The aim of our study was to determine the effect of a rectocolonic preimmunization with ovalbumin on the systemic immune response induced by a subsequent subcutaneous injection of this antigen in Balb/c mice. One rectocolonic, but not intragastric, administration of 25 mg of ovalbumin induced a detectable increase in serum anti-ovalbumin antibody level. The level reached was however much lower than after subcutaneous injection. Both intragastric and rectocolonic immunization with ovalbumin induced specific systemic cellular tolerance. However, after rectocolonic, but not intragastric, preimmunization there was no systemic humoral tolerance to this antigen. These differences in systemic immune responses after rectocolonic or intragastric administration of ovalbumin could be due to different stimulation of the systemic immune system or to differences between the colonic and small bowel mucosal immune system, which remain to be elucidated. The document is only available in French Main frame: Opiate dependant patients are followed in a methadone maintenance program in family practice. Drug abuse of those patients has been studied on both quantitative and qualitative basis. A bibliographic review of Rohypnol™ use as a street drug and a short survey among family doctors of different countries reinforce the study. Results: 53 of 89 methadone maintenance patients have used medicinal substances as drugs. 90% (47/53) have used at least once Flunitrazepam by mouth. Rohypnol™, a potent hypnotic which is the Belgian brand name of Flunitrazepam sold by the Roche company, is given as first choice by 66 % (35/53). The study of 56 interviews of these patients, written or recorded (audio) give a very good insight of the clinical picture "Etre en Roche"[To be on Roche], as the patients call it. A pill of Rohypnol™ is called a "Roche" and a compulsive user is called a "Rocheman". The compulsive use as an heroin substitute, often with alcohol, is the main characteristic of this picture. Euphoric semiconscious state ("to be ravaged, stoned, dead"), dysarthria, ataxia, feeling of being invincible, loss of inhibition with possible uncontrollable aggression and acting out followed by falling asleep and full anterograde amnesia. A bibliographic review on Flunitrazepam abuse and a General practitioner's survey in various countries confirm those facts. Conclusions: The Rohypnol™ is an hard and heroin like drug. This product gives amnesia and rid inhibitions. He has no more place in therapeutic. [less ▲]

A method has been derived for the fire design of concrete columns in the fire situation. This method has been derived from numerical calculations made with the non linear software SAFIR and has been calibrated to obtain the best fit with the results of 76 full scale experimental tests: 16 performed in Belgium (University of Liege and University of Gent), 39 in Germany (Braunschweig) and 21 in Canada (NRC). The model proposed here has been slightly modified in a later version that is now present in Eurocode 2 (EN 1992-1-2), as tabulated data method A (section 5.3.2). This publication is thus a background document of EN 1992-1-2. [less ▲]

This article describes the optimization of the hybridization signal obtained with biotinylated oligonucleotides. Optimal number and positions of biotin moieties on a 33-base oligonucleotide probe were determined. The quality of avidin-peroxidase conjugate and the choice of chromogenic substrate influenced detection sensitivity. A signal amplification method was also developed for avidin enzymatic conjugates. These improvements allowed the detection of less than 0.02 fmol of target DNA. [less ▲]

A simple stochastic model of Darwinistic evolution generating phylogenetic-like trees is developed. The model is based on a branching process taking competition-correlation effects into account. In presence of finite and short range correlations, the process self-organizes into a critical steady-state in which intermittent bursts of activity of all sizes are generated. On a geological-like time scale, this behaviour agrees with punctuated equilibrium features of biological evolution. The simulated phylogenetic-like trees are found to be self-similar. The dynamics of the transient regimes show a power law decrease of the order parameter towards the 0(+) value which characterizes an unstable critical state. The genetic range k of competition-correlations between living species is found to be a relevant parameter which determines the universality class of the evolution process. An infinite competition-correlation range destroys however the self-organized critical behaviour. The fractal dimension D-f of the phylogenetic-like trees increases from 2.0 to infinity as k goes from 1 to infinity. The critical exponent tau of avalanche size-distribution decreases from about 3/2 (for k = 1) and reaches about 1.2 for k = 10. A hyperscaling relation seems to relate the various universality classes. Through a mean-field theory, we mention that the evolution process is much more complex than a simple uncorrelated branching process. [less ▲]

Progelatinases A and B were purified from HT1080-conditioned culture medium using a continuous-elution electrophoresis. Initially cell culture medium was ammonium sulfate precipitated. The concentrated proteins were affinity chromatographed on gelatin-Sepharose column. The bound gelatinases were eluted with electrophoresis sample buffer and subjected to continuous-elution electrophoresis. In a single run, under the standardized working conditions, the obtained fractions contained four purified enzymes--progelatinase A (M(r) 72,000), its activated forms (M(r) 62,000 and M(r) 59,000), and progelatinase B (M(r) 92,000). Moreover, the continuous-elution electrophoresis allowed the enzymes separation from their respective inhibitors--TIMP-1 (M(r) 28,500) and TIMP-2 (M(r) 21,000). The purified progelatinases A and B demonstrated high specific activities (150-200 U/micrograms). [less ▲]
