Isoretulinal is a new alkaloid isolated from Strychnos variabilis rootbark. It can also be easily prepared by oxydation of isoretuline. This alkaloid shows an antimitotic effect at a dose of 10 micrograms/ml on different strains of cancer cells. Moreover isoretulinal possesses an analgesic activity. However this alkaloid is sixty-fold less toxic than strychnine (intravenous acute toxicity in mice). [less ▲]

Titers of circulating antigalactosyl antibodies (a-Gal Ab) were assessed by passive hemagglutination using rabbit red blood cells in 40 normal subjects, in 14 patients with immunodeficient states, in 47 patients with active rheumatoid arthritis (RA), and in 15 patients with an Henoch-Schonlein disease (HS). Titers of controls ranged from 1:16 to 1:64. All immunodeficient patients exhibited very low titers (1:1). On the contrary, the existence of an enhanced humoral immune response status, as observed in RA, was not reflected by a parallel increase of a-Gal Ab titers. However, in this disease, a strong relationship existed between titers exceeding control values (greater than 1:64) and the prior occurrence of renal injury under gold or D-penicillamine therapy. Lastly, the discovery of elevated titers (greater than 1:64) in HS only when renal involvement occurred further suggests that such antibodies reflect a renal injury. [less ▲]

A procedure to standardize the position of an esophageal catheter (eso-cat) tip, used to measure intrathoracic pressure, was tested in ten healthy Dutch Friesians. The cattle were in normal condition with body weights (BW) between 52 and 670 kg and thoracic perimeters (TP) between 80 and 210 cm. The position of the neck and head was standardized. The eso-cat was introduced via the nose into the thoracic portion of the esophagus. The distance between the nares and the eso-cat tip (Lcat), which was positioned between the crossing point with the aorta and the caudal mediastinal lymph nodes, was measured for each animal. The regression equation, calculated between the Lcat and the BW and TP was, respectively: Lcat (cm) = 65 + 0.115 x BW (kg) (r2 = 0.99) and Lcat (cm) = 22 + 0.535 x TP (cm) (r2 = 0.97). The corresponding residual standard deviations were respectively 2.52 and 4.37 cm. The multiple and curvi-linear regression equation did not give a significantly better fit of the data [less ▲]

Techniques are now available for culturing well characterized and purified Schwann cells. Therefore, we investigated the role of fibronectin in the adhesion, growth, and migration of cultured rat Schwann cells. Double-immunolabeling shows that, in primary cultures of rat sciatic nerve, Schwann cells (90%) rarely express fibronectin, whereas fibroblasts (10%) exhibit a granular cytoplasmic and fibrillar surface-associated fibronectin. Secondary cultures of purified Schwann cells do not express fibronectin. Exogenous fibronectin has a small effect on promoting the adhesion of Schwann cells to the substrate and does not significantly affect cell morphology, but it produced a surface fibrillar network on fibronectin on the secondary Schwann cells. Tritiated thymidine autoradiography revealed that addition of fibronectin to the medium, even at low concentrations, markedly stimulates Schwann cell proliferation, in both primary and secondary cultures. In addition, when cell migration was measured in a Boyden chamber assay, fibronectin was found to moderately, but clearly, stimulate directed migration or chemotaxis. [less ▲]

The main alkaloid of an extract with CHCl3 at pH 5 has been isolated and identified as descarbomethoxydihydrogambirtannine; that alkaloid is found for the first time in the family of Loganiaceae. The structure of usambaridine Vi, alkaloid previously isolated from leaves, is now elucidated by analysis of NMR and UV spectra. In other respects the curarizing activity of African Strychnos (e.g. Strychnos usambarensis) is not due to isostrychnine as erroneously written by TURKOVIC (J.Pharm Belg. 1976, 31, 277-286) but to quaternary alkaloids like C-dihydrotoxiferine, C-curarine, afrocurarine and C-calebassine (cf L.ANGENOT et al. Strychnos innocua is frequently cited in African traditional medicine. Root bark has been chemically screened.A small amount of alkaloids was detected as well as loganic acid. The plant contains at least two alkaloids that are devoid of an indole skeleton. The authors suggest that they are monoterpenic and not very different from gentianine.
