The purpose of this study consisted in the identification of the macroscopic and microscopic characters of the vegetative and reproductive organs of Bryonia alba L., by the analysis of vegetal material, both integral and as powder. Optical microscopy was used to reveal the anatomical structure of the vegetative (root, stem, tendrils, leaves) and reproductive (ovary, male flower petals) organs. Histo-anatomical details were highlighted by coloration with an original combination of reagents for the double coloration of cellulose and lignin. Scanning electronic microscopy (SEM) and stereomicroscopy led to the elucidation of the structure of tector and secretory trichomes on the inferior epidermis of the leaf. Micrographic analysis of the powder obtained from aerial parts revealed segments of each organ (e.g. stomata, trichomes) and confirmed furthermore the results obtained by the histo-anatomical studies. Sections achieved through vegetative organs reveal typical anatomical structures: a transition to the secondary structure for the root, a typical dicotyledons’ structure with bicollateral vascular bundles for the stem, a bifacial structure of the leaf and stem-like structures for the tendrils and petioles, which prove they are metamorphosis of the stem. Anatomical structure of reproductive organs was performed hereby for the first time and revealed a typical anatomical structure for the 3-lodged ovary and a leaf-like structure of the male flower petal. Some of the results obtained confirm existing data from the scientific literature and additional information have been provided, outlining features that were not previously reported, such as SEM analysis of the leaf trichomes and histo-anatomical structure of the reproductive organs. Management of livestock grazing in highly-productive mountain meadows is an important aspect for the economic viability and the environmental impact of a grassland-based farm. The main aim of this study was to build near infrared models to determine the chemical composition and in vitro organic matter digestibility of Romanian meadow forages. The treatments were organic and mineral fertilizer combinations, and forage samples were obtained from three fertilization experiments conducted in the Apuseni Mountains; these samples were analysed using classical and NIR methods. The samples were scanned in the NIR wavelength band. The CRA-W Gembloux ‘local’ calibration models were validated with Romanian meadow forages and then used in order to predict the forage quality of samples. A second objective of the study was to determine the effects of fertilization on forage quality. The results showed a decrease in crude protein content from the NPK treatment (150:75:75), which can be explained by a reduction of Fabaceae plants with this treatment from 17.25% of the populations in the control (semi-natural meadow not fertilized) to 6.25% in the fertilized plots. The decrease in protein content and in vitro organic matter digestibility was related to a reduced Fabaceae presence. Our recommendation is to use mineral fertilization with NPK doses less than 100:50:50 to improve meadow productivity; meanwhile organic fertilization can also be used to complement and maintain biodiversity and forage quality. Among the food crops, yam takes up quantitatively the first place in the gabonese diet. Unfortunately, it can stay available only 6 to 7 months in the year because of difficulties of harvest and post- harvest. This problem is little studied in the case of Dioscorea cayenensis-D. rotundata complex. In order to optimize the use of micro tubers for the growing in green house or field, it is important to control the duration of storage before the germination. The present study concerns microtubers obtained by in vitro culture. When microtubers were harvested (after 9 months of culture) and directly transferred on a new medium without hormone, the tubers rapidly sprouted in in vitro conditions. Harvested microtubers were also stored dry in jars in sterile conditions during 2 to 18 weeks before in vitro sprouting. In this case, microtubers stored during 18 weeks sprouted more rapidly than those stored 8 weeks. The size of the tubers used for the storage had great influence on further sprouting. The upper microtubers in 25 mm can be kept to the darkness, under 50% of relative humidity, in 25°C during at least 18 weeks. Sprouting is 100% whatever the substrate of culture. The plant tissue culture technique constitutes a serious alternative for the preservation of plant kinds and for the production of planting material. These techniques allow multiplying in a short time of thousands of copies of new varieties of newly created plants. These in vitro plants can be used on one hand, for the production planting material, and on the other hand for ex vitro storage of breeding grounds with decelerated growth, to struggle against genetic erosion. These results should allow improving in practice the multiplication of yam, while guaranteeing phytosanitary qualities. [less ▲]
