Les biotechnologies font régulièrement appel à la β-oxydation pour produire des composés à haute valeur ajoutée ou dépolluer des effluents industriels. Cette voie métabolique est difficile à maîtriser car elle implique un nombre réduit d'enzymes pour une voie très longue. En particulier, la production d'intermédiaires réactionnels est délicate car les enzymes de la voie de reconsommation sont souvent les mêmes que celles impliquées dans la production. Ainsi, la production de γ-décalactone à partir de ricinoléate de méthyle chez Y. lipolytica implique la β-oxydation (Endrizzi et al., 1996). Cette levure est un bon modèle pour étudier le métabolisme des acides gras car elle possède cinq acyl-CoA oxydases (Aox) catalysant la première étape de la β-oxydation (Wang et al., 1999). L'effet d'une invalidation des gènes codant pour les Aox a été étudié sur la production de γ-décalactones. En fonction des Aox présentes dans les souches, différentes lactones sont produites et leur reconsommation peut être nettement ralentie. Cette étude montre qu'on peut maîtriser la β-oxydation en modifiant le matériel génétique des souches et qu'on peut ainsi améliorer les productions existantes mais aussi produire de nouvelles molécules d'intérêt. Many studies have analysed psychopathological dimensions of relationship that adolescent have with food and body. But few researches have focused attention on links between psychosocial dimensions such as social representations (Moscovici, S. 1961; Lalhou, S.1995) and adolescent typical practices of eating (with family and peers) and body (diet and sport). This research is aimed to describe and to analyse representations and practices of eating and body of 60 adolescents (30 females, 30 males) equally distributed on high and low social class aged from 14 to 16 years. Hypothesis supposes changes of eating and body representations and practices according to sexe and social class of subjects. A multimethodogical approach was adopted. Data were collected in Avril 1999 in two roman colleges. Qualitative and quantitative instruments were administered in this order: a) associative network (words association), b) semi-structured interviews (18 questions about eating and body ideas and practices), c) body drawing, d) eating family drawing, e) questionnaires (Eating Attitudes Test, Eating Disorders Inventory 2). A preliminary interviews content analysis with ALCESTE (Reinert, M.1993) draw out 8 discourse classes. Four classes are linked to eating representations and practices: a) Eating Family Practice (16,85 %); b) Eating with Friends (8, 61%); c) Eating quantity and preferences (15, 97%); d) Daily meals organisation (7,40%). Others four classes are linked to body representations and practices: e) Eating Problems (20, 13%); f) Body Health and beauty (8,07%); g) Diets and Sports ( 10,86%); h) Food quality (12, 10%). Results show a general picture of eating and body representations and practices. Eating with family is a very important social situation: a script characterised by social actors, rules, actions and contexts is outlined. Food intake control and food preferences seem pregnant dimensions in identity construction process face to family and peers. Naive theories about eating disorders are constructed. Finally diet, sport and food quality seem central in shaping and modelling body. [less ▲]

The in vitro inhibitory effect of SR 27417, an antagonist of the platelet-activating factor (PAF) receptor, on PAF-induced platelet aggregation was studied in blood collected from seven healthy Friesien calves. Inhibitory effects of SR 27417 were determined at thirteen different concentrations (0.1-400 nM) by using the dose-response curves of PAF on calf platelet aggregation. In the presence of SR 27417, the maximal slopes of aggregation (%/min) induced by low and high concentrations of PAF were significantly different from the control values obtained without an antagonist at p < or = 0.05 and p < or = 0.01 respectively. In vitro PAF-induced calf platelet aggregation was dose-dependently inhibited by SR 27417. The drug inhibited PAF-induced platelet aggregation in a competitive reversible manner (pA2 = 10.46 +/- 2.36 mol x L(-1)). In conclusion, the results of our study showed that addition of SR 27417 to bovine platelet in vitro inhibits PAF-induced platelet aggregation. [less ▲]

This study is carried out to determine ovine pregnancy associated glycoprotein (oPAG) levels in the serum of Berrichone ewes throughout gestation and the first month post partum. The levels were determinated with heterologous RIA using bovine PAG as standard and tracer and rabbit antiserum against oPAG. The level of oPAG increased from the 4th week of pregnancy till 12-13th week and decreased slowly till 17th week, when increased rapidly and reached the highest level (around 600-700 ng/ml). In the sheep carrying twins, we observed higher levels of oPAG than in the uniparous sheep. After lambing, oPAG decreased rapidly in 4 week to basal values. The oPAG concentrations in the animals carrying twins was higher when comparised to single lamb. We concluded that the oPAG level depends on the number of lambs and term of pregnancy. The estimation of oPAG concentrations in the blood can be used as a indicator for pregnancy diagnosis. [less ▲]

The E. hirae DNA segment lacks the genes which in E. coli encode the ligases Ddl, MurC, MurE and MurF and the integral membrane protein FtsW. Nanoparticles can be used as an attractive carrier for the controlled delivery of drugs by the oral route. This nanoparticular delivery system can protect an active ingredient from enzymatic degradations and enhance its absorption from the intestinal mucosa. We studied the development of a gastroresistant dosage form for the oral administration of nanoparticles. Fast disintegrating pellets were used for the administration of nanoparticles and for their transit to the site of absorption. A gastroresistant film was applied onto the pellets to protect the nanoparticles and/ or the active substance from the acid gastric environment. The integrity of the nanoparticle size was assessed after two critical manufacturing steps: the extrusion/spheronisation process and the coating process. The determination of the nanoparticles size distribution was performed by photon correlation spectroscopy (PCS). The comparison of the PCS results between the original nanoparticles and the nanoparticles incorporated in the pellets shows a very low aggregation. Moreover the coating process was not a critical step since it was not responsible for an additional agglomeration of the nanoparticles. In conclusion, our manufacturing process allows the redispersion of the nanoparticles with a little change of the size distribution. In a next study we well try to quantify the release of the nanoparticles from the pellets. [less ▲]

AGM-1470, a potent angiogenesis inhibitor, is already engaged in phase I clinical trials because of its effectiveness to restrain tumor growth and its lack of major side effects. Recently, we showed that AGM-1470 stimulates in vitro human B lymphocyte proliferation through T lymphocytes. These data prompted us to explore the in vivo effects of AGM-1470 on the immune system in a mouse model. In this study, we showed that AGM-1470, in synergy with phytohemagglutinin, stimulates the proliferation of murine lymphocytes isolated from lymph nodes. This effect was similar to the one observed with human lymphocytes. When injected subcutaneously or intraperitoneally into mice at pharmacological doses, AGM-1470 induced a significant increase of axillary and mesenteric lymph nodes, respectively. Histological and morphological analyses showed that this phenomenon is mostly due to a hyperplasia of the germinal centers. On average, the area of the germinal center of lymph nodes from AGM-1470-treated mice were three times larger than in lymph nodes from control mice. Interestingly, no effect was observed when AGM-1470 was injected subcutaneously into T-deficient nude mice. Our data demonstrate that AGM-1470 stimulates B cell proliferation in vivo as suggested by the in vitro experiments. This effect should be taken into account in the follow-up of patients treated with this molecule and calls for additional studies to determine the biological consequences of such a stimulation on the host immune system. A DNA-methyl green colorimetric microassay for the detection of agents that interact with DNA was assessed with reference molecules (doxorubicin, ellipticine, melinonine F, dihydroflavopereirine, cis-Pt, mitomycin C, emetine or colchicine). This test showed some promise as a screening technique for substances that intercalate partly or totally into the DNA helix but it is not suitable for substances that bind covalently to DNA. An application of this test to a number of indole alkaloids was also performed. Cryptolepine, serpentine, alstonine and matadine induced a reduction of the colour of the solution, so indicating an interaction with DNA. [less ▲]

Foaming properties of surfactin were investigated and compared to those of sodium dodecyl sulfate (SDS) and bovine serum albumin (BSA). Foams were formed by a bubbling technique. Evolution of the foam volume and the liquid in the foam was monitored with optical and conductimetric methods to characterize foam formation and stability. Excellent foaming properties of surfactin were shown by its higher ability to form and stabilize the foam at a concentration as low as 0.05 mg/mL, in comparison with SDS and BSA. Surfactin produced a foam with intermediate maximum density and stabilized the liquid in foam, as well as BSA. [less ▲]
