In this paper, some algorithmic and computational advances are presented for power system dynamic simulations. The heart is a Schur-complement-based solution algorithm, stemming from domain decomposition methods, applied to the differential-algebraic equation model. This algorithm is then accelerated computationally, by employing parallel computing techniques, and numerically, by exploiting time-scale decomposition and localization. Models of a real medium-scale system and a realistic large-scale test system are used for the performance evaluation of the proposed methods. [less ▲]

Thiamine triphosphate (ThTP) was discovered over 60 years ago and it was long thought to be a specifically neuroactive compound. Its presence in most cell types, from bacteria to mammals, would suggest a more general role but this remains undefined. In contrast to thiamine diphosphate (ThDP), ThTP is not a coenzyme. In E. coli cells, ThTP is transiently produced in response to amino acid starvation, while in mammalian cells, it is constitutively produced at a low rate. Though it was long thought that ThTP was synthesized by a ThDP:ATP phosphotransferase, more recent studies indicate that it can be synthesized by two different enzymes: (1) adenylate kinase 1 in the cytosol and (2) FoF1-ATP synthase in brain mitochondria. Both mechanisms are conserved from bacteria to mammals. Thus ThTP synthesis does not seem to require a specific enzyme. In contrast, its hydrolysis is catalyzed, at least in mammalian tissues, by a very specific cytosolic thiamine triphosphatase (ThTPase), controlling the steady-state cellular concentration of ThTP. In some tissues where adenylate kinase activity is high and ThTPase is absent, ThTP accumulates, reaching ≥ 70% of total thiamine, with no obvious physiological consequences. In some animal tissues, ThTP was able to phosphorylate proteins, and activate a high-conductance anion channel in vitro. These observations raise the possibility that ThTP is part of a still uncharacterized cellular signaling pathway. On the other hand, its synthesis by a chemiosmotic mechanism in mitochondria and respiring bacteria might suggest a role in cellular energetics. [less ▲]

Among all classes of drugs, the type 5 phosphodiesterase inhibitors (PDE5-i) (sildenafil, vardenafil and tadalafil) are the most counterfeited and copied in industrialized countries. This is why it is very important for regulatory agencies to have a panel of analytical methods to analyse these drugs and to be able to detect counterfeit or substandard medicines. During this chapter, several analytical techniques will be described and discussed. Among spectroscopic techniques, Raman, NIR, FT-IR and chemical imaging techniques will be envisaged. Their ability to detect counterfeits and predict if a new sample is genuine or not will be presented. Other techniques such as XRay powder diffraction and NMR spectroscopies will be briefly presented. Chromatographic impurity fingerprints will be considered and described for the analysis of suspect samples of Cialis® and Viagra®. Once again, the possibility to predict the authenticity of a new sample by the mean of this technique is envisaged. Finally, a MS-compatible UHPLC-UV method for the analysis of the three authorised PDE5-i and some of their analogues will be shortly described. This technique has been validated using spiked placebo samples in a vegetal matrix representing a suspect sample with a complex matrix. As a conclusion, a generic strategy for the detection and quality evaluation of counterfeit drugs will be presented. [less ▲]

Industrial activities have deeply impacted the quality of biophysical environment at places. This review focuses on consequences of soil contaminations by metallic trace elements: what conditions the mobility of contaminants in soils and what are the remediation practices based upon physico-chemical stabilization? This technique is an in situ remediation which aims at limiting the dispersion of metals in the environment by use of soil conditioners and amendments in contaminated sites. First of all, we reviewed the concepts of mobility of trace elements and their indicators. Then, we compiled the different kinds of amendments applied to stabilize contaminants in soils and the processes involved. [less ▲]
