Strains R39 and R61 are soil isolates. Their designations are arbitrary. In strain R39, the cross-link between the peptide units of the wall peptidoglycan extends from the C-terminal D-alanine of one unit to the amino group at the D-center of meso-diaminopimelic acid of another unit (peptidoglycan of chemotype I). The interpeptide bond is in position to a free carboxyl group. In strain R61, the cross-link extends from a C-terminal D-alanine of a peptide unit to a glycine residue attached to the amino group of LL-diaminopimelic acid of another peptide unit (peptidoglycan of chemotype II). The exocellular DD carboxypeptidases-transpeptidases produced by both strains catalyze hydrolysis, react with β-lactam antibiotics. This chapter explains the assay methods for DD-Carboxypeptidase activity like the standard reaction, chemical estimation of free Alanine, as well as, assay method for β-Lactamase. Membrane bound LD-transpeptidase of Streptococcus faecalis ATCC 9790 was used to catalyse transpeptidation reactions between non radioactive peptide donors of the general type: L-Ala-D-Glu (L)meso-A2pm(L)-D-Ala and D-(14C) alanine acceptor. The presence of one or two amide residues on the carboxyl groups of glutamic acid and meso-diaminopimelic acid increases the transfer reactions and subsequently the yield in radioactive peptides L-Ala-D-Glu (L)meso-A2pm(L)-D-(14C) Ala. Among 150 temperature-sensitive Saccharomyces cerevisiae mutants which we have isolated, 15 are specifically affected in ribonucleic acid (RNA) synthesis. Four of these mutants exhibit particularly drastic changes and were chosen for a more detailed study. In these four mutants, RNA synthesis is immediately blocked after a shift at the nonpermissive temperature (37 C), protein synthesis decays at a rate compatible with messenger RNA half-life, and deoxyribonucleic acid synthesis increases by about 40%. All the mutations display a recessive phenotype. The segregation of the four allelic pairs ts-/ts+ in diploids is mendelian, and the four mutants belong to three complementation groups. The elution patterns (diethylaminoethyl-Sephadex) of the three RNA polymerases of the mutants grown at 37 C for 3.5 h show very low residual activities. The in vitro thermodenaturation confirms the in vivo results; the half-lives of the mutant activities at 45 C are 10 times smaller than those of the wild-type enzymes. Polyacrylamide gel electrophoresis shows that the synthesis of all species of RNA is thermosensitive. The existence of three distinct genes, which are each indispensable for the activity of the three RNA polymerases in vivo as well as in vitro, strongly favors the hypothesis of three common subunits in the three RNA polymerases. After a treatment based on the administration of gonadotropic drugs, aimed at reduced school difficulties and caracterial difficulties by a premature onset of puberty, a young male aged 13 1/2 presents: 1 degree somatic anomalies; 2 degrees increase of intellectual, affective and behavioral troubles; 3 degrees perturbations of EEG prophiles; 4 degrees organicity tests significantly disturbed. The interruption of treatment (4 years of observation) was followed by a progressive remission, however heterogenous, of all initial troubles. The observation underlines the dangers of utilising these treatments in a pre-puberty boy. [less ▲]

This article examines the recent development of urbanized areas in Belgium and endeavours to measure the space consumed by some of the important human activities: industrial operations, main arteries, housing and urban functions. Notwithstanding a considerable lack of data, the gravity of the situation in Belgium is clear : since 1929, the amount of urbanized area has more than doubled, and, in 1970, the proportion of national territory which was urbanized (28 %) was far higher than in any of the neighbouring countries. It is therefore essential to control the use of land by means of a dynamic policy in country planning and development. The waste of land must be stopped by promoting the concentration of activities in specific centres, by keeping the majority of functions in the heart of the urban network, by assisting the use and re-use of available sites and by controlling any new developments, particularly housing schemes. As a pure compound, escine-Na inhibits exsudative oedema induced locally in the rat skin by dextran (MW (0000). This inhibition is due, for a small part, to the release of adrenaline by adrenal medulla; for an other part, to an interaction with glyco-cortico-steroids from the adrenal cortex and, finally, to direct potent vasoconstriction. The exocellular DD-carboxypeptidase-transpeptidase of Streptomyces R39 is inhibited by beta-lactam antibiotics according to the same general scheme of reaction as the exocellular DD-carboxypeptidase-transpeptidase of Streptomyces R61. However, the values for the kinetic constants involved in the reaction are very different for the two enzymes and provide an explanation for the observation that the R39 enzyme is more sensitive to beta-lactam antibiotics than the R61 enzyme. Further, particular beta-lactams influence the kinetic constants to different extents depending on the source of the enzyme, so that a physical basis for the spectrum of antibiotic activity against particular enzyme systems is provided.
