[more ▼] Protein quantification based upon mass spectrometry is gaining ground in diverse applications of biological and clinical relevance. The present article focuses on one of the most complex biological fluids - serum - and provides a novel ICPL based quantification protocol. The results are compared to a label-free (data independent alternate scanning) absolute quantification method. The validation is performed using MRM based protein quantification technique. Regarding the ICPL approach, serum samples used in this study were depleted of high abundant proteins, labeled with ICPL and fractionated according to their respective pI (3-5, 5-7 and 7-12). The samples were further subjected to tryptic digestion followed by treatment with the Glu-C enzyme. The peptides were analyzed on a 2D-nano-LC system using four different concentrations of salt injections (45, 75, 150 and 500 mM ammonium acetate). The LC system was connected on-line with the electrospray ion-trap mass spectrometer. For the label-free quantification the serum samples were depleted and digested with trypsin. A proteome-wide comparison was performed using highly reproducible LC and data independent alternate scanning in conjunction with a high mass accuracy orthogonal time-of-flight mass spectrometer. Selected proteins, found by both methods, were validated using the MRM approach. For this purpose non-depleted tryptically digested serum samples were analyzed by LC coupled with a triple-quadrupole MS. The relative protein quantification using ICPL and mass spectrometry allowed for the detection of approximately 200 proteins, whereas about 2/3 of those contained the ICPL label and could therefore be quantified. Label-free approach used no fractionation, less sample and was able to identify and quantify over 110 proteins. The identified proteins covered generally 3-4 orders of magnitude of protein concentration in human serum. Changes in relative abundance of eight proteins were validated using MRM. This study, for the first time, shows the ability of the relative protein quantification based upon ICPL and 2D-LC-MS/MS to quantify serum biomarkers. It provides two additional label-free approaches that could validate and bring additional value to the label-based results, offering a starting point for comprehensive proteomics studies aiming at revealing biomarkers of clinical relevance. Brain regions associated to recollection were evidenced by contrasting activations for inclusion and exclusion conditions whereas brain regions related to familiarity were explored with the mean effect of the two conditions (at P < .05 corrected). Twelve patients had null recollection estimates (AD-), whereas 16 patients did experience some recollection although significantly less than controls (AD+). In AD+ and controls, recollection activated the posterior cingulate cortex (PCC). In contrast, familiarity estimates were equivalent in the 3 groups and were associated with brain activations around the intraparietal sulcus (IPS). Thus, in AD, impaired recollection is related to damage of the PCC whereas preserved familiarity is supported by the IPS. We hypothesized that networks involved in attentional and executive processes, as well as networks involved in serial order processing, underlie STM for both verbal and visual list information, with neural specificity restricted to sensory areas involved in processing the specific items to be retained. Participants were presented sequences of nonwords or unfamiliar faces, and were instructed to maintain and recognize order or item information. For encoding and retrieval phases, null conjunction analysis revealed an identical fronto-parieto-cerebellar network comprising the left intraparietal sulcus, bilateral dorsolateral prefrontal cortex, and the bilateral cerebellum, irrespective of information type and modality. A network centered around the right intraparietal sulcus supported STM for order information, in both verbal and visual modalities. Modality-specific effects were observed in left superior temporal and mid-fusiform areas associated with phonological and orthographic processing during the verbal STM tasks, and in right hippocampal and fusiform face processing areas during the visual STM tasks, wherein these modality effects were most pronounced when storing item information. The present results suggest that STM emerges from the deployment of modality-independent attentional and serial ordering processes toward sensory networks underlying the processing and storage of modality-specific item information. [more ▼] BACKGROUND: Gemcitabine (2',2'-difluorodeoxycytidine) administration after resection of pancreatic cancer improves both disease-free survival (DFS) and overall survival (OS). Deoxycytidine kinase (dCK) mediates the rate-limiting catabolic step in the activation of gemcitabine. The authors of this report studied patient outcomes according to the expression of dCK after a postoperative gemcitabine-based chemoradiation regimen. METHODS: Forty-five patients with resected pancreatic adenocarcinoma received adjuvant gemcitabine based-therapy in the context of multicenter phase 2 studies. Their tumors were evaluated retrospectively for dCK protein expression by immunohistochemistry. A composite score based on the percentage of dCK-positive cancer cells and the intensity of staining was generated, and the results were dichotomized at the median values. RESULTS: The median follow-up was 19.95 months (95% confident interval [CI], 3.3-107.4 months). The lymph node (LN) ratio and dCK protein expression were significant predictors of DFS and OS in univariate analysis. On multivariate analysis, dCK protein expression was the only independent prognostic variable (DFS: hazard ratio [HR], 3.48; 95% CI, 1.66-7.31; P = .001; OS: HR, 3.2; 95% CI,1.44-7.13; P = .004). CONCLUSIONS: dCK protein expression was identified as an independent and strong prognostic factor in patients with resected pancreatic adenocarcinoma who received adjuvant gemcitabine therapy. The authors concluded that it deserves prospective evaluation as a predictive biomarker for patient selection.
