VO2 max, maximum oxygen uptake, has been measured in 4 normal young men, before and after beta-adrenergic blockade (0.5 to 5 mg Pindolol by mouth). Pindolol induces bradycardia and reduces VO2 max. A statistically significant positive correlation appears between posology of Pindolol and bradycardia, this posology and reduction of VO2 max, and finally between bradycardia and reduction of VO2 max. These correlations indicate that the reduction of VO2 max is best explained by a circulatory limitation of oxygen supply to active muscles. [less ▲]

In some domestic animal species, cow, ewe, sow, mare, luteal regression and therefore oestral regulation, depends on the action of a luteolytic of uterine origin. This luteolysine should be but a prostaglandin F2 alpha or an analogue. Progesterone is the regulator of the cycle; plasma level is low (basic level) at day of oestrus (0,5ng/ml) then increases gradually to reach a peak value of 6 to 9 ng/ml at days 6 to 16 of the cycle. Beta-oestradiol level fluctuates including a main peak before ovulation (9 pg/ml) and three accessory peaks at days 4-5, 8, 12-14 of the cycle; however the peak of the 8th day is inconstant. FSH and LH levels are constant during dioestrus, peak level values are observed at the beginning of oestrus, they are practically superposed and of a 6 to 9 hours duration. Administration of prostaglandins during luteal phase produces hormonal changes similar to those observed during the normal cycle. Because of its luteolytic action and its effect on uterine fibre, prostaglandin F2 alpha offers a special interest for stock farming and veterinary medicine. So a full expression can be given to AI and a more extended application of egg transfer can be foreseen. It constitutes an effective therapy in the case of corpus luteum persistence and a way to induce parturition. [less ▲]

The hormonal pattern was studied in 54 animals: 34 heifers with seasonal anoestrus, 13 cows with post-partum anoestrus, 5 cows with nymphomania and 2 cows superovulated. Seasonal and post partum anoestrus are characterized by basal levels of progesterone, FSH and LH, while oestradiol level is more variable and higher in post partum anoestrus due to the proper activity of the ovary. Nymphomania is a hyperfolliculinemic syndrome; cystic puncture leads to a fall in oestradiol and a surge in FSH and LH but of inequal intensity. The level of the latter hormone is often too low to induce luteinization; cystic degeneration of the ovary is associated with LH deficiency. In superovulated cows, hormonal pattern is like in normal cyclic cows but more pronounced. [less ▲]

Chick embryo fibroblasts cultivated in vitro were treated with ethidium bromide (E.B.) or with DNA-E.B. complex (DNA-E.B.). E.B. (5 mug/ml) provokes morphological alterations and cell death, inhibits DNA synthesis and mitotic activity. DNA-E.B. (E.B. 5 mug/ml) is less toxic to the fibroblasts as far as cell structure, DNA synthesis and mitotic activity are concerned. DNA alone has no apparent effect on the fibroblasts. As shown by fluorescence microscopy, the lower toxicity of DNA-E.B. seems to be related to its mode of penetration into the cells. [less ▲]

We have examined whether glucocorticoids control the activity and (or) the subcellular distribution of protein kinase dependent on cyclic AMP (adenosine 3':5'-monophosphate), since they influence cyclic-AMP-dependent responses to other hormones. Protein kinase activity was determined in rat liver homogenates and subcellular fractions, nuclear, large granular, microsomal and supernatant obtained by differential sedimentation in 0.25 M sucrose. 63% of the tissue protein kinase activity detected in absence of cyclic AMP reside in the particulate fractions. Upon addition of exogenous cyclic AMP, protein kinase activity is stimulated 1.8, 1.2, 1.2 and 4.5-fold in nuclear, large granular, microsomal and supernatant fractions, respectively. Under these conditions, 66% of tissue activity are found in the supernatant fraction. The activity sensitive to exogenous cyclic AMP resolves into a major (84%) cytosoluble and a minor (16%) nucleomicrosomal component. The latter activity resists elution with isotonic saline and is increased in the presence of Triton X-100. Three groups of rats were studied: control and adrenalectomized with or without cortisol treatment. In whole liver homogenates, both protein kinase activity detected in absence of exogenous cyclic AMP and sensitivity of the enzyme to cyclic AMP were comparable in all groups. Moreover, the distribution patterns of proteins kinase activity amoung the fractions were essentially the same in all groups of animals, whether or not particles had been treated with Triton X-100. Finally, in cell-free experiments, glucocorticoids alone or in combination with their intracellular receptor did not modify protein kinase activity of rat liver. Thus the results reported do not support the possibility that glucocorticoids influence cyclic AMP-dependent protein kinase in rat liver. Yet, this study provides data, not available before, on subcellular distribution of this enzyme in rat liver. [less ▲]

Relative lifetimes and energies above the dissociation limit have been determined for the rotational predissociation of several quasibound levels of the X1Sigma+ state of 4HeH+. In particular the lifetimes are very sensitive to the shape of the potential energy curve. These measurements are used to discriminate between two ab initio potential curves which differ by only 0.00004 a.u. (about 1 meV). Using the lifetime data, relative population factors were determined for the observed levels. [less ▲]
