We report the identification of a new 17-bp-long cis sequence located between positions -506 and -489 from the transcription start site. This sequence is recognized by a trans-activating factor that we tentatively named HER2 transcription factor (HTF). This factor, involved in the increased transcription of the HER2 gene in the BT-474 mammary tumor cells, has a molecular weight of about Mr 50,000. HTF can also bind, but with a lower affinity, to a related cis sequence present in the epidermal growth factor receptor promoter. Interestingly, the HTF binding activity is high in nuclear extracts from several mammary tumor cells overexpressing the HER2 gene. Because this method is time-consuming, it is not typically performed in routine analysis of nerve biopsies. We applied the Bodian-Luxol technique to detect unmyelinated axons by light microscopy on semithin sections from resin-embedded nerve tissue. Estimates were compared to ultrastructural counts. The staining appeared highly specific for axons. Excellent correlation was found between optic densities and the population of UF larger than 0.5 microm. The smallest profiles detected by light microscopy had a diameter close to 0.6 microm. This new technique is not a substitute for ultrastructural quantitative morphometry of UF, as very small unmyelinated axons, especially regenerating ones, can not be reliably visualized. However, it provides a valuable light microscopic method for evaluating axonal loss among UF. [more ▼] Oral daily and weekly bisphosphonates represent the current mainstay of treatment for postmenopausal osteoporosis (PMO). However, the inconvenience of frequent dosing is known to negatively affect adherence to therapy in the long term. This has prompted the development of convenient oral bisphosphonate regimens that feature simple, less frequent dosing schedules. Such regimens require high potency agents, which can be given at low effective doses and that also have good tolerability. Ibandronate is a potent, nitrogen-containing bisphosphonate with proven efficacy when given intermittently to estrogen-depleted beagle dogs, rats and cynomolgus monkeys. Clinically, a pivotal prospective study has established that oral ibandronate has significant vertebral fracture efficacy in PMO, whether given daily (2.5 mg) or intermittently (20 mg every other day for 12 doses every 3 months; extended between-dose interval > 2 months). Both oral regimens were well tolerated, which is noteworthy as patients with a history of gastrointestinal (GI) disturbance were not specifically excluded. As a result of these findings, a large, multinational, randomized, double-blind study (Monthly Oral iBandronate In LadiEs: MOBILE) is currently exploring the non-inferiority of once-monthly oral ibandronate (100 or 150 mg) to the oral daily ibandronate (2.5 mg) regimen with proven anti-fracture efficacy, in terms of lumbar spine bone mineral density (BMD) change. As with the trials investigating the weekly administration of other bisphosphonates, vertebral fracture efficacy will be inferred if the study demonstrates the non-inferiority of once-monthly ibandronate to the proven oral daily regimen in terms of spinal BMD change. The availability of this once-monthly ibandronate regimen is expected to offer benefits in terms of convenience (by having to follow dosing recommendations once a month vs. once daily or weekly) and potentially tolerability (by reducing the potential for upper GI irritation that can result from frequent, repeated exposure). Greater convenience and tolerability may enhance the therapy adherence and, hence, improve long-term therapeutic outcomes in PMO. [more ▼] We report on high contrast near-infrared (~2.2 μm) observations of Vega obtained with the Palomar Fiber Nuller, a dual sub-aperture rotating coronagraph installed at the Palomar Hale telescope. The data show consistent astrophysical null depth measurements at the ~= 10[SUP]–3[/SUP] level or below for three different baseline orientations spanning 60 deg in azimuth, with individual 1σ uncertainties <=7 × 10[SUP]–4[/SUP]. These high cancellation and accuracy levels translate into a dynamic range greater than 1000:1 inside the diffraction limit of the 5 m telescope beam. Such high contrast performance is unprecedented in the near-infrared and provides improved constraints on Vega's immediate (sime20 to 250 mas, or sime0.15 to 2 AU) environment. In particular, our measurements rule out any potential companion in the [0.25-1 AU] region contributing more than 1% of the overall near-infrared stellar flux, with limits as low as 0.2% near 0.6 AU. These are the best upper limits established so far by direct detection for a companion to Vega in this inner region. We also conclude that any dust population contributing a significant (>=1%) near-infrared thermal excess can arise only within 0.2 AU of the star, and that it must consist of much smaller grains than in the solar zodiacal cloud. Dust emission from farther than sime2 AU is also not ruled out by our observations, but would have to originate in strong scattering, pointing again to very small grains. The first one corresponds to the usual colour-singlet mechanism. The second one has not been considered so far. We treat it in a gauge-invariant manner, and introduce new 4-point vertices, suggestive of the colour-octet mechanism. These new objects enable us to go beyond the static approximation. We show that the contribution of the new cut can be as large as the usual colour-singlet mechanism at high P-T for J/psi.
