Entire plants were regenerated from nodes explants of Jatropha curcas L. following a procedure of bud aggregate induction on MS (Murashige and Skoog) medium supplemented with 25 mg.l-1 citric acid, 12.2 mg.l-1 adenine sulfate, 15 mg.l-1 L-arginine, 2.46 µM IBA (indole-3-butyric acid), 30 g.l-1 sucrose and 7 g.l-1 of agar, and enriched with different balances of BA (benzyladenine) and L glutamine. The histological studies performed on aggregates showed that the buds result from both the development of axillary buds and adventitious budding starting from underlying tissues of the explant. The culture medium containing 6.65 µM BA and 25 mg.l 1 L-glutamine gave the best results with an average of 64 buds per aggregate after three weeks for all accessions tested. The buds developed into shoots when placed in a MS medium supplemented with 2.21 µM BA, 5.70 µM IAA (indole-3-acetic acid) and 15 mg.l-1 L arginine. These shoots were isolated and then rooted in MS containing 2.46 µM of IBA, 2% sucrose and 0.7% agar. The entire process took 13 weeks with a 98% survival rate in terms of plantlets acclimatization. We obtained a multiplication rate of 13 buds per explant and per subculture which is the double of those obtained in other recent works based on the micropropagation of J. curcas from node explants. This protocol is economically more profitable. [less ▲]

The aim of this study was to describe the embryos abortion process and the inheritance of the embryos abortion trait in Phaseolus vulgaris plants deficient in seed development. These plants were isolated within the second generation of an ethyl methanesulfonate (EMS) TILLING population of P. vulgaris cv. 'BAT93'. Mutant embryos show abnormalities mainly in suspensors, shoot apical meristem (SAM) and cotylédons from the globular to the cotyledon stages and abort before maturity compared to those observed in wild-types samples. Mutant embryos show also hyperhydricity and contain low amount of chlorophyll. Genetic analyses of F1, F2 and F3 populations from the crosses carried out between the mutagenized plants with aborting embryos and the wild-type plants indicated that the embryo abortion phenotype is maternally inherited and controlled by a single recessive gene. These Phaseolus mutant plants with aborting embryos constitute a valuable material for plant embryogenesis studies. [less ▲]

The extension of Jatropha curcas L. cultivation as a biofuel feedstock species requires the distribution of a very large number of plants to the producers in a very short period of time. These plants have to be able to give a high oil yield and be morphologically and phenotypically homogeneous to facilitate cultural operations. If high oil content can be obtained by varietal selection, the achievement of homogeneous material passes by the in vitro propagation. Various methods of mass production of plant material by axenic culture have been published. This study reviews the protocols published for in vitro propagation of J. curcas and discusses their applicability to an industrial scale. [less ▲]

Gossypium australe F. Muell (2n + 2x = 26) is a wild perennial species possessing agronomic useful traits that would be interesting to introgress into G. hirsutum L. (2n = 4x = 52), the main cultivated cotton species. To isolate monosomic alien addition lines (MAALs) of G. australe in G. hirsutum, the [2(G. hirsutum X G. australe) X G. hirsutum] pentaploid (2n = 5x = 65) was backcrossed as male parent to G. hirsutum. Analysis of 42 BC1 plants and seven alien addition lines, already available, with 150 SSR markers developed from G. hirsutum revealed a cross-species amplification rate of 100% and a polymorphism rate of 56%. Eighty polymorphic SSR markers generated 87 G. australe-specific loci that have been assigned by a hierarchical cluster analysis to 13 linkage groups corresponding to the 13 chromosomes of G. australe. Analysis by SSR markers and genomic in situ hybridization of the self-progeny of disomic alien addition lines, backcross progeny of the pentaploid, allowed the isolation of five new MAALs. [less ▲]

Hybrid embryos resulting from crosses between Phaseolus species often fail to reach maturity and some combinations frequently abort at early developmental stages. The genetic or molecular basis for these consistent developmental defects is at present not clear. However, an extremely complex genetic system, thought to be caused by major epigenetic changes associated with gene expression changes, has been shown to be active in plant species. We have investigated DNA methylation in two interspecific hybrids, Phaseolus vulgaris X Phaseolus coccineus and it reciprocal crosses, using methylation sensitive amplification polymorphism (MSAP). The potential use of MSAP for detecting methylation variation during embryogenesis in interspecific hybrids is discussed. Significant differences in the DNA methylation patterns were observed in abortive (interpecific hybrids) and non abortive (parental) genotypes. Taken together, our results strongly suggest that generalized alterations in DNA methylation profiles could play a causative role in early interspecific embryo abortion in vivo. A considerable change in the methylation pattern during embryogenesis could be involved in the disruption of the regulation or maintenance of the embryogenesis process of Phaseolus interspecific hybrids. The results also support the earlier hypothesis that DNA methylation is critical for the regulation of plant embryogenesis and gene expression. [less ▲]

In order to improve the in vitro multiplication rate (number of shoots/explant/subculture) of Jatropha curcas L. axillary nodes taken from young plants of two accessions of this species (originating from Cameroon and Senegal) have been cultivated for three weeks on a MS medium supplemented with 8,87 µM BAP, 4.92 µM IBA, and 30 g.l-1 sucrose at pH 5.7±0.1, and solidified with 0.7% agar. The shoots obtained from each original explant were then transferred to proliferation media (PM) consisting of MS medium supplemented with 2.21 to 8.9 µM BA or 2.21 to 8.9 µM kinetin in combination with 2.46 µM IBA. Each combination was completed with 33.12 µM adenine sulfate, 82.92 µM of glutamine and 30 g.l-1 sucrose. The best multiplication rate was obtained for the PM medium containing 6.65 µM BA and 2.46 µM AIB. On this medium 42.72±3.22 and 38.15±4.7 shoots/explant were obtained respectively for the accessions from Cameroon and Senegal after 6 weeks of culture, and the mean multiplication rates were 8.27±1.27 (accession from Cameroon) and 7.89±1.13 (accession from Senegal) shoots per explant during the 7 following subcultures (3 weeks/subculture). This medium was also the one that allowed the best overall growth in shoot height. Leafy shoots obtained have been rooted in a medium containing half of the major mineral components of MS supplemented with 5.7 µM IBA, 1.5% sucrose and solidified with 0.7% agar, then acclimated with a survival rate of 97%. These results allow considering the establishment of industrial units of plantlet multiplication from elite clones of J. curcas. [less ▲]

This study aimed at classifying nineteen veteran lime trees (Tilia cordata Mill., T. platyphyllos scop. and T. x europaea L.) in their development. Tilia spp. was chosen because it is long lived and widely present in Belgium. A classification in five steps (A to E) is proposed, based on crown and trunk status, quantity of decaying wood and mean annual girth increment (MAI) as an additionnal criterion. We tested the relationship between veteran tree development and MAI. Threshold values were determined to help distinguish steps A to E. The present work was carried out to establish rapid in vitro propagation of the indigenous oilseed Citrullus lanatus. Efficiency of 3 protocols for seed decontamination was investigated. High level of seed sterilization was obtained after removing seed coat and soaking the seeds in 1.6 % sodium hypochlorite with a drop of Tween 20. Shoot tips, single node and cotyledon explants of 3 morphotypes have been screened for adventitious shoot formation in tissue culture. Best response in termis of multiple shoot induction was obtained from cotyledon proximal part with hypocotyl segmenet on Murashige and Skoog medium supplemented with 1 mg/l 6-Benzylaminopurine, 30 g/l sucrose and 8 g/l agar. After 3 weeks of culture, 90% of cotyledon proximal parts induced shoot. An average of 12.6 shoots per explants and a mean shoot length of 8 mm were obtained after multiplication stage. Shoot induction appeared to be strongly influenced by genotype and explant type. The percentage of shoot induction from cotyledon proximal parts ranged from 23.3% to 64.0% according to the genotype. Efficient rooting was achieved on half-solid MS medium containing 0.1 mg/l 1-Naphthaleneactic acid or without growth regulator. Acclimatised plantlets were transferred to greenhouse where they grew and flowered like seeded plantlets. [less ▲]
