L’équipe travaille notamment sur la dépendance à l’alcool, la douleur, l’obésité, le bégaiement, etc… Pour ces recherches, elle utilise des environnements virtuels en 3D en temps réel. Ces environnements virtuels permettent de naviguer librement dans des scénarios, d’incorporer progressivement des sensations physiques et émotionnelles et de graduer les interactions au rythme des utilisateurs. AMPA receptors (AMPARs), one of three sub-groups of ionotropic glutamate receptors present in the central nervous system, are recognised for their involvement in long-term potentiation (LTP), and learning and memory processes.1 They represent a valid cognitive enhancer target, particularly in the fight against Alzheimer’s disease (AD).2,3 Benzothiadiazine 1,1-dioxides, such as BPAM121, have emerged as important allosteric modulators of AMPARs, working solely in the presence of the endogenous transmitter.4 Synthesis of BPAM121 labelled with fluorine-18 was proposed, to investigate the utility of this molecule as a PET probe in vivo, and evaluate its potential as an AD diagnostic tool (Figure 1). Vitamin B1, in the form thiamin diphosphate (ThDP), serves as an enzymatic cofactor for several enzymes involved in glucose metabolism, the Krebs cycle, and branched-chain amino acid biosynthesis. Vitamin B6 in its form as pyridoxal 5'-phosphate (PLP) is required as a cofactor for more than 140 enzymatic reactions in cells, primarily in amino acid, sugar and fatty acid metabolism. Mammals lack the ability to biosynthesize both vitamins B1 and B6 de novo, and plants represent one of their major sources. Symptoms of severe vitamin B1 deficiency include degeneration in the nerves and heart, a disease known as beriberi, which can be fatal. Acute vitamin B6 deficiency can lead to various chronic diseases in humans, including neurological disorders, cardiovascular disease and diabetes. Due to their importance in regions where vitamins B1 and B6 deficiencies are reported, we identified cassava and rice as good target crops for biofortification. In the present work, we performed a screening for B1 and B6 accumulation in selected varieties and landraces representative of the rice and cassava diversity. HPLC analysis allowed precise quantification of vitamin accumulation in selected genotypes contrasting for vitamin accumulation. We also report the first profiling of B1 and B6 vitamers in cassava storage roots and rice endosperm. This work is of primary importance given the reported varying degree of bioavailability for certain vitamers. Identification and expression analysis of B1 and B6 biosynthetic genes from rice and cassava was instrumental to find correlation between expression of selected biosynthetic genes and vitamin accumulation. Genetic engineering of vitamin biosynthetic pathways led to the generation of transgenic rice and cassava lines with high vitamin B6 content. Importantly, our Caco-2 cell assay demonstrated that field-grown storage roots from transgenic cassava lines accumulate high levels of bioavailable vitamin B6. Despite their key role in plant metabolism and their antioxidant activities, increased accumulation of vitamin B6 in transgenic rice and cassava lines could not be associated with improved tolerance to selected biotic or abiotic stresses under lab conditions. De novo protein design is a growing field in protein chemistry, where artificial proteins are first designed in silico and then validated experimentally. Our group has a long tradition in the design of artificial (β/α)8-barrel proteins, known as Octarellins [1]. This fold, also known as TIM-barrel, is widespread in nature, particularly in enzymes, and represent an interesting target for therapeutic or biological applications. Here we present a novel protocol for the de novo design of TIM-barrels. Both Rosetta and Modeller modeling softwares were used to create the backbone structure of a TIM-barrel fold and to generate more than 10.000 artificial amino acid sequences. Stability was tested for the more interesting models by running molecular dynamics (MD) simulations, using GROMACS. The best models were chosen for protein production and preliminary biophysical characterization. The design of artificial proteins and the improvement of bioinformatics tools for protein modeling, structure prediction and MD simulations seem to be essential for a comprehensive knowledge of protein structure in general, as well as for an optimal use of the massive amount of data resulting from the numerous genome sequencing projects. Proxima Cen b is possibly the nearest rocky exoplanet orbiting in the habitable zone of its star and might represent by consequent a formidable opportunity for astrobiology. In this presentation, we discuss several possible atmospheric compositions and present the corresponding infrared spectrum computed with modern planetary atmosphere models. To be specific, we consider (1) a bare planet, which has lost its atmosphere; (2) a water-ocean planet; (3) an Earth-analog planet; and (4) a planet similar to Earth but with a lower O2 pressure (< 1mbar) that produces a false positive for the triple signature (H20, O3, and CO2). We discuss the information contained in each infrared spectrum and the possibility to constrain the nature of the planet by remote sensing. We end this presentation by describing an instrumental concept recently proposed to ESA and optimised for this task. [less ▲]

The discovery of an increasing number of terrestrial planets around nearby stars marks the dawn of a new era in the exoplanet field: the characterization and understanding of their atmospheres. To make significant progress, it becomes clear that a large number of exoplanetary atmospheres have to be studied at various wavelengths. This is particularly relevant for identifying possible bio-signatures. In this talk, we present a concept of a space-based mid-infrared nulling spectrograph that can characterize a large number of exoplanetary atmospheres and provide key information on their size, surface temperature, and the presence of key molecules such as CO2, H2O, CH4 and O3. The proposed mission concept would be particularly suited to characterize Proxima Cen b. [less ▲]

Vortex coronagraphs are among the most promising solutions to perform high contrast imaging at small angular separations from bright stars. They enhance the dynamic range at very small inner working angle (down to the diffraction limit of the telescope) and provide a clear 360 degree discovery space for high-contrast direct imaging of exoplanets. In 2013, we installed and commissioned an L-band coronagraph in LBTI/LMIRCam and obtained outstanding images of the four planets around HR8799 during the first hours on sky. In this presentation, we will present the results of the latest data reduction performed with the VIP software that is developed at the University of Liège and that features state-of-the-art image processing algorithms inherited from the field of background subtraction in computer vision (including machine learning algorithms and low rank modeling algorithms). We will also present the results obtained with the second L- and M-band coronagraph that was recently installed in LMIRCam to enable binocular Vortex observations. During the first observations (October 2016), we tested and validated a new real-time post-coronagraphic tip-tilt sensing technique (called QACITS) to quickly align each beam on the center of their respective Vortex coronagraph and obtained observations of a young star showing disk features near the resolution limit of each aperture. Finally, we will present some exciting prospects for the Vortex coronagraph that will be installed on VISIR and ELT/METIS. [less ▲]

C'est une guerre qui est menée sur de nombreux fronts, du Texas à l'Arabie saoudite, des salons feutrés de l'ONU et de l'organisation mondiale du commerce aux conseils d'administration des majors du pétrole, des laboratoires chinois en panneaux photovoltaïques aux champs éoliens de la mer du Nord. Dans ce tumulte guerrier, chaque nation essaie de défendre ses propres intérêts, d'avancer ses pions de la manière la plus intelligente possible, sachant très bien qu'une énergie abondante et bon marché est une des clés d'une économie puissante. Au cours de ce cours-conférence, le Prof. Ernst décodera cette guerre qui est en train de se passer sous nos yeux et que souvent, l'on ne comprend pas. Il expliquera à quel point la technologie pourrait changer la géopolitique de l'énergie et plonger certaines nations dans la tourmente. Il mettra aussi en garde contre tout espoir excessif en la technologie pour garantir un approvisionnement en énergie suffisant, tellement la soif d'énergie de nos sociétés est grande. At least 9 CGM species are infecting cassava and CGM diversity is particularly high on the African continent. In order to engineer resistance against African cassava mosaic virus (ACMV), we developed transgenic cassava expressing dsRNA against the ACMV Rep (AC1) sequences that are conserved amongst CGMs. Transgenic dsAC1 cassava lines were selected based on the accumulation of transgene-derived small RNAs. We established a rapid and high-throughput agroinoculation method in order to assess virus resistance in cassava. Selected transgenic cassava lines displayed robust resistance against increasing loads of ACMV delivered by gene gun and by agroinoculation. We also observed a fast recovery phenotype in transgenic lines following graft inoculation of ACMV. Best performing dsAC1 cassava lines were also inoculated with South African cassava mosaic virus (SACMV) and East African cassava mosaic virus (EACMV). Recovery from SACMV and EACMV infections in transgenic cassava lines was delayed as compared to the recovery phenotype observed with ACMV infection. Collectively, our results suggest that high similarity between transgene-derived dsRNAs and target virus sequences is required to engineer transgenic plants rapidly recovering from virus infection. Selected transgenic dsAC1 cassava lines also showed delayed development of CMD symptoms in the field. Molecular analysis of CGM species in the field is being performed in order to better characterize the level of resistance of dsAC1 lines against multiple CGM species. [less ▲]

To be specific, we consider (1) a bare planet, which has lost its atmosphere; (2) a water-ocean planet; (3) an Earth-analog planet; and (4) a planet similar to Earth but with a lower O2 pressure (< 1mbar) that produces a false positive for the triple signature (H20, O3, and CO2). We discuss the information in each infrared spectrum and the possibility to identify the nature of the atmospheres by remote sensing. With the aim of studying the recent Greenland ice sheet Surface Mass Balance (SMB) decrease with respect to the last century, we have forced the regional climate MAR model (version 3.6) with the ERA-Interim (1979-2015), ERA-40 (1958-2001), NCEP1 (1948-2015), NCEP2 (1979-2015), JRA-55 (1958-2015), 20CRv2(c) (1880-2012) and ERA-20C (1900-2010) reanalysis. While all of these forcing products are reanalyses, MAR simulates differences in SMB over the common period. A temperature correction of +1°C (resp. -1°C) had notably to be applied to the MAR boundary conditions given that ERA-20C (resp. 20CRv2) is ~1° colder (resp. warmer) over Greenland than ERA-Interim data over 1980-2010. Comparisons with PROMICE daily temperature measurements valid these corrections. In most of regions, the SMB discrepancies between the different simulations are not significant except in the South-East where the maximum of precipitation occurs and where SMB measurements are missing. This suggests that uncertainties in the current SMB reconstruction remain and that observations are still needed. Comparisons with SMB measurements from the PROMICE data set, ice cores and satellite derived melt extent allows to select the best reanalysis forced data set. All of these simulations show that i) the period 1961-1990 usually chosen as reference for SMB and ice dynamics (stable ice sheet) over Greenland is a period when the SMB was abnormally high in respect to the last 120 years; ii) SMB has been significantly decreasing after this reference period due to increasing melt. Both ERA-20C and 20CRv2 forced simulations suggest a precipitation increase since the beginning of the last century and the ERA-20C forced simulation only suggests that SMB during the 1920-1930 warm period over Greenland was comparable with the SMB of the 2000's. Finally, the sensitivity of switching on the erosion of the snow by the wind in MARv3.6 will be discussed. [less ▲]

We revisit the thermoelectric (TE) transport properties of two champion materials, PbTe and SnSe, using fully first principles methods. In both cases the performance of the material is due to subtle combinations of structural effects, scattering, and phase space reduction. In PbTe anharmonic effects are completely opposite to the predicted quasiharmonic evolution of phonon frequencies and to frequently (and incorrectly) cited extrapolations of experiments. This stabilizes the material at high T, but also tends to enhance its thermal conductivity, in a non linear manner, above 600 Kelvin. This explains why PbTe is in practice limited to room temperature applications. SnSe has recently been shown to be the most efficient TE material in bulk form. This is mainly due to a strongly enhanced carrier concentration and electrical conductivity, after going through a phase transition from 600 to 800 K. We calculate the transport coefficients as well as the defect concentrations ab initio, showing excellent agreement with experiment, and elucidating the origin of the double phase transition as well as the new charge carriers. The impacts on different parts of the electrical system are presented: the day-ahead energy market, the secondary reserve and the distribution system. The modeling techniques used to carry the analysis are optimization, game theory and agent-based modeling. Since flexibility is the base block of this thesis, two methods to obtain flexibility from an actual consumption process are broached: direct control of a portfolio of heat pumps and dynamic pricing to control electric heaters and boilers.
