Despite its intensive use in adjuvant breast cancer therapy for more than 30 years, the exact mechanisms of action of tamoxifen have not yet been fully characterized. Tamoxifen was recently shown to restore the E-cadherin function of human breast cancer MCF7/6 cells and to suppress their invasive phenotype. Because tamoxifen interacts with targets implicated in Ca2+ homeostasis, we explored the possibility that the restoration of E-cadherin function in MCF7/6 cells induced by this drug could be affected by Ca2+ modulators. Two different Ca2+ channel antagonists (verapamil and nifedipine) potentiated the effect of tamoxifen on E-cadherin function, as evaluated with a fast cell aggregation assay. These molecules decreased the tamoxifen concentration needed to restore the E-cadherin function from 10(-6) M to 10(-7) M. When incubated with a Ca2+ channel agonist, Bay K8644 (methyl-1,4-dihydro-2,6-dimethyl-3-nitro-4-(2-trifluoro-methylphenyl)- pyridine-5-carboxylate), the effect of tamoxifen on E-cadherin function was completely abolished. These results demonstrate that the restoration of the E-cadherin function induced by tamoxifen depends, at least in part, on a Ca2+ pathway, and support the evidence of an effect of tamoxifen on Ca(2+)-dependent mechanisms. Our data also suggest that Ca2+ channel modulators could make it possible to decrease the dose of tamoxifen administered to patients without reducing the therapeutic effects. [less ▲]

Block copolymers of the ABA type, where B is polybutadiene (PBD) and A is poly(ethyl methacrylate) (PEMA), poly(tert-butyl methacrylate) (P-t-BMA), poly(methyl methacrylate) (PMMA), or poly(isobornyl methacrylate) (PIBMA), have been successfully synthesized by sequential anionic polymerization of butadiene and methacrylates with the diadduct of tert-butyllithium to m-diisopropenylbenzene as a difunctional initiator. Block copolymers of a narrow molecular weight distribution (1.10) have been analyzed by differential scanning calorimetry, transmission electron microscopy, and dynamic mechanical analysis. These materials are phase-separated and have high mechanical performances. Special attention has been paid to the service temperature of these thermoplastic elastomers in comparison with a styrene−butadiene−styrene (SBS) triblock copolymer. The upper service temperature (UST) has been estimated from the temperature dependence of the tensile properties in the 25−150 °C range and found to change with the outer blocks. Polystyrene (Tg = 100°C) is at the origin of the lower UST, which is however comparable to PEMA (Tg = 90 °C) and P-t-BMA (Tg = 116 °C) containing triblock copolymers of similar molecular weight and composition. PMMA (Tg = 132 °C) outer blocks increase the UST of the triblocks, which is further increased by hydrogenation of the PBD midblock. The higher UST has been found for a PIBMA (Tg = 202 °C)-containing triblock copolymer that shows an ultimate tensile strength higher than 2 MPa at 150 °C. [less ▲]

Thermoreversible gelation of syndiotactic poly(methyl methacrylate) (sPMMA)−polybutadiene (PBD)−sPMMA, triblock copolymers in o-xylene has been studied in the presence of isotactic PMMA (iPMMA) in a large range of syndio/iso mixing ratio (s/i). Differential scanning calorimetry (DSC) shows that two types of association contribute to the gel formation, i.e., the self-aggregation of the sPMMA outer blocks and the stereocomplexation of the sPMMA outer blocks with iPMMA. At low iPMMA contents (s/i = 30/1), self-association of the outer blocks accounts for the gelation process, although stereocomplexation occurs. Stereocomplexation becomes the dominant gelation mechanism at a high enough iPMMA content (s/i = 2/1). Thus, gelation is observed at a copolymer concentration as low as 1 wt %. The time dependence of the storage (G‘) and loss (G‘‘) moduli has been measured in the 0.08−1Hz frequency range in order to probe the gelation process. At the gel point, where the loss angle (tan δc = G‘‘/G‘) is independent of the probing frequency, the sample fits in the typical power law G‘(ω) G‘‘(ω) ωΔ. While the gelation time is greatly influenced by the s/i mixing ratio, the scaling exponent Δ is essentially constant in the range of 0.70−0.75. The Δ value agrees with theoretical predictions as well as with experimental values reported for some chemical gels but it is different from the experimental values reported for most physical gels. The Δ value is also independent of the PBD microstructure and it also holds for sPMMA-polystyrene (PS)−PBD−PS−sPMMA (MSBSM) pentablock copolymers. Therefore, the value of Δ appears to be a typical exponent for thermoplastic gels based on triblock copolymers of the ABA type in o-xylene. [less ▲]

A 37LRP/p40 polypeptide is of major interest because it is consistently up-regulated in cancer cells in correlation with their invasive and metastatic phenotype. Furthermore, this polypeptide presents intriguing multifunctional properties because it has been characterized as the precursor of the metastasis-associated 67-kD laminin receptor (67LR) and as a cytoplasmic ribosomal-associated protein. The isolation of the 37LRP/p40 gene is a prerequisite for identifying the molecular mechanisms responsible for the constant up-regulation of the 67LR expression in cancer cells. To date, the active 37LRP/p40 gene has never been identified in any species due to the existence of multiple pseudogenes in most vertebrates genomes. In this study, we report for the first time the gene structure and potential regulatory sequences of the 37 LRP/p40 gene. The chicken genome was selected to undergo this characterization because it is the only known vertebrate that bears a single 37 LRP/p40 gene copy. The 37 LRP/p40 active gene is composed of 7 exons and 6 introns and bears features characteristic of a ribosomal protein gene. It does not bear a classical TATA box and it exhibits several transcription initiation sites as demonstrated by RNase protection assay and primer extension. Analysis of potential regulatory regions suggests that gene expression is driven not only by the 5' genomic region but also by the 5' untranslated and intron 1 sequences. On the basis of gene structure and extensive protein evolutionary study, we found that the carboxyterminal domain of the protein is a conserved lock-and-key structure/function domain that could be involved in the biosynthesis of the higher-molecular-weight 67-kD laminin receptor in vertebrates, whereas the central core of the protein would be responsible for the ribosome associated function. The first identification of the active 37LRP/p40 gene presented in this study is a critical step toward the isolation of the corresponding human gene and the understanding of the molecular mechanisms involved in the up-regulation of its expression during tumor invasion and metastasis. A large body of work has been devoted to tumor necrosis factor alpha or interleukin-1 beta (IL-1 beta) signaling leading to the activation of the transcription factor nuclear factor-kappa B (NF-kappa B) in various cell types. Several studies have indicated that NF-kappa B activation depends strictly on the production of reactive oxygen intermediates. In this report, we first demonstrated that IL-1 beta is a potent activator of NF-kappa B in various epithelial transformed cell lines (OVCAR-3, SKOV-3, MCF7 A/Z). In these cells, IL-1 beta rapidly induces NF-kappa B through a complete degradation of I kappa B-alpha, while H2O2 activates NF-kappa B with slower kinetics through a partial degradation of I kappa B-alpha, p100 and p105. We showed that IL-1 beta-mediated induction of NF-kappa B in OVCAR-3 and in other epithelial cell lines does not proceed through the production of reactive oxygen intermediates, while the same cytokine activates NF-kappa B in lymphoid cells through the intracellular generation of H2O2. Our study demonstrated that several signaling pathways lead to the activation of NF-kappa B, following IL-1 beta treatment in different cell types. [less ▲]

The distribution of 211 particles, the superconducting 123 grain boundary and the solid-liquid interface features are emphasized. The quality of the materials has been investigated by thermopower and thermal conductivity measurements. A simple alkyl lithium initiator (sBuLi) complexed with LiCl (10 molar equiv.) has been used to initiate the anionic polymerization of various methacrylic monomers, i.e. methyl methacrylate (MMA), tert-butyl methacrylate (tBuMA), glycidyl methacrylate (GMA), and dimethyl amino ethyl methacrylate (DMAEMA) in tetrahydrofuran at -78°C. Only the homopolymerization of tBuMA proceeds in a living manner, as evidenced by both the linear plot of experimental molecular weight (Mnexp) vs theoretical ones (Mncal,) (slope % 0.93) and monomer resumption experiments. However, three types of block copolymers, i.e. PtBuMA-b-PMMA, PtBuMA-b-PGMA, PtBuMA-b-PDMAEMA, have been successfully synthesized with a predictable molecular weight, and narrow molecular weight distribution. Positron emission tomography (PET) is now primarily used in oncological indication owing to the successful application of fluorine-18 fluorodeoxyglucose (FDG) in an increasing number of clinical indications at different stages of diagnosis, and for staging and follow-up. This review first considers the biological characteristics of FDG and then discusses methodological considerations regarding its use. Clinical indications are considered, and the results achieved in respect of various organs and tumour types are reviewed in depth. The review concludes with a brief consideration of the ways in which clinical PET might be improved. [less ▲]
