Fractionated whole body X-irradiation (4 X 1.75 Gy at weekly intervals) induces a high percentage of thymic lymphomas in C57BL/Ka mice. These tumors develop after a long latency period during which the thymic lymphopoiesis is deeply altered. In the present work, we test wether those modifications are due to lack of prothymocyte homing to preleukemic thymuses. Our results show that the preleukemic state of the thymus don't prevent the homing of normal marrow precursors grafted immediately after an irradiation of 4 Gy. Thus the alterations of thymic lymphopoiesis observed after a leukemogenic irradiation are not due to a modification in the thymus receptivity to thymocyte precursors. [less ▲]

The ploidy of the thymus was studied in C57B1/Ka mice irradiated with 4 weekly X-Ray doses of 1.75 Gy. The determination of nuclear DNA content was performed by flow cytometry of intact thymocytes labeled with propidium iodide in presence of a mixture of chicken and rainbow trout red blood cells as internal reference standards. The method has been tested by detecting the sex difference in DNA content of G0/G1 of normal thymic mouse cells. The mean value was 2.9% higher in female mice. The thymus of almost 60% of irradiated male mice present a slight hypoploidy of 2.6% one month after the last irradiation. [less ▲]

To study the role of follicular dendritic cells (FDC) in germinal centers, we tried to keep them alive in vitro by culturing entire lymphoid follicles. Ultrastructural studies of those cultures in different conditions of culture technique, medium and temperature have been made. In any considered conditions living FDC were not found in the cultures after the 7th day. But during that period, only lymphocytes in close contact with the cytoplasmic processes of FDC survived. FDC seem thus to exert a positive action on lymphocyte survival in vitro. Moreover, FDC and lymphocytes appeared associated in situ in clusters similar to those obtained after isolation of FDC (Lilet-Leclercq et al., J. Immunol. Meth., 1984, 59, 235). [less ▲]

Human pituitary adenomas were fixed in glutaraldehyde and embedded in epon. Ultrathin sections were incubated either with anti-hGH, anti-hPRL or anti-hLH. They were incubated with second step goat anti-rabbit immunoglobulins linked to gold particles. Two PRL secreting adenomas, one GH and PRL secreting adenoma, one ACTH secreting adenoma and two non secreting adenomas were studied. The specificity and the limits of the method were discussed in relation with the results obtained in light microscopy with the PAP method. [less ▲]

5 micrograms of human beta-endorphin were labelled with 2 mCi 125I by the chloramine T technique. After two gel filtrations on Sephadex G-15 and on Sephadex G-50 in phosphate buffer with EDTA, Trasylol and mercapto-ethanol, a pure tracer was obtained with a specific activity about 150 microCi/ug. Kept at + 4 degrees C, the tracer remained utilizable for 30 days without loss of immunoreactivity. The labelling with lactoperoxydase and the use of another gel filtration method (filtration on Aca 202) gave a 125I beta-END tracer with the same immunoreactivity. The binding of this tracer to the antibody of an anti-beta-END antiserum diluted at 1/8000 was 32% with a non specific binding of 2%. 5 micrograms of human beta-lipotropin were labelled with 0.5 mCi 125I by the lactoperoxydase method. After two gel filtrations on Sephadex G-25 and on Sephadex G-75 in phosphate buffer with EDTA, Trasylol and mercapto-ethanol, a pure tracer with a specific activity of 140 microCi/micrograms was obtained. It remained utilizable for 30 days when kept at + 4 degrees C. Gel filtration on Aca 202 did not give good purification, while gel filtration on Aca 54 was good but slower than on Sephadex G-75. The binding to antibody in absence of unlabelled beta-LPH was 32% for an anti-beta-LPH antiserum diluted at 1/4000. The non specific binding was 2.5%. [less ▲]

Serum activity of angiotensin converting enzyme (ACE) were measured during extra-corporeal circulation in five patients undergoing aorto-coronary bypass surgery. We observed a significant decrease of serum ACE levels in the absence of pulmonary circulation, suggesting that in man the lungs were the major source of circulating ACE. An effective extra-pulmonary liberation of ACE could take place during cardiopulmonary bypass. The levels of serum ACE increased with pulmonary recirculation, but preoperative levels were not reached 24 h later. [less ▲]

Cultivated animal cells (mouse peritoneal macrophages, chick fibroblasts or mouse Ehrlich tumor cells) or Zea mays root tips are treated with cis-Pt (II). Various effects (chromatin dispersion or condensation, pycnosis) are observed under some experimental conditions in all cell types. Cytoplasmic ribosomes in helical aggregates appear but only in vegetal cells. Mitosis and cell cycle disturbances due to cis-Pt (II) are probably related to chromatin alterations. We suggest that the latter and helical polyribosomes are produced by cis-Pt (II) reaction with nucleic acids in these structures. [less ▲]

This infusion reduced MBP (-7 mm Hg, p less than 0.05) after 13 days of protein-supplemented fasting. The graft into the footpad of Jackson mice of B16 melanoma cells which are only slightly pigmented, induces the simultaneous appearance of achromatic and well-pigmented metastatics. We have analysed histologically both types of metastases from the 8th to the 20th day after the resection of the primary tumor. The percentages of mitoses are higher in the achromatic metastases than in the pigmented ones. Both are infiltrated by lymphocytes and by a small number of macrophages and polymorphonuclears. A statistical analysis demonstrates however that the percentage of lymphocytes is not related to the importance of the metastase pigmentation. The discussion is devoted to the relations between the infiltration by lymphocytes, the immunogenicity and the differentiation of the tumor. [less ▲]

The effects of a single oral dose of 40 mg propranolol on endurance time and on the maximum oxygen consumption were studied in eight healthy young men. They were running on a motor-driven treadmill (work rate from 80 to 150% VO2 max. The VO2 max and endurance time were decreased to an average of 10 and 30% of controls respectively. The relationship between the endurance time and the relative work load (VO2/VO2 max) remained unchanged after beta-adrenoceptor blockade. The reduction of the endurance time following beta-adrenoceptor blockade is a consequence of the reduction of the VO2 max. [less ▲]
