GC/MS (Gas Chromatography coupled to Mass Spectrometry) is the analytical method of choice for carboxylic acids analysis because of good sensitivity, low limit of detection and the possibility to compare the pattern of fragmentation with existing databases for identification. However GC requires that the analytes are volatile. If it is not the case, the use of chemicals in order to perform the derivatization is mandatory, this may induce analytical bias. CE (Capillary electrophoresis) is a technique of choice for ions separation without prior treatments. The method is fast and do not require highly technical skills. A UV detector is the most common detection method; however electrospray mass spectrometry detection is recently gaining interest, while it really helps for structural information of the detected compounds. In this poster, the preliminary results of CE/MS analysis of several carboxylic acids are presented. All carboxylic acids are analyzed without any sample pretreatment. These acids looked at are from the “Citric Acid Cycle” including pyruvate and some isotope labeled analogues but also glyoxylate, lactate, oxalate and tartrate. Moreover, the preliminary results of a sample preparation approach to remove phosphate salts are presented. Phosphate is a very common salt that is often used in biological buffers but prevents the derivatization of carboxylic acid for GC/MS analysis and reduces the reproducibility of results for both GC/MS and CE/MS analysis. Egg yolk immunoglobulins (IgY) are considered as a promising tool of establishing passive immunity against enteric pathogens (e.g. Salmonella) in poultry (Kovacs-Nolan and Mine, 2005). In our previous works, in-feed incorporation of freeze-dried yolk powder (FYP) improved performances of Salmonella-infected broilers (Marcq et al., 2009). Nevertheless, no extra value could be attributed to the presence of specific IgY when a comparison was done between impact of hyperimmune or non immune FYP both concerning growth performances and Salmonella infection. We hypothesized that these inconclusive in vivo results were related to the sensitivity of IgY to gastrointestinal conditions. Egg yolks with high IgY titers directed against Salmonella Enteritidis and Typhimurium were obtained from laying hens immunized as previously described (Chalghoumi et al., 2008). Three feed additives derived from these eggs were produced: FYP, spray-dried yolk powder (SYP), and freeze-dried water-soluble fraction of yolk powder (WSFP). A force-feeding assay was realized on these additives with 48 five-week-old males Ross broiler chicken (Salmonella spp.-free status). Survival of undigested IgY and bioactive Fab fragments was evaluated in the intestinal tract by ELISA. Statistical analyses were done using GLM procedure of Minitab 15.1. Software. Differences were considered significant when P<0.05. As a result of the digestive process, the levels of total and specific undigested IgY found throughout the intestine were dramatically reduced for the three additives. For FYP, no difference was observed with intestinal contents of control animals (P>0.05) and for WSFP, which correspond to semi-purified IgY, the immunological activity reached the same level than that for SYP (P>0.05) whereas its initial level was almost ten times higher. The results suggest a protective effect of whole yolk on IgY with SYP but such effect was not observed with FYP. High levels of Fab fragments were identified in intestinal contents, highlighting the importance of protein digestion in the digestive tract and especially pepsin digestion. These Fab fragments conserve their immunological functionality and remained detectable in all intestinal segments including the caecum which represents the major site of Salmonella infection in poultry. These results can explain our previous challenge data and reveal that a protection treatment should be applied to dried egg yolk to improve the resistance of IgY to gastrointestinal conditions in broilers. The potential of kenaf (Hibiscus cannabinusL.) and corn (Zea mays L.) for accumulation of cadmium and zinc was investigated. Plants have been grown in lysimetres containing dredging sludge, a substratum naturally rich in trace metals. Biomass production was determined. Sludge and water percolating from lysimeters were analyzed by atomic absorption spectrometry.No visible symptoms of toxicity were observed during the three- month culture. Kenaf and corn tolerate trace metals content in sludge. Results showed that Zn and Cd were found in corn and kenaf shoots at different levels,2.49 mg/kg of Cd and 82.5 mg/kg of Zn in kenaf shoots and2.1mg/kgofCdand10.19mg/kgincornshoots. Quantities of extracted trace metals showed that decontamination of Zn and Cd polluted substrates is possible by corn and kenaf crops. Tolerance and bioaccumulation factors indicated that both species could be used in phytoremediation. [less ▲]

Protecting and supporting local production systems, regional authorities, as well as producers, give a very important role to milk quality. Therefore, this study was aimed to investigate the potential of mid-infrared spectroscopy (MIR) for certifying the geographic origin of milk. Because milk MIR spectral databases and extra phenotypes (breed, testday, livestock herd and origin appellation of traditional products) were available in the Belgium Walloon Region via European project OptiMIR (INTERREG IVB North West Europe Program), discrimination studies were conducted to distinguish Ardennes region (which is linked to PDO “Beurre d’Ardennes”) from the rest of Wallonia. A total of 542,733 spectral records linked to their geographic origin coming from Wallonia milk recording were used (97,369 of MIR spectra -Ardennes region and 450,326 -rest of Wallonia). A mixed model (fixed: breed, year and month of record, random: herd x year) was applied to obtain predicted MIR spectral values for all testdays and prediction errors (residuals) representing the factors not present in the model. In order to test the MIR ability to milk authentication, chemometric tools, such as partial least squat regression and linear discriminant analysis were applied to residuals for three MIR spectral regions (e.g. 930-1600 cm-1, 1710-1810 cm-1 and 2560-2990 cm-1). The classifications on not-corrected MIR spectral data were 95% and the cross-validation were 95% for Ardennes region. Results showed after correction of MIR spectra, the discriminant function constructed on the residuals spectra allowed a good discrimination. The results show that MIR spectroscopy techniques may provide useful fingerprints to detect geographic origin and could be potentially used in routine management decision and quality assurance tools. The use of a rotating-baseline nulling interferometer for exoplanet detection was proposed several decades ago, but the technique has not yet been fully demonstrated in practice. Here we consider the faint companion and exozodiacal disk detection capabilities of rotating-baseline nulling interferometers, such as are envisioned for space-based infrared nullers, but operating instead within the aperture of large single telescopes. In particular, a nulling interferometer on a large aperture corrected by a next-generation extreme adaptive optics system can provide deep interferometric contrasts, and also reach smaller angles (sub λ/D) than classical coronagraphs. Such rotating nullers also provide validation for an eventual space-based rotating-baseline nulling interferometer. As practical examples, we describe ongoing experiments with rotating nullers at Palomar and Keck, and consider briefly the case of the Thirty Meter Telescope. [less ▲]

The Darwin and TPF-I space missions will be able to study the atmosphere of distant worlds similar to the Earth. Flying these space-based interferometers will however be an extraordinary technological challenge and a first step could be taken by a smaller mission. Several proposals have already been made in this context, using the simplest nulling scheme composed of two collectors, i.e., the original Bracewell interferometer. Two of these projects, viz. Pegase and the Fourier-Kelvin Space Interferometer, show very good perspectives for the characterisation of hot extra-solar giant planets (i.e., Jupiter-size planets orbiting close to their parent star). In this paper, we build on these concepts and try to optimise a Bracewell interferometer for the detection of Earth-like planets. The major challenge is to efficiently subtract the emission of the exo-zodiacal cloud which cannot be suppressed by classical phase chopping techniques as in the case of multi-telescopes nulling interferometers. We investigate the potential performance of split-pupil configurations with phase chopping and of OPD modulation techniques, which are good candidates for such a mitigation. Finally, we give a general overview of the performance to be expected from space-based Bracewell interferometers for the detection of extra-solar planets. In particular, the prospects for known extra-solar planets are presented. [less ▲]

The aim of this study was to demonstrate the ability of a recently developed beta(+)-range sensitive intracerebral probe (beta-Microprobe) to measure the binding kinetics of [(18)F]MPPF, a well-documented 5-HT(1A) serotoninergic receptor ligand, in the dorsal raphe nucleus (DRN) of the anaesthetised rat. This midbrain nucleus presents a high concentration of 5-HT(1A) receptors known to be implicated in the effects of antidepressants. The difficulty confronting this study lay in the fact that the dimensions of the DRN are smaller than the detection volume of the beta-Microprobe. In the first part of the study, we studied the feasibility of this measurement from a theoretical point of view by autoradiography and a Monte Carlo simulation. We determined the optimal beta-Microprobe location close to the DRN and verified that this configuration allowed accurate determination of [(18)F]MPPF specific binding in the nucleus. In the second part of our study, we measured the in vivo time-concentration curves of [(18)F]MPPF binding in the DRN in comparison with the cerebellum. The specificity of [(18)F]MPPF binding in the DRN was confirmed by its displacement after non-labelled 5-HT(1A)antagonist injection (MPPF or WAY-100635). Moreover, we verified the feasibility of using beta-Microprobe monitoring and simultaneous validation by microdialysis to study the effect of an increase in extracellular serotonin, induced by fenfluramine injection, on [(18)F]MPPF binding in the DRN. Our theoretical simulations, confirmed by our experimental results, demonstrate the ability of this new device to monitor in vivo the binding of [(18)F]MPPF in the DRN of anaesthetised rodents. Most of agricultural biogas plants do not operate at optimum loading rate to avoid overload and process failure. This study aims at the development of an electronic nose technology to serve as a specific monitoring tool for anaerobic digestion processes. Twelve semi-continuous anaerobic mini-digesters (1.6 L useful capacity) were fed with three different substrates: sucrose, maize oil and a mix 1:1 of sucrose and maize oil. Two feeding strategies were followed (i) a cautious organic load (1.3 gVS.L-1.day-1) and (ii) an increasing load strategy (1.3 to 5.33 gVS.L-1.day-1). Digesters were monitored during two months by analysing the biogas composition and collecting electronic nose measurements. The electronic nose was able to detect the impact of organic load variations. Overload situations were distinguished by the e-nose apparatus and the different feeding substrates induced distinct responses from the e-nose system. [less ▲]
