Abdominal obesity plays a key role in the development of metabolic syndrome. Similarities between metabolic syndrome and Cushing disease suggest that excessive local tissue exposition to glucocorticoids, despite normal circulating plasma levels, might contribute to the pathophysiology of metabolic syndrome. To this respect, 11β-hydroxysteroid dehydrogenase type 1 (11HSD1), the enzyme which converts cortisone (inactive) to cortisol (active) in target tissues, raises much interest. Indeed, several studies showed both increased expression and activity of this enzyme in adipose tissues in presence of obesity. Even more striking, experimental data in rodents showed a direct link between increased 11HSD1 activity and the development of metabolic abnormalities. Furthermore, studies in mice KO for 11HSD1 confirmed the potential of inhibiting this enzyme to attenuate metabolic abnormalities related to visceral adiposity. Selective inhibitors of 11HSD1 are currently in development, and preliminary results obtained in rodents appear promising, with significant improvements in glucose and lipid profiles. The development of potent and selective 11HSD1 inhibitors may open new prospects in the treatment of metabolic syndrome or type 2 diabetes associated with abdominal obesity in humans. [less ▲]

La 11β-hydroxystéroïde déshydrogénase de type 1 (11HSD1), qui convertit la cortisone (inactive) en cortisol (actif) dans les tissus cibles, est surexprimée dans certains tissus en présence d’une obésité, surtout abdominale, et pourrait jouer un rôle dans les anomalies métaboliques associées. Des inhibiteurs non sélectifs de la 11HSD1 ont déjà été testés chez l’homme et se sont révélés peu efficaces. Le développement de nouveaux inhibiteurs, plus puissants et plus sélectifs, est en cours. Des résultats préliminaires obtenus avec divers inhibiteurs sont prometteurs, avec une diminution de l’insulinorésistance, une amélioration de la tolérance au glucose ou du contrôle glycémique et une correction de certaines anomalies liées au syndrome métabolique. Si ces résultats se confirment, les inhibiteurs sélectifs de la 11HSD1 pourraient représenter à l’avenir une nouvelle classe pharmacologique d’antidiabétiques oraux. Cet article donne au lecteur un aperçu des différentes étapes de réflexion et d’expérimentations humaines qui ont mené au développement d’inhibiteurs de la 11HSD1 à visée thérapeutique. [less ▲]

From 1992 to 2004 upstream migrating eels were collected in a trap (0,5 cm mesh size) installed at the top of a small pool-type fish-pass at the Visé-Lixhe dam (built in 1980 for navigation purposes and hydropower generation; height : 8,2 m; not equipped with a ship-lock) on the international River Meuse near the Dutch -Belgium border (290 km from the North Sea; width: 200 m; mean annual discharge: 238 m3/s; summer water temperature 21-26°C).The trap in the fish-pass was checked continuously (three times a week) over the migration period from March to September each year, except in 1994. We caught a total number of 32157 eels (biomass 1,955 kg) with a size from 14 cm to 85 cm and a mean value of 31,6 cm corresponding to yellow eels. The study based on a constant year-to-year sampling effort revealed a regular decrease of the annual catch from a maximun of 5613 fish in 1992 to a minimum of 423 in 2004. This demographic trend is fitted by the equation: number per year = 5.614 - 299 t, where t is time in years with 1992 as year 1. According to this model, the upstream migrating yellow eelstock in the Belgian Meuse should drop to near zero within the next ten years, as an expression of a collapsing recruitment of glass eels in the estuary in the Netherlands. In the discussion of these results, we examine the possible role of two other factors on the decrease over time of the number of yellow eels caught in the fish-pass. i) the effect of environmental variables (river discharge, water temperature and dissolved oxygen content) on the timing and intensity of upstream migration waves and ii) the existence and use of alternative migration routes. The presence of corticotropin-releasing hormone (CRH) receptors has been previously demonstrated in corticotrophs from normal pituitaries using a method combining immunocytochemistry and liquid emulsion autoradiography. The aim of this study was to compare the characteristics of the 125I-Tyr0-hCRH binding in corticotrophs from normal pituitaries (three obtained at autopsy and one obtained at surgery) with corticotrophs from pituitary adenomas (six corticotroph adenomas responsible for Cushing's disease and two silent corticotroph adenomas secreting a biologically inactive ACTH molecule). In normal corticotrophs, the larger part of the 125I-Tyr0-hCRH binding was localised in patchy conglomerates at the centre of the cell and, to a much lesser degree, in a diffuse pattern at the cell periphery. In adenomatous corticotrophs, CRH receptor expression is disturbed both quantitatively and qualitatively. Except for a minority of cells in one adenoma, all adenomatous corticotrophs showed only peripherally bound 125I-Tyr0-hCRH and no centrally localised binding. Furthermore, adenomatous corticotrophs revealed a statistically significant lower signal intensity when compared to normal corticotrophs and a strongly negative correlation was found between the labelling area in adenomatous corticotrophs and both the basal and CRH-stimulated plasma ACTH levels. These findings suggest defective processing of CRH receptors and could be relevant to the sustained ACTH secretion by adenomatous corticotrophs in Cushing's disease and, more generally, provide an explanation to its pathology. The silent corticotrophs secreting a biologically inactive ACTH molecule were characterised by a very faint signal intensity, although present on almost every cell. [less ▲]

Context. Measuring the carbon isotope abundance ratio in comets allows one to constrain the conditions in the outer protosolar nebula. Different measurements of the [SUP]12[/SUP]C/[SUP]13[/SUP]C ratio, using various molecules, have already been published for different solar system objects, such as the Sun, the Earth, the Moon, asteroids, planets, or comets. So far, all these measurements are consistent with [SUP]12[/SUP]C/[SUP]13[/SUP]C ~ 90, but significant differences have been observed. This ratio is remarkably constant in comets (91.0 ± 3.6) for studies based on the CN radical, but it presents stronger variations in studies based on other radicals. <BR /> Aims: This paper aims at measuring the [SUP]12[/SUP]C/[SUP]13[/SUP]C ratio in two bright Oort cloud comets using the [SUP]12[/SUP]C_2 and [SUP]12[/SUP]C[SUP]13[/SUP]C emission lines and an improved method. The ratios will be compared to those obtained for the same comets with another radical, CN. <BR /> Methods: We used the (2,1) and (1,0) bandheads of the [SUP]12[/SUP]C[SUP]13[/SUP]C, near 4723 and 4745 Å to measure the [SUP]12[/SUP]C/[SUP]13[/SUP]C ratio and compared their intensity to the [SUP]12[/SUP]C_2 lines of the same bands. We developed a model for interpreting observational data obtained at high resolution (~70 000) using the 8.2-m Kueyen telescope (UT2) of the Very Large Telescope (VLT) with the Ultraviolet and Visual Echelle Spectrograph (UVES) in two comets: C/2001 Q4 (NEAT) and C/2002 T7 (LINEAR). <BR /> Results: Our modeling has provided [SUP]12[/SUP]C/[SUP]13[/SUP]C = 85 ± 20 for C/2002 T7 (LINEAR) and 80 ± 20 for C/2001 Q4 (NEAT). These values are compatible with previous measurements performed with the CN radical. Based on observations made with ESO Telescopes at the Paranal Observatory under programmes ID 073.C-0525. The measurement of the carbon isotope abundances ratio in comets allows to constrain the conditions in the outer protosolar nebula. Different measurements of the 12C/13C ratio, using various molecules, have already been published for different solar system objects, such as the Sun, the Earth, the Moon, asteroids, planets or comets. So far all these measurements are consistent with 12C/13C' 90 but some significant differences have been seen. In comets this ratio is remarquably constant and equal to 91.0±3.6 [3] for studies based on CN radical but present larger variations with studies based on other radicals [1][5]. In this work we present a new analysis of this ratio, based on two different approachs for modeling the 12C2 and 12C13C emission spectrum and the (1,0) and (2,1) bandheads of 12C13C. Observational data obtained at high resolution (' 70, 000) using the 8.2-m Kueyen telescope (UT2) of the Very Large Telescope (VLT) with the Ultraviolet and Visual Echelle Spectrograph (UVES) instrument have been used to test our modeling and measure the 12C/13C ratio in two different comets: C/2001 Q4(NEAT) and C/2002 T7 (LINEAR). Our modeling and numerical results will be presented. [less ▲]

L’alcool remonte le moral. Une petite cuite n’a jamais tué personne. Boire un café atténue l’effet de l’alcool. Le binge drinking est un fléau nouveau… De nombreuses idées reçues, certaines fondées, d’autres pas, sont véhiculées à propos de l’alcool et de ses conséquences. L’alcool soulève aussi de multiples questions : L’alcool est-il une drogue ? L’alcool est-il aphrodisiaque ? L’alcoolisme est-il héréditaire ? Combien l’alcool coûte/rapporte-t-il à la société ? Peut-on guérir de l’alcoolisme ?… Ce livre a pour but de démont(r)er certaines idées reçues sur l’alcool et d’apporter des réponses aux questions que chacun se pose. Les auteurs ne se bornent pas à répondre par vrai ou faux, ils fournissent les explications, appuyées sur l’état des connaissances scientifiques actuelles, qui permettent d’infirmer ou de confirmer ces idées reçues ou de répondre à ces questions. Ils nuancent le propos lorsque la réponse n’est pas de l’ordre du tout ou rien. Il est indéniable que l’excès d’alcool est nuisible à la santé. Il existe cependant une littérature scientifique démontrant des effets positifs sur la santé de la consommation en quantités modérées de certaines boissons alcoolisées. Ce mélange d’effets positifs et négatifs explique que le public a développé une relation d’amour-haine avec l’alcool. Ainsi, les abstinents complets sont parfois qualifiés de rabat-joie. Les alcooliques chroniques (5 à 10 % des occidentaux, selon les études épidémiologiques !) sont, quant à eux, souvent trop vite jugés. Ce qui est certain c’est que l’alcoolo-dépendance est source de beaucoup de souffrances pour la personne et son entourage. Ces 41 réponses à des questions sur l’alcool visent à donner des balises au lecteur, littérature scientifique à l’appui. [less ▲]
