The Hedgehog (Hh) signaling pathway plays an important role in human development. Abnormal activation of this pathway has been observed in several types of human cancers, such as the upper gastro-intestinal tract cancers. However, activation of the Hh pathway in colorectal cancers is controversial. We analyzed the expression of the main key members of the Hh pathway in 7 colon cancer cell lines in order to discover whether the pathway is constitutively active in these cells. We estimated the expression of SHH, IHH, PTCH, SMO, GLI1, GLI2, GLI3, SUFU and HHIP genes by RT-PCR. Moreover, Hh ligand, Gli3 and Sufu protein levels were quantified by western blotting. None of the cell lines expressed the complete set of Hh pathway members. The ligands were absent from Colo320 and HCT116 cells, Smo from Colo205, HT29 and WiDr. GLI1 gene was not expressed in SW480 cells nor were GLI2/GLI3 in Colo205 or Caco-2 cells. Furthermore the repressive form of Gli3, characteristic of an inactive pathway, was detected in SW480 and Colo320 cells. Finally treatment of colon cancer cells with cyclopamine, a specific inhibitor of the Hh pathway, did not downregulate PTCH and GLI1 genes expression in the colorectal cells, whereas it did so in PANC1 control cells. Taken together, these results indicate that the aberrant activation of the Hh signaling pathway is not common in colorectal cancer cell lines. (c) 2007 Wiley-Liss, Inc. [less ▲]

One strategy employed by bacterial strains to resist beta-lactam antibiotics is the expression of metallo-beta-lactamases requiring Zn+2 for activity. In the last few years, many new zinc beta-lactamases have been described and several pathogens are now known to synthesize members of this class. Metallo-beta-lactamases are especially worrisome due to: (1) their broad activity profiles that encompass most beta-lactam antibiotics, including the carbapenems; (2) potential for horizontal transference; and (3) the absence of clinically useful inhibitors. on the basis of the known sequences, three different lineages, identified as subclasses B1, B2, and B3 have been characterized. The three-dimensional structure of at least one metallo-p-lactamase of each subclass has been solved. These very similar 3D structures are characterized by the presence of an alpha beta beta alpha-fold. In addition to metallo-beta-lactamases which cleave the amide bond of the beta-lactam ring, the metallo-beta-lactamase superfamily includes enzymes which hydrolyze thiol-ester, phosphodiester and sulfuric ester bonds as well as oxydoreductases. Most of the 6000 members of this superfamily share five conserved motifs, the most characteristic being the His116-X-His118-X-Asp120-His121 signature. They all exhibit an alpha beta beta alpha-fold, similar to that found in the structure of zinc beta-lactamases. Many members of this superfamily are involved in mRNA maturation and DNA reparation. This fact suggests the hypothesis that metallo-beta-lactamases may be the result of divergent evolution starting from an ancestral protein which did not have a beta-lactamase activity. La première partie de cet article approche le problème de la véridiction de l’image et du jugement épistémique en prenant en considération la relation entre la textualité photographique et ses pratiques de production et de réception. On porte l’attention sur le fait que la textualité, qu’elle soit ou non photographique, ne peut pas être étudiée uniquement à partir de ses configurations plastiques et figuratives et de la constatation d’une cohérence sémiotique interne à l’image. En effet sa signification dépend fortement de ses pratiques productives (ou mieux encore de ce qu’on connaît des pratiques productives), de ses dispositifs « présentationnels » (comme par exemple l’album de famille, les affiches, la presse, Internet, etc.), de ses statuts (scientifique, documentaire, testimoniale, artistique, publicitaire) et de son histoire interprétative. La seconde partie de l’article analyse les stratégies énonciatives d’un reportage sportif, les Jeux Olympiques d’été de 2004 à Athènes. Le corpus d’images pris en considération construit différentes configurations de point de vue et diverses manières de mettre en présence de l’observateur le corps de l’athlète : des corps en action, des corps fatigués, des corps en équilibre, des corps concentrés, des corps tendant vers le but, enfin des corps avec lesquels nous construisons une « relation inhérente » grâce à une saisie analogisante. Nous avons ainsi individualisé trois types de mise en présence du corps et trois types de temps présent. Dans le premier type de présent, appelé dimensionné, le mouvement corporel entre en résonance avec la « traînée » des moments retenus et la « protension » de ceux qui se profilent comme possibles ; le second type est un présent d’appariement : dans ce deuxième cas l’exploration perceptive du photographe et du récepteur de l’image se coordonnent avec l’alternance des « points d’attaque » synchronisés des corps dansants. Le troisième type de présent, intervalle-échantillon, enfin, construit l’athlète en tant qu’identité et conscience en acte. [less ▲]

Bone sialoprotein (BSP) is a bone matrix glycoprotein whose expression coincides with terminal osteoblastic differentiation and the onset of mineralization. In this study we show that BSP expression is considerably increased in confluent Saos-2 human osteosarcoma cells and in differentiating normal human osteoblasts, concomitantly with the decrease of Runx2, a key transcription factor controlling bone formation. Therefore, we investigated the role of Runx2 in the regulation of BSP expression in Saos-2 cells. Using a mobility shift assay, we demonstrated that Runx2 binds to the BSP promoter only in preconfluent cells. Histone deacetylase 3 (HDAC3) has been recently shown to act as a Runx2 co-repressor. Chromatin immunoprecipitation assays demonstrated that both Runx2 and HDAC3 are detectable at the BSP promoter in preconfluent Saos-2 cells but not when they are confluent and overexpress BSP. Consistently, nuclear Runx2 protein level is down-regulated, whereas Saos-2 cells became increasingly confluent. Finally, the suppression of HDAC3, Runx2, or both by RNA interference induced the expression of BSP at both mRNA and protein levels in Saos-2 cells. Our data demonstrate that Runx2 and HDAC3 repress BSP gene expression and that this repression is suspended upon osteoblastic cell differentiation. Both the nuclear disappearance of Runx2 and the non-recruitment of HDAC3 represent new means to relieve Runx2-mediated suppression of BSP expression, thus allowing the acquisition of a fully differentiated and mineralization-competent phenotype by osteoblast cells. Bacterial cell division requires the coordinated action of cell division proteins and murein (peptidoglycan) synthases. Interactions involving the essential cell division protein FtsN and murein synthases were studied by affinity chromatography with membrane fraction. The murein synthases PBP1A, PBP1B, and PBP3 had an affinity to immobilized FtsN. FtsN and PBP3, but not PBP1A, showed an affinity to immobilized PBP1B. The direct interaction between FtsN and PBP1B was confirmed by pulldown experiments and surface plasmon resonance. The interaction was also detected by bacterial two-hybrid analysis. FtsN and PBP1B could be cross-linked in intact cells of the wild type and in cells depleted of PBP3 or FtsW. FtsN stimulated the in vitro murein synthesis activities of PBP1B. Thus, FtsN could have a role in controlling or modulating the activity of PBP1B during cell division in Escherichia coli. [less ▲]

This paper aims at reporting on the design of polymeric drug nanocarriers used in cancer therapy, with a special emphasis on the control of their biodistribution. First, the prominent role of poly(ethylene oxide) in the lifetime of nanocarriers circulating in the blood stream is highlighted, and the origin of a passive targeting based on a difference in the anatomy of tumors and normal tissues is discussed. The main body of the review is devoted to the targeting of nanocarriers towards tumors and the underlying concepts. As a rule, either the constitutive polymer is stimuli-responsive and the locus of drug release is where the stimulation occurs, or a ligand endowed with specific recognition is grafted onto the nanocarrier. Finally, the fate of the nanocarrier after drug delivery and the bioelimination of the polymer(s) involved are briefly considered. [less ▲]

Il apparaît que la zone étudiée est sortie de la « Grande Sécheresse » à la fin des années 1980. Cependant, les conséquences de ce retour à des conditions plus humides ne se seraient pas accompagné d’une amélioration significative de la longueur de la saison des pluies. [less ▲]
