Eyeworms were retrieved from the left eyes of two dogs presented for unilateral ocular discharge in Belgium. Morphological and molecular identification were performed and the parasites were identified as Thelazia callipaeda. The history suggested that the infection had been acquired in South-Western France and Southern Italy where the disease has been observed regularly for the last 6 and 12 years, respectively. In these two regions, the disease is considered endemic and spreading. To the best of our knowledge, this is the first complete case report of canine thelaziosis in Belgium. The risk of introduction of the parasite in Northern Europe and particularly in Belgium is discussed. [less ▲]

The objective of this prospective study was to investigate the success rate, practicality and complications of a new perilimbal pocket technique for the replacement of prolapsed nictitans gland in 30 dogs (44 eyes). A first incision was made in the bulbar conjunctiva, 2-3 mm from and parallel to the infero-nasal limbus, a second incision on the bulbar aspect of the nictitating membrane (NM), 2-3 mm parallel to the free edge. The gland was returned to its normal position by suturing the subconjunctival tissues of the NM to the episcleral tissues, using four to six interrupted horizontal mattress sutures. The English bulldog, Neapolitan mastiff, great dane and American cocker spaniel were commonly presented. Nictitans gland prolapse occurred prior to one year of age in 83.3 per cent of dogs, and unilaterally in 15 patients. The procedure was easy to perform, and had a 90.9 per cent success rate, with minimal complications. The median duration of follow-up, conducted by ophthalmic examination or telephone contact with the owners, was 21.5 months. Tear production and ocular health were not affected in 17 eyes with at least six months follow-up. There was a statistically significant increase between preoperative and postoperative Schirmer tear test-1 measurements. [less ▲]

The cataract corresponds to the total or partial opacification of the lens of the eye preventing the passage of the light. At present, the surgery is the only effective treatment to overcome the cataract. The surgical intervention consists in removing the cloudy lens and to replace it by an artificial intraocular lens (IOL). The in vivo implantation of these synthetic lenses involves the evaluation of several factors as their physico-chemical properties, their capacities to interact with lens epithelial cells and proteins, as well as their biocompatibility. During a previous study, we demonstrated major differences concerning the tackiness (atomic force microscopy), the cellular adhesion and the protein adsorption of various polymer disks intended for the manufacturing of intraocular lenses. The aim of this work was to correlate a histological analysis to a mass spectrometry imaging analysis performed on the same sample. To estimate the biocompatibility of the biomaterials, an animal testing was realized in rabbits. The various polymers were implanted subcutaneously. After one month, the 2 cm x 3 cm pieces of rabbit skin and underlying muscle with a 2 cm thickness were removed, fixed with formaldehyde 10% during six days, treated for the paraffin inclusion and stored at room temperature until use. Slices of 5 µm thickness were performed using a microtome. Paraffin was removed and tissue sections were washed in graded ethanol baths. The slices were then stained with the hematoxylin and eosin dyes. The analysis of stained sections showed different histo-morphological features according to the implanted polymer. For MALDI MSI purposes, on tissue protein digestion was performed using trypsin (1) and the MALDI matrix (α-cyanohydroxycinnamic acid, 5 mg/mL in ACN/0.2% TFA 70:30) was deposited using an ImagePrep automated sprayer (Bruker Daltonics, Bremen, Germany). Experiments were carried out using an UltraFlex II TOF/TOF mass spectrometer (Bruker Daltonics, Bremen, Germany). MALDI imaging can show the detection of different proteomic profiles according to the tested biomaterials, which may be considered as biocompatibility markers. The MALDI images of these markers are then correlated with the histo-morphological profiles. Consequently, mass spectrometry imaging can become a powerful tool in the evaluation of the biocompatibility of artificial implants in biomedical application. [less ▲]

REASONS FOR PERFORMING STUDY: There is a clinical impression that tetracaine hydrochloride (THCl) eyedrops is a suitable topical anaesthetic in horses. OBJECTIVE: To determine the duration of corneal anaesthesia following instillation of multiple doses and 2 concentrations of THCl in 10 healthy horses. METHODS: The corneal touch threshold (CTT) was determined, in both eyes, before (basal CTT) and after application of one drop of 0.5% THCl, 2 drops at a 1 min interval of 0.5% THCl or one drop of 1% THCl. CTT was measured in mm every 5 min until complete recovery of the basal CTT. Treatments were separated by an interval of at least one week. RESULTS: Corneal sensitivity was significantly reduced from baseline values for 30, 60 and 50 min after application of one drop of 0.5% THCl, 2 drops of 0.5% THCl and one drop of 1% THCl, respectively. Mean maximal anaesthetic effects, corresponding to a CTT of 0 mm, lasted 5.5, 16 and 15.25 min and maximal anaesthetic effect was present in 55, 90 and 80% of eyes, 5 min after application of one drop of 0.5% THCl, 2 drops of 0.5% THCl and one drop of 1% THCl, respectively. CONCLUSIONS: The application of a second drop or the use of more concentrated eyedrops significantly increases duration of both anaesthesia and maximal anaesthetic effect. POTENTIAL RELEVANCE: Duration of corneal anaesthesia following tetracaine instillation was established enabling a better use when performing diagnostic and therapeutic procedures. Comparison of tetracaine with other ocular anaesthetics needs to be published in the future. [less ▲]

REASONS FOR PERFORMING STUDY: Tetracaine hydrochloride (THCl) has been reported to cause irritation in dogs. In man, some topical anaesthetics have been shown to disrupt the tear film. Tear break-up time (TBUT) is a useful test allowing an assessment of the quality of the precorneal tear film. Only one TBUT value has been reported in horses with no information on the technique used. OBJECTIVES: To provide a method for performing the TBUT in horses and to report any side effects of a single application of THCl in clinically normal horses, particularly on the stability of the tear film. METHODS: In Study 1, one drop of 0.5 or 1% THCl was applied to one eye of 20 horses divided in 2 groups. Treated eyes were assessed for the development of side effects 2.5 and 5 min after treatment. In Study 2, the TBUT was measured in both eyes of 2 groups of 10 horses, before and 2.5 and 5 min after, instillation of one drop of either 0.5 or 1% THCl. RESULTS: No animals developed any ocular side effect after instillation. Basal TBUT was 8.3±1.3 s. TBUT decreased from baseline 5 and 2.5 min after application of one drop of 0.5% THCl and one drop of 1% THCl, respectively. CONCLUSIONS: A technique to measure the TBUT in healthy horses is described and normal range values that could be used as a reference were obtained. POTENTIAL RELEVANCE: THCl is well tolerated in horses but lowers the TBUT. [less ▲]

Several molecules available in equine medicine provide profound sedation and good analgesia when performing diagnostic or surgical procedures. They differ slightly in degree of action, as well in time as in depth. The aim of this study was to evaluate the depth of sedation and quality of performing a standardized procedure after injection of an 2-agonist alone or associated with butorphanol. Horses presented to the Equine Clinic of Liege University from December 2007 to June 2008 for an endoscopic, dental, radiological or ophthalmological examination of which owner and clinician agreed to include the horse in the study were retained. All horses (n=133) were sedated in a randomized double blind fashion using one of the six following protocols: detomidine (D) (0.01mg/kg IV), romifidine (R) (0.04mg/kg IV), or xylazine (X) (0.5mg/kg IV) alone or associated with butorphanol (B) (0.02mg/kg IV). Stability, head position and the reaction to a stimulus (auditive and visual) were evaluated before the start of the procedure. An overall subjective grade using a 10 cm line scale was given to evaluate sedation depth (ranging from no effect to maximal depth on line) and the quality of the performed procedure (ranging from not realised to ideal examination). For each specific procedure, the same clinician experienced in his particular field evaluated the parameters. Data were related by a variance analysis using SAS system and considered significant if P<0.01. The used protocols significantly influenced both sedation depth and examination quality. Sedation depth increased significantly when B was added in all the three 2-agonists used (mean ± SD): D: 6.46±0.33; D+B: 7.06±0.37; R: 4.93±0.46; R+B: 7.04±0.45; X: 4.94±0.43; X+B: 7.75±0.40. Detomidine provided a significantly better quality of the examination compared to romifidine and xylazine. The addiction of butorphanol significantly decreased the examination quality when added to detomidine and increased the quality when combined with romifidine and xylazine. Mean (± SD) of the quality of performed procedures: D: 8.41±0.54; D+B: 6.06±0.64; R: 6.68±0.71; R+B: 7.16±0.67; X: 4.47±0.67; X+B: 4.52±0.64. Comparison of sedation depth and quality of performed examination revealed an increase in quality of examination scores with increasing depth of sedation until a certain level. As from sedation depths with scores of 8 or more, a decrease in quality of examination was encountered. This report shows the important effect of butorphanol addition on sedation. However, increasing sedation depth was not necessarily linked to a better quality of procedure, notably due to difficulties in handling strongly sedated horses for the procedures performed in this study. X-ray microtomography coupled with image analysis was tested as a non-destructive alternative method for the textural characterization of the trabecular part of deer antlers (Cervus Elaphus). As gas adsorption and mercury intrusion cannot be applied on this soft and spongy material, its pore texture was, up to now, determined from histological sections that give only two-dimensional information. In this work, X-ray microtomography is used to scan entire or half pieces of antlers and three-dimensional image analysis is performed in order to assess the differences between samples collected at various antler locations. Results clearly show a porosity profile along the sample diameter. The pore size distribution is showed to be dependent on the sample original site. [less ▲]

A family of five privately owned rough collie dogs was referred for corneal lipidosis and also suffered from hypercholesterolaemia. The hypercholesterolaemia was characterised by an increase in the alpha-2 high density lipoprotein-1 band and was due to an increase in the cholesterol content of the very low density lipoprotein, low density lipoprotein and possibly the high density lipoprotein-1 fractions. A low-fat and energy-restricted diet did not reduce either total cholesterol or the corneal lipidosis. Corneal lipidosis regressed with short-chain fructo-oligosaccharide supplementation. However, the effects of short-chain fructo-oligosaccharides on total cholesterol were transient and variable. [less ▲]

OBJECTIVE: To assess morphologic and metabolic abnormalities in dogs with early left ventricular dysfunction (ELVD) induced by rapid right ventricular pacing (RRVP). ANIMALS: 7 Beagles. PROCEDURE: Plasma carnitine concentrations were measured before and after development of ELVD induced by RRVP. At the same times, transvenous endomyocardial biopsy was performed, and specimens were submitted for determination of myocardial carnitine concentrations and histologic, morphometric, and ultrastructural examination. RESULTS: In 4 dogs in which baseline plasma total carnitine concentration was normal, RRVP induced a decrease in myocardial total and free carnitine concentrations and an increase in myocardial esterified carnitine concentration. In 3 dogs in which baseline plasma total carnitine concentration was low, plasma and myocardial carnitine concentrations were unchanged after pacing. Structural changes associated with pacing included perinuclear vacuolization in 3 dogs. Morphometric analyses indicated there was a decrease in myofiber cross-sectional diameter and area following pacing. Electron microscopy revealed changes in myofibrils and mitochondria following pacing. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that moderate to severe alterations in myocyte cytoarchitecture are present in dogs with ELVD induced by RRVP and that in dogs with normal plasma carnitine concentrations, myocardial carnitine deficiency may be a biochemical marker of ELVD. Results also indicated that transvenous endomyocardial biopsy can be used to evaluate biochemical and structural myocardial changes in dogs with cardiac disease. [less ▲]

OBJECTIVE: To add objective measurements of the characteristics of evoked injury potentials (EIP) and their relations to clinical severity in dogs with thoracolumbar spinal cord damage. ANIMALS: 25 dogs with naturally acquired spinal cord compression attributable to disk extrusion or vertebral fracture at the level of the thoracolumbar junction and with various degrees of paresis/paralysis. PROCEDURE: Spinal cord potentials evoked by tibial nerve stimulation were recorded every 5 to 10 mm at the lamina level in the vicinity of the cord compression. This allowed an EIP to be recorded even in the least handicapped dogs. A computer model yielded information about the waveform changes of the EIP in the vicinity of conduction blocks. RESULTS: The EIP waveform changed from biphasic to monophasic a short distance caudad to the location of spinal cord compression. Location of a maximal conduction block was measured in relation to position of the electrodes recording this waveform change. The distance between the assumed conduction block and the actual spinal cord compression was larger in the most affected dogs. The amplitude of the EIP was not related to severity of the clinical picture; however, the proximity of the recording electrode to the spine influenced the amplitude and the waveform of the EIP. CONCLUSION AND CLINICAL RELEVANCE: Change in the EIP waveform from biphasic to monophasic makes it possible to estimate the conduction block location along the spinal cord. A large distance between the assumed conduction block and site of actual cord compression could be an objective argument to confirm severity of a lesion. [less ▲]

The aim of this study was to determine cardiac contractility using indices derived from cardiac catheterisation in conscious healthy dogs during dobutamine infusion. Eight dogs were studied. An ECG was recorded together with left ventricular pressure and volume which were measured using a conductance catheter with an integrated microtip pressure sensor. Eight indices of left ventricular systolic performance were derived from these records. Measurements were realised under basal conditions and during an incremental dobutamine challenge. The maximal rate of rise in ventricular pressure (max dP/dt), max dP/dt divided by the developed pressure and the mean systolic ejection rate were the most sensitive indices to detect dobutamine induced changes in contractility with maximal percentage changes of 122+/-11 per cent, 130+/-7 per cent and 102+/-24 per cent respectively. Ejection fraction increased significantly during dobutamine infusion (maximal percentage change of 43+/-9 per cent) whereas the pre-ejection period (PEP) and the left ventricular ejection time (LVET) decreased significantly (maximal percentage change of -41+/-2 per cent and -28+/-3 per cent respectively). All these six indices were significantly correlated with each other. Conversely, the ratio PEP/LVET and the LVET corrected for heart rate dependency showed a maximal percentage change of only -10+/-1 per cent and -16+/-7 per cent, respectively, during the dobutamine infusion and were not significantly correlated with the other contractility indices. This study demonstrated the feasibility of the conductance method to determine cardiac contractility in conscious healthy dogs submitted to a pharmacological stress testing and provides control values for eight indices of left ventricular contractility during dobutamine infusion at increasing dosages.
