DNA methylation and histone deacetylation are two key epigenetic modifications that play central role in regulation of gene expression. Several studies have shown that histone deacetylases (HDAC) and DNA methyltransferases (DNMT) inhibitors are potent anti-angiogenic compounds. Though combination of HDAC and DNMT inhibitors are now being examined in clinical trials of hematological malignancies, little work has been done to understand the effect of this combination on physiological and tumoral angiogenesis. We have designed and tested a family of twin drugs with intrinsic HDAC and DNMT inhibitory activities in relevant models of angiogenesis in vitro (Human Umbilical Vein Endothelial Cells – HUVEC and aortic ring) and in vivo (chick chorioallantoic membrane and Zebrafish). We have identified a lead compound having quantifiable anti-angiogenic effect without cytotoxicity affecting global histone acetylation and DNA methylation levels. In order to elucidate its anti-angiogenic mechanism, we characterized gene expression pattern simultaneously with the methylation profile of HUVEC cells treated with the lead compound and reference epigenetic modulators. This approach based on parallel microarray analyses permitted us to underscore a list of genes exclusively affected by the lead compound but not by other HDAC or DNMT inhibitors. These genes were then analyzed using the Ingenuity Pathway software revealing potential involvement of a subset of genes in angiogenesis. Our present aim is to validate the expression levels of a series of genes with respect to epigenetic mechanisms (histone modifications and DNA methylation). Finally, the biological relevance of the target genes will be explored by RNA silencing. Hence, we are using these novel epigenetic modulators as a tool to understand the regulatory mechanism of angiogenesis and to develop effective approaches to treat cancer. [less ▲]

The Chaucerian attitude to the set of viragos of his Man of Law's Tale, Monk's Tale, and Knight's Tale is enigmatic. By discrediting the reliability of his narrators, he blurs the categories of difference that they strongly advocate, thus creating a space in which the medieval racial and racist clichés concerning Oriental viragos may be reconsidered. Attention has been drawn to Chaucer’s formation of ‘English’ identity in the poet’s late writings. The writer’s exclusive use of London English, a colonized language, was a political decision, but he resists easy binaries of identification and his English nationalism is a ‘present-looking and eclectic multiculturalism.’ The Knight’s as well as the Man of Law’s tales are retellings of imperial aggression but also accounts of failed imperialist ambitions. In a way, the Monk addresses a similar issue, but here the initial imperial belligerence comes from the Orient and there is a return to Western imperial order. Needless to say, narratives dealing with military campaigns and crusades, and deviant women were of high relevance to a late fourteenth-century audience. The period marked a turning point from universal forms of truth to a greater interest in individual identities; and late medieval English society was not monolithic, and neither was the social sample going to Canterbury. In this chapter, we first present an original two-phase flow model for the water-sediment mixture, acting as a unified basis for all our subsequent developments. Next, we focus on two topics in which we have made original contributions, namely sediment routing on alluvial and non-alluvial beds and modelling of transient hyperconcentrated flows. In both cases, we use our original two-phase flow modelling framework to derive specific governing equations, for which we discuss their mathematical properties, detail appropriate finite volume numerical scheme and demonstrate their validity through a number of test cases. Key factors in the formation of cell aggregates in tissue engineering and other fields are the cell-cell and cell-matrix interactions. Other important factors are culture conditions such as nutrient and oxygen supply and the characteristics of the environment (medium versus hydrogel). As mathematical models are increasingly used to investigate biological phenomena, it is important that processes such as cell adhesion are adequately described in the models. Recently a technique was developed to incorporate cell-cell and cell-matrix adhesion in continuum models through the use of non-local terms. In this study we apply this technique to model adhesion in a cell-in-gel culture set-up often found in tissue engineering applications. We briefly describe the biological issues underlying this study and the various modeling techniques used to capture adhesive behaviour. We furthermore elaborate on the numerical techniques that were developed in the course of this study. Finally, we consider a tissue engineering model that describes the spatiotemporal evolution of the concentration of cells, matrix, hydrogel, matrix degrading enzymes and oxygen/nutrients in a cell-in-gel culture system. Sensitivity analyses indicate a clear influence of the different adhesive processes on the final cell and collagen density and distribution, demonstrating the significance of cell adhesion in tissue engineering and the potential of the proposed mathematical technique. Alzheimer’s disease (AD) is characterized by a progressive loss of controlled cognitive processes (processes requiring mental effort and attentional resources), and functional neuroimaging at early stages of AD provides an opportunity to tease out the neural correlates of controlled processes. Controlled and automatic memory performance was assessed with the Process Dissociation Procedure in 50 patients diagnosed with questionable Alzheimer’s disease (QAD). The patients’ brain glucose metabolism was measured using FDG-PET. After a follow-up period of 36 months, 27 patients had converted to AD, while 23 remained stable. Both groups showed a similar decrease in controlled memory processes but preserved automatic processes at entry into the study, suggesting that impairment of controlled memory would not be specific for AD. Patients who subsequently converted to Alzheimer type dementia showed significantly decreased brain metabolism at baseline compared to stable QAD in associative cortices known to be involved in AD (the left precuneus, the right inferior parietal lobule and bilateral middle temporal cortex).Voxel-based cognitive and metabolic correlations showed that a decrease in controlled memory processes was preferentially correlated with lower activity in the dorsomedial prefrontal and posterior cingulate cortices in very early AD patients. The dorsomedial prefrontal cortex would play a role in controlled memory processes as they relate to reflective and monitoring processes, while the posterior cingulate cortex is involved in the controlled access to previously encoded episodes. In stable QAD patients, reduced controlled performance in verbal memory correlated with impaired activity in the left anterior hippocampal structure, which would alter the reactivation of associations created at encoding. In this paper, we introduce a min max approach for addressing the generalization problem in Reinforcement Learning. The min max approach works by determining a sequence of actions that maximizes the worst return that could possibly be obtained considering any dynamics and reward function compatible with the sample of trajectories and some prior knowledge on the environment. We consider the particular case of deterministic Lipschitz continuous environments over continuous state spaces, nite action spaces, and a nite optimization horizon. We discuss the non-triviality of computing an exact solution of the min max problem even after reformulating it so as to avoid search in function spaces. For addressing this problem, we propose to replace, inside this min max problem, the search for the worst environment given a sequence of actions by an expression that lower bounds the worst return that can be obtained for a given sequence of actions. This lower bound has a tightness that depends on the sample sparsity. From there, we propose an algorithm of polynomial complexity that returns a sequence of actions leading to the maximization of this lower bound. We give a condition on the sample sparsity ensuring that, for a given initial state, the proposed algorithm produces an optimal sequence of actions in open-loop. Our experiments show that this algorithm can lead to more cautious policies than algorithms combining dynamic programming with function approximators. [less ▲]

The performance of a market timer can be measured through the Treynor and Mazuy (1966) model, provided the regression alpha is properly adjusted by using the cost of an option-based replicating portfolio, as shown by Hübner (2010). We adapt this approach to the case of multi-factor models with positive, negative or neutral betas. This new approach is applied on a sample of hedge funds whose managers are likely to exhibit market timing skills. We stick to funds that post weekly returns, and analyze three hedge funds strategies in particular: long-short equity, managed futures, and funds of hedge funds. We analyze a particular period during which the managers of these funds are likely to magnify their presumed skills, namely around the financial and banking crisis of 2008. Some funds adopt a positive convexity as a response to the US market index, while others have a concave sensitivity to the returns of an emerging market index. Thus, we identify “positive”, “mixed” and “negative” market timers. A number of signs indicate that only positive market timers manage to acquire options below their cost, and deliver economic significant performance, even in the midst of the financial crisis. Negative market timers, by contrast, behave as if they were forced to sell options without getting the associated premium. We interpret this behavior as a possible result of fire sales, leading them to liquidate positions under the pressure of redemption orders, and inducing negative performance adjusted for market timing. South East Asia displays a high level of mammal endemism and the highest number of threatened and data deficient mammal species. However, the South East Asian biodiversity is still highly unexplored. Because of the runaway global changes, a better biological knowledge of this region is urgently required to improve the conservation and the management of its biodiversity. The first aim of this chapter is to present recent published data on a biodiversity inventory of the Rattini murine rodents from this region based on molecular markers (Page`s et al., 2009). In this first study, we applied the method proposed by Pons et al. Our second aim concerns the intraspecific genetic structure of a rare and threatened South East Asian mammal species: the murine rodent Leopoldamys neilli, endemic to karst habitats . In this latter study, our results evidenced a high geographic structure of the genetic diversity of this species. The observed highly divergent genetic lineages would have to be considered as distinct evolutionary units or at least as Management units. These results are essential for the best conservation issues of species endemic to karsts and to South East Asia, in general. In this chapter, we therefore highlight that South East Asia would not be only a hotspot of interspecific but also of intraspecific biodiversity. [less ▲]

New energy conversion technologies are required in order to ensure the production of electricity without generating environmental pollution. Among them, low-grade heat sources or low power heat sources present an important potential of development. The Organic Rankine Cycle (ORC) is a well known technology since the early 80’s. A large amount of ORC power plants have been built, mainly for geothermal, waste heat recovery and combined heat and power applications. This technology shows a number of advantages over the traditional steam Rankine cycle that make it more profitable for power plants with a limited electrical output power (typically lower than 1 MWe), despite a lower efficiency. The optimization of the ORC is quite different from that of the steam cycle, mainly because of the heat source temperature limitation, and because there is usually no constraint regarding the vapour quality at the end of the expansion. This chapter presents an overview of the current state of the art in the ORC technology and exposes the main target applications. The modelling of such a cycle is described and issues such as fluid selection, optimization or control of the cycle are thoroughly reviewed. [less ▲]
