[more ▼] The abundances of nine elements in the atmospheres of 20 extreme metal-poor stars are derived from green spectra obtained with the ESO Cassegrain Echelle Spectrograph (CASPEC). Sodium is found to be overabundant with respect to aluminum by as much as one dex. There is some scatter in Ca/Fe which cannot be accounted for by random errors in equivalent widths or atmospheric parameters. Scandium is overabundant relative to iron. Some barium lines may be affected by strong departures from local thermodynamic equilibrium. The nitrogen-rich metal-poor star HD 74000, which was found in previous work to be also Al-rich, also presents higher than usual abundances of Na and of s-process elements. It can thus be considered as a mild barium dwarf of Population II. In group 1, Limousin embryos were transferred nonsurgically into Ayrshire recipients (10 heifers and 1 cow), and in group 2, 11 Ayrshire heifers and 1 cow were inseminated artificially to Ayrshire bulls. Blood samples were collected daily from d 265 of gestation until d 15 postpartum from 5 heifers of each group. Milk yield was recorded on alternate weeks during the first 20 wk postpartum. Calf birth weight was higher (44.2 vs. 35.4 kg) and gestation was longer (297.4 vs. 280.2 d) in Ayrshire dams bearing Limousin fetuses than in those bearing Ayrshire fetuses. Daily milk production for the first 20 wk was lower (18.1 vs. 20.8 kg) in Ayrshire dams bearing Limousin fetuses than in those bearing Ayrshire fetuses. Prepartum decrease in progesterone concentrations and increase in estrone concentrations were faster in Ayrshire heifers bearing Limousin fetuses than in those bearing Ayrshire fetuses. Profiles of peripartum concentrations of bovine placental lactogen differed between Ayrshire dams carrying different families of Limousin fetuses but were similar in those carrying families of Ayrshire fetuses. The concentrations of 15-keto-13,14-dihydroprostaglandin F2 were lower during the postpartum period in heifers that gave birth to Limousin calves than in those that had Ayrshire calves. In conclusion, the breed of fetus influences physiological and endocrine responses of the dam, which might have some effect on milk production of the dam. [more ▼] The sites of androgen and estrogen action on sexual behavior were studied in the preoptic area of castrated male Japanese quail by stereotaxic implantation of hormones, antihormones and metabolism inhibitors. The first experiment demonstrated that bilateral implantation of the aromatase inhibitor, androstatrienedione (ATD), in the sexually dimorphic nucleus (POM) of the preoptic area can completely suppress the behavioral activation produced by a systemic treatment with testosterone. The effects of ATD were only observed if the implants were located in the POM. In the second experiment, implants in the POM of the synthetic estrogen, diethylstilbestrol, restored copulatory behavior in castrated males while implants of the synthetic nonaromatizable androgen, methyltrienolone, were almost ineffective. During the third experiment, the activating effects of a systemic treatment with testosterone were blocked by stereotaxic implants in the POM of the antiestrogen, tamoxifen, or the antiandrogen, flutamide. The effects of tamoxifen were more pronounced than those of flutamide. In addition, tamoxifen was active in all parts of the POM while a behavioral inhibition was observed only for flutamide implants which were located in the caudal part of the nucleus. Taken together, these results demonstrate that the sexually dimorphic POM is the area where the behaviorally active estrogenic metabolites of T have to be produced. The estradiol derived from T aromatization presumably acts within the POM to activate copulation as demonstrated by the effectiveness of DES implanted in this region. Androgens also have a direct action on sexual behavior as suggested by the partial inhibition observed in flutamide-treated birds. It is, however, suggested that androgens and estrogens do not act in the same brain area to activate behavior. All three residues of the SDN loop in Streptomyces albus G beta-lactamase were modified by site-directed mutagenesis. The mutant proteins were expressed in Streptomyces lividans, purified from culture supernatants and their kinetic parameters were determined for several substrates. Ser130 was substituted by Asn, Ala and Gly. The first modification yielded an almost totally inactive protein, whereas the smaller-side-chain mutants (A and G) retained some activity, but were less stable than the wild-type enzyme. Ser130 might thus be involved in maintaining the structure of the active-site cavity. Mutations of Asp131 into Glu and Gly proved to be highly detrimental to enzyme stability, reflecting significant structural perturbations. Mutation of Asn132 into Ala resulted in a dramatically decreased enzymic activity (more than 100-fold) especially toward cephalosporin substrates, kcat. being the most affected parameter, which would indicate a role of Asn132 in transition-state stabilization rather than in ground-state binding. Comparison of the N132A and the previously described N132S mutant enzymes underline the importance of an H-bond-forming residue at position 132 for the catalytic process.
