A performant method for the simultaneous quantification of daidzein, genistein, formononetin and biochanin A in forages using an UPLC®-MS/MS was developed and fully validated. The ultrasound-assisted extraction and enzymatic hydrolysis used in the sample preparation step were optimized using the Box-Behnken experimental design. The optimal extraction conditions used for a representative mix of forage plants were 80°C, 10 min and 55% methanol, and for hydrolysis they were 20°C, 18 h and pH=6. The chromatographic separation was achieved using an Acquity® UPLC® HSS T3 column, with a water/methanol linear gradient containing 0.01% of formic acid at a 0.55 mL min-1 flow rate. The four isoflavones were detected by ESI mass spectrometry in positive ion MRM mode. The method allows high throughput analyses of samples and showed an adequate linear regression model for all isoflavones over a range from 5 to 125 ng mL-1. There was good intra- and inter-day precision (≤ 8.2% and ≤ 7.6%) and accuracy (≤ 11.4% and ≤ 7.1%). The recovery rates were in an acceptable range of 70-120%, except for biochanin A, where the rate was about 50%. Good method repeatability was also observed, and there was no matrix effect or carry-over problem. The sample extracts were stable for at least 6 days of storage at -21°C and 6°C. The method proved to be sensitive, precise and accurate for discriminating a wide variety of forages likely to be grazed by ruminants according to their isoflavone content and to observe the impact storage process on isoflavone content in forages. This paper provides an update and comprehensive review of the analytical methods used for quantifying isoflavones and their metabolites in cow’s milk. Isoflavones are secondary plant metabolites that are similar to 17 β-estradiol in chemical structure. They form one of the most common categories of phytoestrogens. Numerous health benefits have been attributed to isoflavones, but many of these compounds are also considered to be endocrine disruptors, with adverse effects on health. These contradictory trends offer an attractive prospect for future research, and therefore, sensitive and reliable analytical methods are required to clarify various issues about isoflavones. For this review, a structured methodology was used to select 26 relevant articles published between 2005 and 2015 from the Scopus and CAB Abstract databases. The review discusses individual steps of the analytical procedures described in these articles, including sample preparation, instrumental analysis and validation. The most commonly used analytical procedure is sample preparation involving liquid-liquid extraction and an enzymatic hydrolysis step followed by liquid chromatography with mass spectrometry analysis. Currently, however, there is no standardized procedure for the sample preparation and analysis of isoflavones in milk. [less ▲]

Isoflavones constitute one of the most common categories of nonsteroidal estrogen-like substances belonging to the broad group of phytoestrogens. The highest concentrations in the plant kingdom are found in the Fabaceae family. They have become a focus of research because of their estrogenic or anti-estrogenic effect and potential impact on human health. In recent years, several studies have focused on the impact of biotic and abiotic factors and farming management on the isoflavone concentration in plants and their impact on the composition of cow’s milk. Nevertheless, knowledge about the effect of the ensiling process on isoflavone concentration remains limited. The objective of this work was to study the evolution of the concentrations of four compounds (daidzein, formononetin, genistein and biochanin A) in red clover (Trifolium pratense L.) ensiled at harvest. The impact of the field-drying process was also assessed. The plant material was ensiled in laboratory scale silos using vacuum-packed plastic bags and stored over a 6-month period. The quality of the silages was checked throughout the experiment by determining the pH, lactic acid concentration, volatile fatty acids, crude proteins, cellulose and other chemical characteristics. The isoflavone concentration in fresh plant material was 2050, 1766, 306 and 127 µg/g DM for formononetin, biochanin A, genistein and daidzein, respectively. After 4 days of drying in the field, no significant change in isoflavone concentration was found, except for daidzein, which increased twofold. The laboratory-scale silos experiment, however, showed a decrease in isoflavone concentration during the first 2 weeks, followed by stabilization over the 5 remaining months. The concentrations fell by 26, 39, 66 and 73% for daidzein, genistein, biochanin A and formononetin, respectively. Animals fed with silage would therefore absorb fewer isoflavones than those fed with fresh plant material or hay. [less ▲]

Dans le cadre d’un projet de valorisation des prairies, intitulé « PhytoHealth », une méthode d’analyse quantitative constituée d’une étape d’extraction à l’aide d’un bain à ultrasons, suivi d’une hydrolyse enzymatique et d’une analyse par UPLC®-MS/MS a été développée pour assurer le dosage de phyto-œstrogènes dans les fourrages. Pour alléger l’optimisation de la préparation des échantillons, la méthodologie des surfaces de réponses a été utilisée. Un plan d’expérience de Box-Behnken a été appliqué aux étapes d’extraction et d’hydrolyse. Parmi les différents facteurs pris en compte, les impacts de la nature du solvant utilisé lors de l’extraction ainsi que la température et le pH lors de l’hydrolyse, ont clairement pu être mis en évidence. L’ensemble des données générées a également permis de définir des conditions optimales d’extraction et d’hydrolyse pour chacun des composés. Ces conditions n’étant pas identiques pour chacun d’entre eux, un compromis a été trouvé afin de pouvoir analyser l’ensemble de ces composés simultanément. L’ensemble de la méthode a ensuite été validée selon les recommandations de l’EMA/CVMP/VICH/463202/2009, pour confirmer sa fiabilité et robustesse. Cette dernière, combinée au model d’ensilage en sacs sous vide, a permis pour la première fois de suivre la cinétique d’évolution de 4 phyto-œstrogènes (daidzéine, génistéine, formononétine et biochanin A) durant une période d’ensilage. The term ‘phytoestrogen’ is used to define a wide variety of nonsteroidal compounds that occur naturally in many plants. When they are absorbed by cows, these polyphenolic compounds undergo biotransformation by specific bacteria present in the gastrointestinal tract. The resulting metabolites are absorbed in the organism and some are excreted in milk. The impact of these compounds on human health divides opinion. Some scientists believe that they might have adverse health effects. Others believe the opposite, especially with regard to their microbial metabolites, such as equol. Whatever one's view on the subject, scientists need accurate, sensitive and rapid analytical methods in order to continue the research on clarifying the phytoestrogen issue. Cow's milk is an animal product that is common in the human diet, and it is therefore important to evaluate its content of phytoestrogen metabolites. In order to study the human intake of equol via commercial milk, an analytical method was developed and validated following EMA/CVMP/VICH/463202/2009 guidelines. Enzymatic hydrolysis was used to release the equol. It was then extracted using double liquid/liquid extraction and analyzed using UPLC®-MS/MS, with an analysis runtime of only 5 min. This analytical method produced a linear calibration curve with a high correlation coefficient (R2≥0.996) between 5 and 1,000 ng mL−1. Good intra- and inter-day precision (RSDs≤5.3% and ≤5.2%, respectively) and accuracy (bias≤8.6%) were achieved. The recovery rate differed slightly among the different types of milk, ranging between 60.6±1.09% and 82.3±5.21%. Good method repeatability was observed (RSDs<15%). There was neither a matrix effect nor a carry-over effect, and the sample extracts were stable during storage for at least 7 days at −21°C and 5°C. In order to apply the proposed method and obtain an initial estimate of equol concentration in cow’s milk in Belgium, 44 samples of various brands found in several supermarkets and 5 raw milk samples from Walloon farms were analyzed. Equol was found in all the samples analyzed, with a concentration ranging from 10 to 50 ng mL−1 for conventional milk and from 70 to 130 ng mL−1 for organic milk. These results were compared with other European studies and the same trend was observed. The results obtained show the specificity, sensitivity and precision of this method for analyzing oestrogenic metabolite-equol in cow’s milk. This study should be extended to include a greater number of samples and be implemented over a longer period to see if there is a correlation between livestock practice and/or sample origins with the equol content in milk. The addition of other phytoestrogen compounds with this method, using UPLC®-MS/MS technology, could also lead to more reliable monitoring of these compounds in dairy production. [less ▲]

Cow’s milk can be used as a potential source of equol in the human diet. In order to study human intake, however, it is necessary to develop a reliable and sensitive analytical method. This paper reports on the validation of an analytical method using ultra-performance liquid chromatography coupled with a tandem mass spectrometry detector to quantify the equol in commercial milks (raw, whole, semi-skimmed, and skimmed milk). The equol was initially released using enzymatic hydrolysis, and it was then extracted using a double liquid/liquid extraction. The analytical method produced a linear calibration curve with a high correlation coefficient (R 2 ≥ 0.996) between 5 and 1,000 ng.mL−1. Good intra- and inter-day precision (≤5.3% and ≤ 5.2%, respectively) and accuracy (≤8.6%) were achieved. The recovery rate differed slightly among the different types of milk, ranging between 60.6 ± 1.09% and 82.3 ± 5.21%. Good method repeatability was observed (<15%). There was neither matrix effect nor carry-over effect, and the sample extracts were stable for at least 7 days of storage at −21 °C and 5 °C. The method proved to be specific, sensitive, precise, and accurate and was used for the first time to quantify the equol content in Belgian commercial cow’s milk. In all the samples analyzed, equol was present at a concentration ≥10 ng.mL−1 and had a significantly higher content in organic than in conventional milk. The study also found that the mean concentrations of equol were similar for each type of commercial conventional cow’s milk. [less ▲]

In a project named PhytoHealth, the development of analytical methods for studying the impact of phytoestrogens rich diet on the «health value» of animal products is in progress. Despite the ambiguous image that have phytoestrogens, some of their metabolites seemed to have potentially beneficial effects to human health. In a first time, a microbial metabolite (equol) was selected and its metabolism in dairy cow is studied. A first method using the UPLC®-MS/MS technology has been validated and has achieved a screening of equol content in milk consumed in Wallonia. Equol was found in all milk samples analyzed and a significant difference between farming methods has been highlighted. A second analytical method to quantify the equol precursors was then developed and a study of forage plants consumed by Belgian dairy cows will be conducted to select the richest fodder varieties. Other methods will be developed to better understand the metabolism in the cow and estimate the impact of enriched milk on human health. An original approach involving the use of minipigs will be considered. In the end, creating of a differentiated quality animal products chain could be interesting for the consumer, but also recoverable for the producer. [less ▲]

Kashin-Beck disease (KBD) is an endemic and chronic osteochondropathy. This disease principally occurs in the Tibet Autonomous Region and in several provinces of the People’s Republic of China. The etiology of the disease remains obscure although environmental factors are assumed to be involved. Diet, in particular, differentiates the rural community, affected by KBD, from the other communities (nomads and city-dwellers), who remain unaffected. In anticipation of a nutrition survey, this study aimed to measure the mineral content (Ca, P, Mg, Fe, Zn, Mn, Cu, Ni, Se, Al, Sr, Mo, Cd, As, Pb, Hg, Cr, and Co) of eight Tibetan staple foods and to compare the results against two food composition tables (FCTs). Foods were sampled in twenty households selected from both an endemic and a non-endemic area of rural Tibet. Ten minerals involved in bone metabolism were measured using atomic and molecular spectrometric methods. Results revealed that a very limited number of food/constituent pairs showed a variation in mineral composition during a single year of testing for a given region. In addition, results showed significant differences in mineral content between the endemic and the non-endemic area, especially for wheat flour. Following our analysis of the mineral content of the Tibetan food samples, results were statistically compared with similar foods listed in two food composition tables: the USDA National Nutrient Database (USDA Food Search for Windows, Version 1.0, database version SR23), and the China Food Composition Table (book 1, 2nd edition). More than 50 to 60% of p-values < 0.05 were highlighted, suggesting the inappropriateness of using FCTs as a reference for nutrition surveys in rural Tibet, and emphasizing the need for analysis of traditional foods. Differences were found to be more or less marked depending on the element considered, and calcium content seemed to show the greatest difference. Although it is obviously too early for definite conclusions to be reached (insufficient number of samples by food and insufficient number of foods analyzed), it seems that the present pilot-study indicates significant discrepancies between measured and tabulated values of the mineral content of certain foods. A more complete survey would therefore seem mandatory. [less ▲]

Knowing the contents of minerals in milk like Ca or Na could be interesting to improve the nutritional quality of milk and to assess the animal health status. This study had two aims: 1) development of mid-infrared equations for mineral contents in milk by using an approach combining multiple countries, breeds, and production systems and 2) study of the genetic variability of these traits in the Walloon Holstein dairy cattle. Samples included in the calibration set were collected in Belgium, Luxembourg and France over 5 years. The calibration set included at least 400 samples analyzed by coupled plasma atomic emission spectrometry to quantify the contents of Na, Ca, Mg, P and K. The calibration coefficient of determination ranged between 0.69 for K and 0.93 for Na. The standard error of cross-validation was 63.35, 49.24, 64.33, 7.04, and 93.22 mg/kg of milk for Na, Ca, P, Mg and K. From these results, the quantification of milk minerals by mid-infrared is feasible. These equations were applied to more than 140,000 spectral records collected from 43,797 first parity Holstein cows in 1,233 herds. The variance components were estimated using Gibbs Sampling using single trait random regression models derived from the one used for the Walloon genetic evaluation of milk production traits. First results gave a daily heritability of 0.26 for Na, 0.45 for Ca, 0.48 for P, 0.46 for Mg, and 0.41 for K. Moderate negative genetic correlations were found between Na and the other studied traits. The highest correlation (0.69) was observed between P and Mg. These results confirmed the genetic variability of these traits. Further studies will be conducted to study the relationship between these traits and other traits (e.g., production, health).
