Among the 25 hematological RI that were determined and compared with generic canine RI established in the same laboratory, only those of some red blood cells and platelet indices were sufficiently different to have a significant impact on clinical decision. Most red blood cells indices had higher, whereas most platelet indices had lower RI.&#xD;
As we also hypothesized that some apparently healthy adult DDB could suffer from JGN, we decided to look, in a second study, for the biological abnormalities that were previously described in association with the disease. We thus recruited a large cohort of adult DDB and found that proteinuria (urine protein-to-creatinine ratio (UPC) &gt; 0.5) was present in 33%, and proteinuric CKD in 2% of the dogs. When assessed by UPC, proteinuria increased with age and was associated with changes in several biological variables. Urine specific gravity, plasma albumin and total calcium decreased, whereas cholesterol, fibrinogen and thyroid-stimulating hormone increased with UPC. Furthermore, changes in total renal volume, evaluated by ultrasound, were also described to be affected by magnitude of proteinuria. Having a UPC&gt;0.5 increased the probability of observing urinary granular casts by an odds-ratio of 4. Amplitude of biological variations observed in this study was however generally moderate.&#xD;
Finally, to investigate if proteinuric DDB could be suffering from a glomerular disease related to the JGN, we attempted in a third study to characterize the nature of their proteinuria. For that purpose, we used urinary proteins electrophoresis and specific tubular (retinol binding protein (uRPB) and N-acetyl-β-D-glucosaminidase (uNAG) enzyme) and glomerular (albumin (uAlb) and immunoglobulins G (uIgG)) urinary markers. One hundred and two DDB, including 2 dogs with proteinuric CKD, were retrospectively recruited for this study, irrespective of UPC values, to facilitate comparison. Final interpretations of urine proteins electrophoresis attributed a “normal” pattern to 39 dogs, a tubular pattern to 9 of them, a glomerular pattern to 38, and a mixed pattern to 16. Most of these results were highly correlated to UPC values. However, tubular and mixed patterns were only attributed to entire male dogs because of the presence of an isolated 25kDa band, which might be due to urinary contamination by seminal proteins. When results for urinary markers were compared to values obtained in non proteinuric (UPC≤0.2) DDB dogs, 37% of the borderline proteinuric dogs (0.2≤UPC&lt;0.5) had all four markers within reference limits. On the other hand, all proteinuric DDB dogs had elevated ratios of uAlb and uIgG when their concentrations were related to urinary creatinine (/c). Only 53% of them had an elevated uRBP/c and 35% an elevated uNAG/c, which corresponded to the dogs with highest UPC values. Results of this last study support the hypothesis that proteinuric DDB dogs have a glomerular disease which is likely related to JGN. Additionally, sensitivity, specificity, positive and negative likelihood ratio of electrophoresis to identify dogs with elevated uAlb/c were 94%, 92%, 11.85 and 0.07, in that order. These indices were respectively 90%, 74%, 3.43 and 0.13, to identify DDB dogs with abnormally elevated uIgG/c.; Les Dogues de Bordeaux (DDB) sont prédisposés à une glomérulopathie juvénile familiale (GFJ), initialement rapportée comme entraînant, avant l’âge de deux ans, les signes classiques d’une insuffisance rénale chronique (IRC). Depuis les premières communications autour de cette maladie, le nombre d’individus diagnostiqués en Europe n’a cessé de croître, et il semble que certains animaux ne développent de signes cliniques que tardivement. Le mode de transmission de cette affection n’a pas encore été élucidé et le dépistage précoce des animaux atteints est devenu l’une des préoccupations centrales des propriétaires de DDB. &#xD;
Il nous a d’abord semblé essentiel de décrire les éventuelles particularités sanguines du DDB cliniquement sain, afin de faciliter l’identification des animaux malades. Compte tenu des données disponibles, nous envisagions en effet que plusieurs spécificités biologiques raciales pouvaient exister et perturber le diagnostic de certaines affections chez le DDB. Lors d’une première étude nous avons donc déterminé les intervalles de référence (IR) biochimiques et hématologiques du DDB cliniquement sain. Parmi les 29 IR biochimiques spécifiquement établis pour le DDB et pour lesquels une comparaison qualitative avec les IR fournis par les fabricants des analyseurs était possible, 6 ont été jugés suffisamment différents pour influencer une décision clinique. Ainsi les limites de références des concentrations de protéines totales et de cholestérol total et les activités de l’aspartate transaminase, de l’alanine transaminase et de la lipase étaient-elles plus élevées, tandis que celles de la concentration de thyroxine totale étaient nettement plus basses. Parmi les 25 IR hématologiques établis pour la race et comparés statistiquement à ceux établis de façon similaire pour une population générale canine, seuls les IR de certains indices des globules rouges présentaient des limites de référence plus élevées tandis que celles des plaquettes étaient généralement plus basses. &#xD;
Une autre de nos hypothèses étant que certains chiens apparemment sains pouvaient être atteints de GFJ, nous avons recherché, dans une deuxième étude, la présence éventuelle des anomalies préalablement décrites lors de cette affection au sein d’une large cohorte de DDB. Cette étude a révélé que dans la population recrutée 2% des chiens étaient atteints d’IRC protéinurique et que la prévalence de la protéinurie (i.e. rapport des concentrations de protéines sur créatinine urinaires (RPCU) &gt; 0.5) d’origine vraisemblablement rénale était de 33%. De plus, cette protéinurie, évaluée quantitativement par le RPCU augmentait avec l’âge des chiens et était associée à des modifications de plusieurs variables biologiques : la densité urinaire, l’albuminémie et le calcium total diminuaient, tandis que le cholestérol, le fibrinogène et la thyréostimuline augmentaient avec le RPCU. Des variations du volume rénal total, évalué par l’intermédiaire de mesures échographiques, ont aussi été mises en évidence en fonction de l’amplitude du RPCU. De plus, le risque d’observer des cylindres granuleux dans les urines des chiens protéinuriques était multiplié par un facteur de 4. Les modifications de l’ensemble des variables étaient pour la plupart modérées.&#xD;
Enfin pour étudier l’hypothèse selon laquelle les DDB protéinuriques souffriraient d’une affection glomérulaire possiblement reliée à la GFJ, nous avons cherché à préciser la nature de leur protéinurie dans une troisième étude. Celle-ci a reposé sur l’électrophorèse des protéines urinaires et le dosage de deux marqueurs urinaires tubulaires (la protéine de liaison du rétinol (uRBP) et l’enzyme N-acétyl-β-D-glucosaminidase (uNAG)) et de deux marqueurs urinaires d’altérations glomérulaires (l’albumine (uAlb) et les immunoglobulines G (uIgG)). Cent deux chiens ont été inclus dans cette étude, dont deux chiens atteints d’IRC protéinurique. Afin de faciliter les comparaisons, les électrophorèses et les dosages de marqueurs urinaires ont été réalisés chez tous les chiens, quelle que soit la valeur du RPCU. Sur la base des résultats des électrophorèses, 39 chiens ont été considérés comme n’ayant pas de protéinurie anormale, 9 comme ayant une protéinurie d’origine tubulaire, 38 une protéinurie d’origine glomérulaire et 16 une protéinurie mixte. La plupart des résultats des électrophorèses étaient fortement corrélés au RPCU. Cependant, seuls les chiens mâles entiers se sont vus attribuer un profil tubulaire ou mixte, du fait de la seule présence d’une bande de 25 kDa, dont l’origine pourrait être une contamination des urines par des protéines séminales. Comparativement aux valeurs établies chez les DDB non protéinuriques (i.e. RPCU ≤ 0.2), les valeurs des 4 marqueurs urinaires étaient considérées comme normales pour 37% des chiens ayant une protéinurie douteuse (i.e. 0.2 ≤ RPCU &lt; 0.5). En revanche 100% des chiens protéinuriques présentaient des dosages d’uAlb et d’uIgG rapportés à la créatinine (/c) anormalement élevés, tandis que seulement 53% d’entre eux avaient un uRBP/c et 35% un uNAG/c anormalement élevés. Les chiens avec des uRBP/c et uNAG/c anormaux présentaient les valeurs de RPCU les plus élevées. Les résultats de cette troisième étude sont en faveur d’une protéinurie d’origine glomérulaire, vraisemblablement reliée à la GFJ, chez le DDB. Par ailleurs de bonnes performances diagnostiques de l’électrophorèse des protéines urinaires ont été démontrées pour identifier les chiens ayant des uAlb/c et d’uIgG/c anormalement élevés. Ainsi la sensibilité, la spécificité et les rapports de vraisemblance positif et négatif de la bande électrophorétique correspondant à l’albumine, évaluée semi-quantitativement, pour prédire une élévation anormale d’uAlb/c étaient-ils de 94%, 92%, 11.8, et 0.07, respectivement. En parallèle, la sensibilité, la spécificité, les rapports positif et négatif de vraisemblance de la présence d’une bande électrophorétique correspondante aux IgG pour identifier les chiens ayant un uIgG/c anormalement élevé étaient respectivement de 90%, 74%, 3.4 et 0.13.</description>

&lt;br/&gt;Résumé: Wildebeests carry asymptomatically alcelaphine herpesvirus 1 (AlHV-1), a gamma-herpesvirus inducing malignant catarrhal fever (MCF) to several ruminant species (including cattle). This acute and lethal lymphoproliferative disease occurs after a prolonged asymptomatic incubation period after transmission. Our recent findings with the rabbit model indicated that AlHV-1 infection is not productive during MCF. Here, we investigated whether latency establishment could explain this apparent absence of productive infection and sought to determine its role in MCF pathogenesis. First, whole-genome cellular and viral gene expression analyses were performed in lymph nodes of MCF-developing calves. Whereas a severe disruption in cellular genes was observed, only 10% of the entire AlHV-1 genome was expressed, contrasting with the 45% observed during productive infection in vitro. In vivo, the expressed viral genes included the latency-associated nuclear antigen homolog ORF73 but none of the regions known to be essential for productive infection. Next, genomic conformation analyses revealed that AlHV-1 was essentially episomal, further suggesting that MCF might be the consequence of a latent infection rather than abortive lytic infection. This hypothesis was further supported by the high frequencies of infected CD8+ T cells during MCF using immunodetection of ORF73 protein and single-cell RT-PCR approaches. Finally, the role of latency-associated ORF73 was addressed. A lack of ORF73 did not impair initial virus replication in vivo, but it rendered AlHV-1 unable to induce MCF and persist in vivo and conferred protection against a lethal challenge with a WT virus. Together, these findings suggest that a latent infection is essential for MCF induction.</description>

This project aims to develop strategies to use milking robots in pasture in order to benefit from advantages regarding feeding costs as well as animal welfare and health. It aims to study the effects of grass height, animal, days in milk, rotation cycle number and distance from grazed paddocks on daily milk yield and milking number of grazed cows.</description>

La majorité des troupeaux traits avec un robot ne pratique pas le pâturage car les exploitants considèrent que cette méthode n'est pas conciliable avec le pâturage. La fragmentation des parcelles est aussi un frein au pâturage. La mise en place d'un robot dans une structure mobile permet d'utiliser le même robot à l'étable en hiver et dans les prairies distantes de l'exploitation en été. L'objectif de cette étude est de tester l'effet de facteurs environnementaux, l'animal, la complémentation, la distance entre le robot et la parcelle, les numéros du jour dans la parcelle et de cycle de rotation sur la production laitière (PL) et le nombre de traites (NT).</description>

&lt;br/&gt;Résumé: Objective: The present study was undertaken to investigate the population dynamics of the intermediate hosts of Fasciola gigantica and the levels of infection in the snails. This would allow identifying the most important transmission periods and suggesting optimal snail control for the area.&#xD;
Methods: Lymnaeid snails were monthly collected by hand picking in Ha Noi (Northern Vietnam) during the period from March 2010 to December 2011 and in Binh Dinh province (Central Vietnam) during the period May and September 2012. Collected snails from different sites were identified based on morphology and the partial 16S rDNA sequence and ITS-2 sequence. Snails were examined for the presence of Fasciola larval by the crushing method and confirmation by multiplex PCR analyses with PCR primer for Fasciola gigantica cathepsin L in parallel with the snail rDNA species specific primer. &#xD;
Result: The density of Lymnaeid populations underwent great changes in relation to the geographical locations and seasons. In Central Vietnam, the Lymnaeid populations reached the peak in the dry/rainy season (May) and decreased sharply in rainy/dry season (September). In contrast, in Hanoi, it reached the peaks in 2 periods (February to April and August to November) of rice cultivation or early stage of growing of rice, and greatly deceased when the rice becomes fully developed. Transmission of fascioliasis in Ha Noi were high when rice cultivation is performed,  while the permanent transmission can takes place through the year in Binh Dinh with peak transmission during the  dry season.</description>

&lt;br/&gt;Résumé: Testudinid herpesvirus infections in tortoises belonging to the Testudinidae family are well known for decades, but their pathogenesis remains poorly understood and treatments are often empirical. This study describes the in vitro evaluation of selected anti-herpesvirus compounds against Testudinid Herpesvirus 3 (THV-3). A-5021, a compound with known broad-spectrum anti-herpetic activity, showed to be 9 times more potent than acyclovir, with an EC50 of 13.2 µM and inducing a complete inhibition of viral replication at 37.7 µM. Initial pharmacokinetic parameters were determined after a single sub-cutaneous administration of 5 and 10 mg/kg in Hermann’s tortoises (Testudo hermanni, n=3). Blood samples were collected at different time points and plasma concentrations of A-5021 were determined. No adverse effects were clinically observed and plasma concentrations remained above the EC50 for 2.8 and 4.2 h after administration of 5 and 10 mg/kg, respectively. These preliminary data provide a basis for further proof-of-concept studies for a potential prophylactic or therapeutic treatment of THV-3 infection in tortoises</description>
