Multifrequency impulse oscillometry (IOS) was compared with a monofrequency forced oscillation technique (MFO) in calves undergoing experimentally induced bronchoconstriction and subsequent bronchodilatation. The dynamic lung compliance (Cdyn) was also measured by conventional methods. For each test, the baseline mean and the responses to saline, a bronchoconstrictive agent (carbachol) and a bronchodilator (fenoterolhydrobromide) were calculated. Using the IOS, the information was markedly frequency-dependent. The resistance (R) and the magnitude of respiratory impedance (Z) were only sensitive at 5 Hz, leading to negative frequency dependence of these parameters as an indicator of peripheral airway obstruction. A high sensitivity for reactance (X) and phase angle phi values was observed between 5 and 20 Hz. For MFO (10 Hz), the parameters Ros (which includes resistive and capacitive components of the respiratory system), phase shift (psi), and the oscillatory derived compliance of the respiratory system (Crs) were of the greatest clinical potential. Crs showed a significant coefficient of linear correlation (r = 0.88, P < 0.001) with Cdyn. At the 10 Hz test frequency similar results were observed with MFO and IOS, suggesting that for healthy calves the measurement effect of an impulse is not significant. With respect to peripheral airway calibre, a test frequency less than 10 Hz appeared to be most sensitive and least variable. [less ▲]

The present paper deals with an analysis of metric (cranial measurements) and non metric (epigenetic cranial characters) variation in black rat populations from S. Miguel and Terceira islands compared to a continental sample. Assuming the Azorean rats originate from the European continent, we tested the similarity between the samples, considering only adult and subadult animals. Having verified that there was no sexual dimorphism in the characters studied, we computed discriminant functions and made a PCA with the most discriminant variables. The three samples are clearly separated by these functions, the Azorean rats showing a larger general skull size and differing between the two islands. The Mean Measure of Divergence (MMD), based on non metric cranial traits frequencies, was also used to express the interpopulational differences: the rats of Terceira differ significantly either from the continental ones or from those of S. Miguel. Finally, the possible origin of these differences is disussed. [less ▲]

The mammalian genome contains hundreds if not thousands of zinc finger protein (Zfp) genes. While the function of most of these genes remains to be determined, it is clear that a few of them play important roles in gene regulation and development. In studies described here, we have used an interspecific mouse backcross mapping panel to determine the chromosomal location of 15 KRAB-containing zinc finger loci. These loci map to nine different mouse autosomes and the X Chromosome (Chr). Two Chrs, 7 and 9, contain cosegregating pairs of KRAB-containing Zfp genes, indicating that the KRAB-containing Zfp genes have evolved through processes involving regional as well as genome-wide duplication events. L'article décrit les activités de recherche et d'enseignement en surveillance de l'environnement à la Fondation Universitaire Luxembourgeoise à Arlon. Il présente le contexte historique qui a initié ces activités, intégrant plusieurs chercheurs de disciplines diverses. Travailler sur le milieu naturel impose des contraintes, mais celles-ci sont compensées par une exigence modérée en ce qui concerne la précision de certains résultats. L'approche de l'unité de recherche utilise le milieu comme source d'information pour développer des méthodes de mesure, en exploitant notamment les concepts de bio-capteurs et de bio-indicateurs. L'agrégation des variables en indicateurs globaux et l'utilisation de modèles comme outils de surveillance de l'environnement figurent également parmi les originalités de la démarche. [less ▲]

A seven-year-old, female golden retriever was referred for a paroxysmal, chronic cough and dyspnea, dysphagia, facial pruritus, anterior uveitis, and deteriorating general condition. A severe, mixed interstitial and alveolar pattern, with poorly defined amorphous lesions, was seen on thoracic radiographs. Multiple, whitish nodules disseminated on the hyperemic respiratory mucosa were noted on bronchoscopy. Escherichia coli and Aspergillus fumigatus were cultured from the bronchoalveolar lavage. Granulomatous lesions in numerous organs were identified during necropsy, and Aspergillus fumigatus and Candida spp. were cultured from lung and kidney tissues. Microscopic granulomatous lesions were compatible with mycotic infection; however fungal organisms were not observed. [less ▲]

Most of the mutations causing deficiency of the pyruvate dehydrogenase (PDH) complex are in the X-linked E1 alpha gene. We have developed a rapid screening method for the detection of mutations in this gene using reverse transcription of total RNA, polymerase chain reaction amplification of the whole coding region of the gene and single-strand conformation polymorphism (SSCP) analysis. With this method, we studied eight patients with a PDH complex deficiency, using cultured fibroblasts. In all patients, aberrant SSCP patterns were found and, after sequencing of the corresponding fragments, we were able to identify six new mutations and two mutations already described previously. The mutations are point mutations leading to amino acid substitutions (5) and direct repeat insertions (3). The presence of the mutations was confirmed in genomic fibroblast DNA. The 4 female patients were shown to carry both a normal and a mutated E1 alpha gene. [less ▲]

The impact of the codon bias of the mouse tumour necrosis factor alpha (mTNF) gene cloned in Streptomyces lividans on the efficiency of expression and secretion was analysed. Minor codons occurring in the mTNF gene were therefore adapted to the codon bias of Streptomyces by site-directed mutagenesis. No improvement in mTNF yield could be detected. The stability of the transcript derived from the construct was shown to be more important for determining the final level of mTNF production. A strong correlation was observed between the yield of secreted biologically active mTNF and the amount of mTNF mRNA present in the cells. [less ▲]

Monofrequency forced oscillometry (MFO) using a sinusoidal test signal of one frequency (10 Hz) was compared with the classical pulmonary function technique (using an oesophageal balloon with airflow measurements) in two breeds of cattle which differ in their respiratory physiology, particularly to assess changes in upper airway resistance. Six healthy male Friesian calves (mean [SD] bodyweight 131 [12] kg) and six healthy male blue Belgian calves (mean [SD] bodyweight 137 [13] kg) were examined successively with their heads either in a normal position or held vertically, using a cross-over study design. In the normal position, higher resistances, a higher respiratory impedance, and lower values of the dynamic lung compliance (Cdyn) were measured in the blue Belgian than in the Friesian calves. In the vertical head position, total pulmonary resistance (RL) increased and Cdyn decreased significantly (P < 0.05). The changes in MFO-derived parameters were in accordance with the changes in classical parameters, and for both methods, the percentage changes were markedly higher in the blue Belgian calves.
