The blockade of the renin-angiotensin system (RAS) improves the prognosis of patients with complications related to diabetes, hypertension or, in general, atherosclerosis. Several observational studies have suggested the use of a dual blockade of the RAS to benefit from a better cardiorenal protection. However, recent randomized controlled studies failed to demonstrate that a dual blockade exert a better protection than single blockade, but showed a higher risk for renal complications and hyperkalemia. To decrease the residual risk, other opportunities may be recommended such as reinforcement of low salt diet, use of supraphysiological dose of a monotherapy inhibiting the RAS (perhaps prescribed at the evening) or addition of an aldosterone antagonist. However, all these approaches, as dual therapy, may also increase the risk of hypotension and renal insufficiency and thus require to be used under strict medical supervision. [less ▲]

If the treatment of hypertension is very useful to decrease the cardiovascular risk, the way to get the normalisation of high blood pressure is difficult in spite of many families of antihypertensive agents with different mechanisms of action. To reach such objective, association of different drugs are most often required. But, the european guidelines for the management of hypertension have just been released and proposed the possibility to begin very-low-dose combination therapy first-line. These associations offer immediate higher efficacy, excellent tolerance and low cost. [less ▲]

Près de 50 % des patients atteints d’un cancer bénéficient, à un moment de leur trajet de soins, d’une irradiation. Celle-ci peut être administrée avec une intention curative ou palliative, en fonction de l’extension de la maladie, de l’état général du patient et de sa volonté. Le but d’une irradiation palliative, sera de contrôler localement la tumeur ou la métastase et, donc, de ralentir l’évolution du cancer. La radiothérapie peut également être utile pour supprimer un symptôme et, ainsi, être un des traitements de confort en fin de vie. La dose totale, la dose par fraction ainsi que la technique d’irradiation sont adaptées à l’intention du traitement. Cet article passe en revue les principales indications d’irradiation en fin de vie. [less ▲]

Involvement of the AP-1 (activator protein-1) transcription factor has been demonstrated previously in the regulation of cell proliferation and cell-cycle progression, in the control of cell migration, invasion and metastasis, and in signal transduction, stress responsiveness, DNA replication and DNA repair. YB-1 (Y-box-binding protein-1) has also been implicated in many of these processes. However, the mechanism by which YB-1 mediates these processes is poorly understood. In the present study, we report that overexpression of a transfected gene encoding YB-1 in human HeLa cervical carcinoma cells significantly represses the transactivation of a minimal AP-1 reporter construct in response to the tumour promoter PMA. YB-1 also represses mRNA expression and PMA-induced promoter transactivation of the endogenous AP-1 target gene encoding matrix metalloproteinase-12 (metalloelastase). YB-1 transrepression of both the minimal and matrix metalloproteinase-12 promoter reporter constructs is dependent on the AP-1 sequence. To identify new nuclear proteins that bind specifically to the AP-1 DNA-binding site, we devised a DNA-affinity-chromatography-based assay termed NAPSTER (nucleotide-affinity preincubation specificity test of recognition) and discovered a 49 kDa protein from human cancer cells that binds in a sequence-specific manner to the AP-1 DNA sequence. By tandem MS fragmentation sequencing analyses we determined that p49 is a YB-1. Immunoblotting of the NAPSTER-purified p49 protein using anti-YB-1 antibodies confirmed YB-1 binding to the AP-1 DNA sequence, as did gel mobility-supershift assays using YB-1 antibodies. This is the first report of YB-1 transrepression and interaction at the AP-1 DNA-binding site. [less ▲]

YBa2Cu3O7-δ thick films have been deposited onto Ag substrates by the Electrophoretic Deposition (EPD) technique. Different microstructures and electrical behaviours were observed depending on the starting powder. Coatings prepared from commercial powder displayed significant porosity and the superconducting transition width was found to be magnetic-field dependent. Films produced from home-made coprecipitated powder are denser but contain some secondary phases. No dependence of the resistive transition as a function of magnetic field (H ≤ 20 Oe) was observed in that case. [less ▲]
