The effect of estradiol-17β (E2) supplementation on superovulation with (PMSG) or (FSH) was investigated in Swamp buffalo. Sixty-eight buffalo were treated in seven groups. Group 1 served as control and was superovulated by standard PMSG or FSH treatment used in routine bovine embryo transfer protocols. Group 2 was superovulated by standard PMSG regimen plus two injections of E2 at a 48 h interval beginning one day before the onset of gonadotropin treatment (short-term supplementation) for a total dosage of 2.5 mg E2; Groups 3 and 4 received the same regimen as Group 2, but in doses of 5.0 and 7.5 mg E2, respectively. Group 5 received the standard FSH regimen (40% LH). Group 6 received short-term E2 (7.5 mg) supplementation of FSH-p. Group 7 was superovulated by standard FSH regimen (40% LH) plus three injections of E2 at 48–72 h intervals beginning five days before the onset of gonadotropin treatment (long-term supplementation) for a total dosage of 7.5 mg E2. The number of corpora lutea (CL) and follicles ≥ 8 mm in diameter were recorded by palpation per rectum and after slaughter. The mean numbers of CL and follicles were 0.99, 5.8, 8.0, 10.6, 4.0, 3.9, 8.1 and 0.25, 6.8, 6.2, 6.2, 1.6, 0.0, 4.1 for Groups 1, 2, 3, 4, 5, 6, 7, respectively. In Group 7, the rates of nonsurgical and postmortem embryo recovery were 46 and 90.4%, respectively and 54.4% of the collected ova were fertilized. These results indicate the possibility of producing viable embryos in buffalo by using E2 supplementation for the gonadotropin treatment. [less ▲]

The aim of this study was to determine the role of octreotide administration in acromegalic patients as a preparation for selective adenomectomy using transsphenoidal route. Octreotide was administered for 3 to 6 weeks before surgery in 12 patients and for 4 to 39 months in 25 patients. The clinical response was judged as excellent or good in 10 of 12 patients from group I and in 23 of 25 patients from group II. Marked reduction (ie, greater than 50% of initial values) in serum growth hormone (GH) levels was seen in all patients, with levels to less than 5 micrograms/L in 68% of patients and less than 2 micrograms/L in 27%. Insulin-like growth factor 1 (IGF-1) levels decreased to within normal limits in half the cases. During long-term treatment, an escape phenomenon could be seen. Varying degrees of tumor shrinkage were seen in more than 50% of cases. During surgery, with regard to the relative ease or difficulty in removing the tumor, the consistency of the tumor and the separation of normal from pathological tissue, no significant difference was observed between patients given octreotide and those from control series. Morphological changes in adenomatous tissue were rather small. The surgical outcome was similar in the pretreated series as in the control series, except in enclosed adenomas, which showed a tendency to a higher success rate. Since octreotide improves both the clinical condition and hormonal parameters and induces varying degrees of tumor shrinkage, it is potentially useful as an adjunct to surgery. Morphological data suggest that octreotide exercises a functional inhibitory effect on GH release. [less ▲]

Techniques and methods required for accurate and reproducible measurements of transdiaphragmatic pressure (Pdi), were studied in seven-week and five-month-old calves. Since reliable pleural pressure measurement in cattle has already been described, separate experiments were conducted to compare retrodiaphragmatic pressures measured (i) with balloon-catheters and needle catheters, (ii) before and after eructation and (iii) in the ruminal and in the peritoneal cavities. Whereas respiratory-induced pressure changes were equally transmitted throughout the rumen content, a marked anteroposterior damping was observed as the recording site in the abdominal cavity was moved away from the diaphragm (P < 0.0001). It was concluded that the most appropriate and accurate way to measure Pdi changes in polygastric animals is to measure the pressure changes existing in the mid-thoracic portion of the oesophagus and in the liquid content of the rumen with balloon-tubing units. Neither growth- and diet-related rumen enlargement nor eructation, which are two typical characteristics of ruminants, altered the reliability of retrodiaphragmatic pressure measurements in the two age groups studied. A photoactive protochlorophyllide-protein complex with absorbance and fluorescence maxima at 648 and 653 nm was detected in greening barley leaves without any re-darkening. The variations of the amplitudes of the absorbance and the fluorescence of the photoactive protochlorophyllide with greening time at two different light intensities indicate a close relationship between the rate of chlorophyll synthesis and the amount of the complex during the first hours. The chlorophyllide resulting from photoreduction during greening has an absorbance maximum at 684 nm, which shifts towards a shorter wavelength within a few seconds, indicating rapid liberation of the pigment from the enzyme. We conclude that chlorophyll accumulation proceeds through continuous regeneration and phototransformation of the photoactive complex. [less ▲]

The synthesis and use of functional diethylaluminum alkoxides (Et2AlORX) as initiators for the ring-opening polymerization of ε-caprolactone have been investigated as a possible strategy to prepare omega-primary amino poly(ε-caprolactone) (PCL). In a first approach, an initiator containing ethylphthalimide as the functional-RX group has been synthesized. That protected amino group has been successfully attached at the end of PCL chains. The conversion of the phthalimide end group into a primary amine has however failed, whatever the experimental conditions. In an alternative approach, an alkyl bromide has been considered as the functional group to be attached to PCL chains, via the appropriate initiator. The end functionalization was quantitative and the bromo end group converted into an azido group which was subsequently reduced to the expected primary amine. Omega-primary amino PCL has proved to be an effective macroinitiator for the ring-opening polymerization of γ-benzylglutamate N-carboxy anhydride with the formation of a poly(caprolactone-b-peptide) diblock copolymer.
