The International Liquid Mirror Telescope (ILMT) consists of a 4-m diameter zenith telescope with a liquid mirror characterized by a f/2 focal ratio. It is a collaborative project between Belgium, Canada, India and Poland. The participating institutions are: Institute of Astrophysics and Geophysics of the Liege University (IAGL), the Royal Observatory of Belgium (ROB), six Canadian universities (British Columbia, Laval, Montreal, Toronto, Victoria and York), the Aryabatta Research Institute of Observational Sciences (ARIES, Nainital, India) and the Observatory of Poznan (UAM, Poland). This telescope is being installed at the Devasthal observatory, India (Longitude 79° 41' 04'' E, +29° 21' 40'' N, altitude 2450m), in the central Himalayas. First light should happen in early 2016. It will image every night a strip of sky having an approximate width of 27' in declination using a 4096 x 4096 pixel CCD detector that will work in the time delay integration (TDI) mode. The ILMT will thus perform a deep survey of a long and narrow strip of sky by looking at stars, galaxies, AGN, quasars, asteroids, space debris, ... crossing its field of view. It will observe in the i', r', g' SDSS spectral bands. This survey will thus be mainly dedicated to photometric and astrometric variability studies of faint objects. Among the main scientific goals, let us mention the detection and follow up of a few tens of multiply imaged quasars produced by gravitational lensing, hundreds of supernovae every year, detection of nearby brown dwarfs, white dwarfs and other faint stars based upon their trigonometric parallax and/or proper motion measurements, photometric variability studies of thousands of quasars and millions of stars as well as space debris detection at dusk and/or at dawn. Right now, numerous data sets have already been obtained of the ILMT strip of sky with smaller telescopes in the TDI mode. These will be used to carry out preliminary scientific projects and calibration of the ILMT data. A pipeline and database are presently in construction at the Poznan Observatory. Some videos and recent photographs will illustrate the present status of the ILMT project. [less ▲]

A new spin trap, the 5-(diethoxyphosphorylmethyl)-5-methyl-4,5-dihydro-3H-pyrrole N-oxide (3), is prepared through a four-step synthetic route. The in vitro spin trapping properties of 3 have been investigated and the hydroxyl and superoxide spin adducts characterised. The persistence of the superoxide spin adduct obtained with 3 is compared with those of the analogues obtained with DEPMPO [5-(diethoxyphosphoryl)-5-methyl-4,5-dihydro-3H-pyrrole N-oxide] and DMPO in both phosphate buffer and pyridine. In 0.1 M phosphate buffer at pH 7.0, the HOO-3? adduct is found to be 2.8 times more persistent than HOO-DMPO?, but 5.7 times less than HOO-DEPMPO?. In phosphate buffer, HOO-3? decomposes to give HO-3?. The stereoselectivity of the addition of free radicals on 3 is generally poor, and mixtures of diastereomers have been observed by EPR. The overall spin trapping behaviour of nitrone 3 is more similar to that of DMPO than that of DEPMPO. [less ▲]

Glioblastoma constitute the most frequent and deadliest brain tumors of astrocytic origin. They are resistant to all current therapies and are associated with a high rate of recurrence. Glioblastoma were previously shown to respond to treatments by 5-aminolevulinic acid (5-ALA)-based photodynamic therapy (PDT) mainly by activating a necrotic type of cell death. The receptor-interacting protein 3 (RIP3) has recently been outlined as a key mediator of this caspase-independent form of programmed cell death. In the present study, we analyzed the necrotic mechanism induced by 5-ALA-PDT in human glioblastoma cells and explored the role of RIP3 in this context. Our results show that PDT-induced necrosis is dependent on RIP3, which forms aggregates and colocalizes with RIP1 following photosensitization. We demonstrate that PDT-mediated singlet oxygen production is the cause of RIP3-dependent necrotic pathway activation. We also prove that PDT induces the formation of a pro-necrotic complex containing RIP3 and RIP1 but lacking caspase-8 and FADD, two proteins usually part of the necrosome when TNF-α is used as a stimulus. Thus, we hypothesize that PDT might lead to the formation of a different necrosome whose components, besides RIP1 and RIP3, are still unknown. In most cases, glioblastoma are characterized by a constitutive activation of NF-κB. This factor is a key regulator of various processes, such as inflammation, immune response, cell growth or apoptosis. Its inhibition was shown to further sensitize glioblastoma cells to PDT-induced necrosis, however, no difference in RIP3 upshift or aggregation could be observed when NF-κB was inhibited. [less ▲]

BACKGROUND: Natural T regulatory cells (Treg) are challenging to expand ex vivo, and this has severely hindered in vivo evaluation of their therapeutic potential. 5-Azacytidine (5-azaC) and Rapamycin (RAPA) are immunosuppressive drugs that promote selectively the expansion of CD4+CD25highFoxp3+ regulatory T cells OBJECTIVE: We investigated whether 5-azaC and RAPA could be used together to promote the ex vivo expansion of Tregs purified from adult human peripheral blood. METHODS: CD4+CD25+ and CD4+CD25- T cells were isolated from PBMC of normal controls using Miltenyi beads and FACS ARIA sorting. These cells subsets were cultured in the presence of anti-CD3/CD28 antibodies and 200 IU/ml of IL2 for 2 weeks. RAPA (100nM) and 5-azaC (1µM) were added to half of the cultures. We monitored cell expansion and after harvest, the cell phenotype, gene expression, T suppression activity and Annexin V binding were determined and compared to values obtained with control cell culture and with freshly-separated CD4+CD25+ and CD4+CD25- T cells. Also we tested the absolute number of Treg in the several culture conditions during the time of expansion. RESULTS: We found that 5-azaC helped maintain FOXP3 expression during the expansion process probably by promoting the conversion of T conventional (Tconv) in Treg, instead Rapa induces selectively apoptosis in Tconv cells and expansion in Treg. The addition of 5-azaC to RAPA treated cultures improved gene expression of FOXP3, CD25, STAT5 and TGF-B resulted in enhanced Treg expansion and suppressive activity. Also Rapa and 5-AzaC combination sustain Bcl-2 protein expression in Treg conferring resistance to apoptosis process. CONCLUSION: 5-azaC may have utility in ex vivo expansion of human Tregs, not as a single agent, but in combination with RAPA. These data may considerably accelerate the development of immunotherapeutic approaches for the treatment of autoimmune disease or posttransplant alloreactions by the adoptive transfer of nTreg cells.
