Human noroviruses (HuNoV) are the leading cause of acute nonbacterial gastroenteritis in humans and can be transmitted either by person-to-person contact or by consumption of contaminated food. Knowledge of an efficient disinfection for both hands and food contact surfaces is helpful for the food sector and provides precious information for public health. The aim of this study was to evaluate the effect of seven disinfectants belonging to different groups of biocides (alcohol, halogen, oxidizing agents, quaternary ammonium compounds, aldehyde and biguanide) on infectious viral titre and on genomic copy number. Due to the absence of a cell culture system for HuNoV, two HuNoV surrogates such as murine norovirus (MNV) and feline calicivirus (FCV), were used and the tests were performed in suspension, on gloves and on stainless steel discs. When, as criteria of efficacy, a log reduction > 3 of the infectious viral titre on both surrogates and in the three tests is used, the most efficacious disinfectants in this study appear to be biocidal products B, C and D, representing the halogens, the oxidizing agents group and a mix of QAC, alcohol and aldehyde, respectively. In addition, these three disinfectants also elicited a significant effect on genomic copy number for both surrogate viruses and in all three tests. The results of this study demonstrate that a halogen compound, oxidizing agents and a mix of QAC, alcohol and aldehyde are advisable for HuNoV disinfection of either potentially contaminated surfaces or materials in contact with foodstuffs. The callipyge phenotype is an ovine muscular hypertrophy characterized by polar overdominance: only heterozygous +Mat/CLPGPat animals receiving the CLPG mutation from their father express the phenotype. +Mat/CLPGPat animals are characterized by postnatal, ectopic expression of Delta-like 1 homologue (DLK1) and Paternally expressed gene 11/Retrotransposon-like 1 (PEG11/RTL1) proteins in skeletal muscle. We showed previously in transgenic mice that ectopic expression of DLK1 alone induces a muscular hypertrophy, hence demonstrating a role for DLK1 in determining the callipyge hypertrophy. We herein describe newly generated transgenic mice that ectopically express PEG11 in skeletal muscle, and show that they also exhibit a muscular hypertrophy phenotype. Our data suggest that both DLK1 and PEG11 act together in causing the muscular hypertrophy of callipyge sheep. [less ▲]

The Kabyle goat breed represents 10.52% of the total Algerian market (3.8 million heads). A survey of 69 farmers has been carried out in order to characterize it and its breeding in the District of Chemini. The questions focused on household agricultural activities, including breeding of goats, cattle, sheep, rabbits, chicken, turkeys, honeybees and the production of olive oil and figs. The goat morpho-biometric characterization was based on 18 corporal measurements. The Hierarchical Cluster Analysis (HCA) of the farm structures defined four groups of farms, variance between groups accounting for 55.7% of total variability. The average number of goats in groups 1 to 4 was 7.2±2.8, 11.1±3.5, 22.3±1.4 and 3.4±1.0, respectively. Group 3 (n=6), showed the greatest number; it consists of older farmers (67 years old or over) not practicing arboriculture. They also have the largest numbers of sheep (48.67), rabbits (50.83), chicken (48.33) and turkeys (42). Group 4 (n=9), with the lowest number of goats, was the group of young farmers (39 years old or less), more dedicated to cattle breeding (~52 heads) and the production of olive oil (~207 trees) and figs (~47 trees). The 18 morpho-biometric variables were significantly higher in males than in females (p<0.05). The Kabyle goat is small (male: 68.23±0.97cm and female: 65.41±0.55cm) with long hair (male: 12.24±0.51cm, and female: 9.51±0.29cm). Its ears are drooping, its convex profile has a slightly pronounced nasal split and its dress color ranges from dark brown to black. This adapted genetic resource should be key in the development of a local production, based on a strong commitment of farmers inside a breed association, for the production of specimens corresponding to a standard, to be determined collectively. OBJECTIVES: To describe the results of a prevention campaign in terms of participation and pet health status and to identify opportunities to improve preventive medicine in cats and dogs. METHODS:An awareness campaign was designed to highlight the role of veterinarians and emphasise the benefits of a veterinary visit. Owners were invited to make an appointment for a free pet health check in a voluntarily participating veterinary clinic. Observations recorded by the veterinarians were entered in a database and subsequently analysed using simple descriptive statistics. RESULTS:A total of 5305 completed health check forms were analysed. The percentages of overweight and obese dogs and cats were 34 and 36%, respectively; this was the most common finding, followed by dental calculus (31% in dogs, 21% in cats). In total 67% of cats did not undergo flea control and 59% were not vaccinated. CLINICAL SIGNIFICANCE: Opportunities for increased quality of care are numerous given the high percentage of intact, unvaccinated or non-permanently identified pets and the low level of worm and flea control. Animal health should benefit from preventive measures, and improved management can be undertaken after early detection of diseases. [less ▲]

Cyprinid herpesvirus 3 (CyHV 3) is causing severe economic losses worldwide in common and koi carp industries, and a safe and efficacious attenuated vaccine compatible with mass vaccination is needed. We produced single deleted recombinants using prokaryotic mutagenesis. When producing a recombinant lacking open reading frame 134 (ORF134), we unexpectedly obtained a clone with additional deletion of ORF56 and ORF57. This triple deleted recombinant replicated efficiently in vitro and expressed an in vivo safety/efficacy profile compatible with use as an attenuated vaccine. To determine the role of the double ORF56-57 deletion in the phenotype and to improve further the quality of the vaccine candidate, a series of deleted recombinants was produced and tested in vivo. These experiments led to the selection of a double deleted recombinant lacking ORF56 and ORF57 as a vaccine candidate. The safety and efficacy of this strain were studied using an in vivo bioluminescent imaging system (IVIS), qPCR, and histopathological examination, which demonstrated that it enters fish via skin infection similar to the wild type strain. However, compared to the parental wild type strain, the vaccine candidate replicated at lower levels and spread less efficiently to secondary sites of infection. Transmission experiments allowing water contamination with or without additional physical contact between fish demonstrated that the vaccine candidate has a reduced ability to spread from vaccinated fish to naïve sentinel cohabitants. Finally, IVIS analyses demonstrated that the vaccine candidate induces a protective mucosal immune response at the portal of entry. Thus, the present study is the first to report the rational development of a recombinant attenuated vaccine against CyHV 3 for mass vaccination of carp. We also demonstrated the relevance of the CyHV 3 carp model for studying alloherpesvirus transmission and mucosal immunity in teleost skin. [less ▲]

RÉSUMÉ L’étude a été conduite pour déterminer les caractéristiques du cycle oestral de la brebis et les caractéristiques spermatiques du bélier de la race à laine Koundoum du Niger. Seize brebis et huit béliers Koundoum ont été utilisés pour cette étude. Les brebis ont été soumises à une observation biquotidienne de leur comportement sexuel en vue de détecter les oestrus, à l’aide de béliers boute-en-train. Les échantillons de sperme ont été recueillis à l’aide d’un vagin artificiel chez les béliers pendant les périodes d’oestrus des brebis. La motilité du sperme a été déterminée à l’aide d’un microscope et sa concentration à l’aide d’un spectrophotomètre. Sur les 16 brebis, 13 ont manifesté au moins une fois des comportements d’oestrus ayant une durée moyenne de 37,8±5,8 heures avec une durée de cycle oestral de 18,1±1,1 jours. La collecte de semence a été réalisée sur 4 béliers sur les 8 inclus dans le protocole. Les éjaculats collectés ont présenté un volume moyen de 1,03±0,3 ml, une motilité de 3,4, et une concentration de 1322±544 millions/ ml. La connaissance de la durée du cycle et de l’oestrus est un élément de base pour la maîtrise de la fonction sexuelle des brebis. La difficulté de collecte de la semence de béliers Koundoum devra être prise en compte dans les plans de conservation par cryogénisation de paillettes. Les informations obtenues à travers cette étude sont une base permettant de poser les premiers jalons du programme de conservation et d’amélioration de cette race. [less ▲]

OBJECTIVES: To describe the complications, short and long-term outcome and owner satisfaction of dogs with cranial cruciate ligament rupture treated with a recently described new osteotomy for the modified Maquet technique (N-MMT). MATERIALS AND METHODS: Medical records and radiographs of 82 dogs (84 stifles) were reviewed. Details regarding short-term outcome and complications were recorded from the medical records. Long-term follow-up information was obtained by telephone interview. Historical data and complications were statistically analysed. RESULTS: Major complications occurred in 34/84 stifles. Intra-operative complications occurred in 26/84 stifles, all of which were fissures or fractures of the cortical hinge. Twenty-one of these fractures or fissures were repaired with a figure-of-eight wire. The second most common major complication was late meniscal tears in 3/84 stifles. One dog sustained a complete tibial fracture. Non-displaced fracture of the cortical hinge was the most common postoperative minor complication, which occurred in 5/84 stifles. The median preoperative lameness score was 3 out of 6. Final in-hospital re-evaluation of limb function was available in 58 dogs. The median lameness score at that time was 0 out of 6. Development of osteotomy related complications was not associated with a significant change in the postoperative lameness score. CLINICAL SIGNIFICANCE: Subjectively assessed clinical outcome with the N-MMT was good to excellent in this cohort of dogs. However, a high rate of intra- and postoperative complications of the N-MMT procedure was also present in these dogs. [less ▲]

In Arabidopsis halleri, the AhHMA4 gene has an essential function in Zn/Cd hypertolerance and hyperaccumulation by mediating root to shoot translocation of metals. Constitutive high expression of AhHMA4 results from a tandem triplication and cis-activation of the promoter of all three copies. The three AhHMA4 copies possess divergent promoter sequences, but highly conserved coding sequences, and display identical expression profiles in the root and shoot vascular system. Here, we expressed an AhHMA4::GFP fusion under the control of each three A. halleri HMA4 promoters in a hma2hma4 double mutant of Arabidopsis thaliana to individually examine the function of each A. halleri AhHMA4 copy. The protein localized non-polarly at the plasma membrane of the root pericycle cells of both A. thaliana and A. halleri. The expression of each AhHMA4::GFP copy complemented the severe Zn deficiency phenotype of the hma2hma4 mutant by restoring root-to-shoot translocation of zinc. However, each copy had different impact on metal homeostasis in the A. thaliana genetic background: AhHMA4 copies 2 and 3 were more highly expressed and provided higher Zn tolerance in roots and accumulation in shoots than copy 1, whereas AhHMA4 copy 3 also increased Cd tolerance in roots. Our data suggest a certain extent of functional differentiation among the three A. halleri HMA4 copies, stemming from differences in expression levels rather than in expression profile. HMA4 is a key node of the Zn homeostasis network and small changes in expression level can have major impact on Zn allocation to root or shoot tissues. [less ▲]

This survey, conducted with 461 racehorse owners-breeders in Algeria between 2009 and 2011, investigates their perceptions, objectives and practices regarding selective breeding. Racehorse breeding is a full-time professional activity for a third of interviewees. The holdings are small-sized with 77% owning one or two mares. The regular practice of mating is here used to categorize breeders according to their degree of professionalization (38.4% professional vs. 61.6% occasional breeders). Experience in the sector was also used to classify breeders, considering as “junior” the breeders under 10 years experience (38.8%) and as “senior” those above 10 years (61.2%). More than professionalization, experience shows a significant impact on practices and objectives. Thus, experience influences breed choice (junior breeders tend to specialize while senior own both Arabian and Thoroughbreds), age at first foaling (sooner among senior breeders), information sources considered for selecting stallions (senior use more diversified sources), the importance granted to the price of mating (greater for junior breeders), the importance granted to the ranking compared to earnings (the ranking being more important to junior breeders), and the priority given to breeding (junior breeders give higher priority to a buy-race-resell activity). Finally, racehorse breeding is poorly professionalized, the only financial goal being cost coverage. Despite inappropriate practices, an interest for selection is noticed. [less ▲]
