[more ▼] Auditory neurons cultured from adult rat acoustic ganglia require for survival either a substrate bound factor(s) present in astrocyte conditioned medium or substrate bound basic fibroblast growth factor (bFGF). Nerve growth factor (NGF) is not a survival factor for these neurons in vitro, but when used in combination with substrate bound bFGF, NGF does vigorously stimulate a neuritogenesis response by these neurons. Transforming growth factor beta (TGF beta 1) enhances the survival effect that bFGF has on these adult auditory neurons but does not by itself promote their survival in dissociated acoustic ganglion cultures. We propose that there may be complex interactions and synergy exerted by these growth factors (i.e. bFGF, NGF, TGF beta 1) during injury to the inner ear. Water-soluble tryptic-digest peptides were selectively produced from PBP5, their N-terminal regions were sequenced and synthetic oligonucleotides were used as primers to generate a 476 bp DNA fragment by polymerase chain reaction. On the basis of these data, the PBP5-encoding gene was cloned in Escherichia coli by using pBR322 as vector. The gene, included in a 7.1 kb insert, had the information for a 678-amino acid-residue protein. PBP5 shows similarity, in the primary structure, with the high-molecular-mass PBPs of class B. In particular, amino acid alignment of the enterococcal PBP5 and the methicillin-resistant staphylococcal PBP2' generates scores that are 30, for the N-terminal domains, and 53, for the C-terminal domains, standard deviations above that expected for a run of 20 randomized pairs of proteins having the same amino acid compositions as the two proteins under consideration. We showed that chemical modification of the carboxylic groups of glutamic acid and aspartic acid residues restored a normal electrophoretic mobility and that the anomalous behaviour of that protein on SDS/PAGE was due to its very large negative charge at neutral pH. We also compared the behaviour of the same enzyme on gel filtration in the presence of SDS with those of other class A beta-lactamases (Mr approx. 30,000). These experiments suggested that the very low electrophoretic mobility of the Actinomadura R39 beta-lactamase upon SDS/PAGE was more probably due to a low degree of SDS binding rather than to an unusual shape of the SDS-protein complex. [more ▼] The effects of gonadectomy combined or not with testosterone (T) therapy on the sexual behavior, cloacal gland area, staining density and alpha 2-adrenergic receptors in the medial preoptic nucleus (POM) were studied in male and female Japanese quail. In castrated males, T increased the cloacal gland size and activated copulatory behavior. In agreement with previous reports, these effects were sexually differentiated: under the influence of T, cloacal gland growth was smaller and this treatment did not activate masculine sexual behavior in females. The optical density of the medial preoptic nucleus stained with thionein blue (Nissl stain) was decreased by castration and increased by T in both males and females. This suggests that T has a profound effect on the synthesis of proteins in the POM and, since POM is a critical site in the activation by T of masculine sexual behavior, these induced proteins are, in all probability, a part of the causal chain of biochemical events giving rise to copulatory behavior. The treatment with T had, by contrast, no clear effect on the number of binding sites and on the affinity of the alpha 2-adrenergic receptors in the POM. Therefore, if the noradrenergic transmission is involved in the induction by T of copulatory behavior, it is probably acting either through the alteration of another adrenergic receptor subtype or through the modulation of the baseline levels or turnover of the transmitter itself.
