We sought to determine whether prolactin (PRL) could influence the neutrophilic inflammation that characterizes chronic mastitis. Most of the genes encoding inflammatory proteins depend on the nuclear factor kappa B (NF-kappa B) for their expression. We addressed the hypothesis that immunomodulatory activities of PRL might arise from an increase in NF-kappa B activity. MAC-T cells, a bovine mammary epithelial cell line, were stimulated with increasing concentrations of bovine PRL ( 1, 5, 25, 125, and 1,000 ng/mL). Level of NF-kappa B binding activity was measured and mRNA was evaluated for IL-1 beta, IL-6, IL-8, granulocyte-macrophage colony-stimulating factor (GMCSF), IFN-gamma, and tumor necrosis factor (TNF)-alpha, cytokines known to require NF-kappa B for their maximal transcription. Prolactin activated NF-kappa B; maximal NF-kappa B activation was weaker with PRL than with TNF-alpha at 30 or 180 min poststimulation. In addition, PRL significantly amplified, in a dose-dependent manner, mRNA expression of IL-1 beta, IL-6, IL-8, GMCSF, and TNF-a. We measured PRL concentrations in blood and milk from healthy and chronic mastitis-infected cows, and studied the relationship between the PRL concentration and the degree of inflammation in the mammary gland as indirectly assessed by somatic cell counts (SCC). Plasma PRL did not differ significantly between healthy and chronic mastitis-affected cows (63.7 and 67.5 ng/mL, respectively). Milk PRL concentration was significantly increased in chronic mastitis-affected quarters with the highest SCC, and had a positive significant correlation between SCC, as well as between the number of neutrophils present in milk samples. The present findings show that PRL promotes an inflammatory response in bovine mammary epithelial cells via NF-kappa B activation, and suggest a role for PRL in the pathogenesis of chronic mastitis. The purpose of this retrospective study was to further identify in the equine species the clinical signs and blood parameters that could be useful to identify a hepatic disease and act as predictors of the outcome in animals suffering from an acute hepatic insufficiency. The study included 31 equids that were hospitalized at the University of Liege and that suffered from a hepatic (group 1, 17 cases of hepatitis; group 2, 11 cases of hyperlipemia) or post-hepatic (group 3, 3 cases of cholelithiasis) disease. Frequency of the clinical signs and values of selected blood parameters on admission were compared statistically between surviving and non-surviving animals. The most frequently presented clinical signs were depression, decreased appetite or anorexia, fever, tachycardia, polypnea, icterus, and congested mucous membranes. Frequency of the clinical signs and blood parameter values were not statistically different in surviving and non-surviving animals, with the exception of gamma-glutamyl transferase (GGT) and alkaline phosphatase (ALP), which were significantly higher in animals with fatal outcome, with a cutoff value of 224 and 820 IU/L, respectively. OBJECTIVE: To evaluate the usefulness of multisite quantitative ultrasonography for noninvasive assessment of bone in horses. SAMPLE POPULATION: 12 healthy horses and both forelimbs from 8 clinically normal horses. PROCEDURE: For in vivo measurements, various regions of interest (ROI) were examined on the third metacarpal bone, radius, and tibia. Precision error for speed of sound (SOS) measurements was obtained by measuring each ROI of 4 horses 10 times with probe repositioning. Additionally, 3 operators measured each aspect of the third metacarpal bone of 6 horses 5 times each. For ex vivo measurements, third metacarpal bones were examined at 9 ROI, and SOS measurements were performed before and after soft tissue removal. One ROI of a single forelimb was subjected to 96 ex vivo measurements with 3 different contact media. RESULTS: The lateral aspect of the third metacarpal bone had significantly higher SOS values than the dorsal and medial aspect of the third metacarpal bone. No difference was obtained between SOS values of the lateral and medial aspect of the radius. The tibia had significantly higher SOS values than the lateral aspect of the radius and the dorsal and medial aspect of the third metacarpal bone. Intraoperator coefficients of variation ranged from 0.62 to 3.15%, and interoperator coefficients of variation ranged from 0.78 to 2.70%. Values of SOS were highest when silicone oil was used as the contact medium. CONCLUSIONS AND CLINICAL RELEVANCE: Speed of sound measurements obtained by quantitative ultrasonography in axial transmission mode can be used to precisely measure superficial cortical bone properties of third metacarpal bone, radius, and tibia in horses.
