For more than a decade, the taxonomy of Phormidiaceae has been problematic, since morphologically similar organisms represent phylogenetically distinct entities. Based on 16S rRNA analyses, the polyphyletic genus Phormidium and other gas-vacuolated oscillatorioids appear scattered throughout the cyanobacterial tree of life. Recently, several studies have focused on understanding the oscillatorioid taxa at the generic level. At the specific level, there are few studies characterizing cyanobacterial strains using combined datasets (morphology, ultrastructure, and molecular multi-locus analyses). Using a multi-facet approach, we propose a new, well-defined genus, Cephalothrix gen. nov., by analyzing seven filamentous strains morphologically 'intermediate' between gas-vacuolated taxa and Phormidium. Furthermore, we characterize two new species: Cephalothrix komarekiana (CCIBt 3277, CCIBt 3279, CCIBt 3523, CCALA 155, SAG 75.79, and UTEX 1580) and Cephalothrix lacustris (CCIBt 3261). The generic name and specific epithets were proposed under the provisions of the International Code of Nomenclature for algae, fungi, and plants. [less ▲]

An intracellular symbiotic bacterium was isolated from the flora of a natural clone of the black bean aphid Aphis fabae. The strain was able to grow freely in aerobic condition on a rich medium containing 1% of each of following substrates; dextrose, yeast extract and peptone of casein. Pure culture was achieved through the use of solid-phase culture on the same medium and was designated CWBI-2.3T. 16S ribosomal DNA sequence analysis revealed that CWBI-2.3T strain was a member of the gamma-3 subgroup of the class Proteobacteria, having high sequence identity (>99%) with the R-Type of secondary endosymbiont found in several aphid species Serratia symbiotica. As strain CWBI-2.3T (=LMG 25624T =DSM 23270T) was the first R-type symbiont to be isolated and characterized, we proposed to designate it as the type strain of Serratia symbiotica species. [less ▲]

A thermotolerant acetic acid bacterium, designated strain CWBI-B418(T), isolated in Senegal from mango fruit (Mangifera indica), was characterized in detail by means of genotypic and phenotypic methods. The novel strain was strictly aerobic and exhibited optimal growth on YGM medium at 35 degrees C. Cells were Gram-negative, motile and coccoid. The strain was assigned to the genus Acetobacter on the basis of 16S rRNA gene sequence analysis. DNA-DNA hybridization experiments with its phylogenetically closest relatives showed that strain CWBI-B418(T) represented a novel Acetobactergenospecies. The DNA G +C content of strain CWBI-B418(T) was 56.0 mol%. Phenotypic characteristics enabling the differentiation of strain CWBI-B418(T) from phylogenetically related Acetobacter species were: production of 2-keto-D-gluconic acid from D-glucose, but not 5-keto-D-gluconic acid, production of catalase but not oxidase, growth on yeast extract with 30% D-glucose, growth with ammonium as sole nitrogen source with ethanol as carbon source, utilization of glycerol and ethanol but not maltose or methanol as carbon sources, and growth in the presence of 10% ethanol. Based on the genotypic and phenotypic data presented, strain CWBI-B418(T) clearly represents a novel Acetobacter species, for which the name Acetobacter senegalensis sp. nov. is proposed. The type strain is CWBI-B418(T) (=LMG 23690(T) = DSM 18889(T)). [less ▲]
