The relationship between the concentration of plasma progesterone (P4) during embryo attachment or at recognition of pregnancy, and that of pregnancy-associated glycoprotein (PAG) was assessed in dairy cows. The outcome of artificial insemination (AI) was classified as positive (AI+), negative (AIÿ), or late embryonic mortality (EM) by measuring circulating PAG concentrations and by ultrasonography. Based on P4 concentrations at either day 21 or day 15, AI+ and EM cows were classified into ‘low’ (P4 concentrations < mean) and ‘high’ (P4 concentrations > mean) P4 groups. In both experiments, the threshold of P4 concentration between the ‘low’ and ‘high’ groups was approximately 6 ng/mL. PAG concentrations were lower in the ‘low’ group only when P4 concentrations were below the threshold. The study findings suggest that a possible P4 threshold exists below which PAG secretion may be impaired [less ▲]

Pregnancy-associated glycoproteins (PAG) constitute a large family of glycoproteins found in the outer placental epithelial cell layer of the placenta in Eutherian species. In ruminants, they are noted to be structurally closely related among the different species. This study was designed to determine PAG concentrations in maternal and fetal plasma, allantoic and amniotic fluids in buffalo species. Antisera (AS) generated in rabbits against distinct PAG molecules were used in three RIA-PAG systems: RIA-1 (raised against bovine PAG67kDa; AS#497), RIA-2 (raised against caprine PAG55+62kDa; AS#706) or RIA-3 (raised against buffalo PAG; AS#859). Samples were collected at slaughterhouse (n = 67). PAG concentrations determined by RIA-2 gave significantly higher results in both allantoic and amniotic fluids (12.7 ± 2.1 ng/ml and 24.0 ± 7.3 ng/ml, respectively). Regarding maternal and fetal plasma, PAG concentrations obtained by RIA-2 (21.8 ± 2.4 ng/ml and 20.2 ± 2.5 ng/ml, respectively) and RIA-3 (25.0 ± 2.2 ng/mL and 21.9 ± 3.2 ng/ml, respectively) were higher than those obtained by RIA-1 (15.5 ± 1.4 ng/ml and 16.1 ± 1.8 ng/ml, respectively). The correlation among the three systems was very high. The study clearly reveals the ability of PAG-RIA systems to determine PAG concentration in swamp buffalo samples. [less ▲]

In the present study we report the development of a new ELISA for PAG measurement (Ref. code E.G.7, CER, Maloie) in ruminant species. Purified boPAG67kDa protein was purified in our laboratory and used as standard. Antiserum raised against caprine PAG 55kDa+62kDa (AS707) was used as capture antibody (1:40,000). Antiserum raised against PAG purified from buffalo placentas (AS859) was used as detection antibody (1:32,000 dilution). Avidin-HRP and TMB completed the ELISA system. Reproducibility was determined by calculating the intra- and inter-assay CV. Accuracy was determined by adding increasing concentrations of purified boPAG67kDa to bovine sera containing low PAG concentrations. Specificity was tested regarding 25 different compounds: carbohydrates, aspartic proteinases, hormones and major plasma proteins. Parallelism was assessed by serially diluting pregnant cow serum containing high PAG concentrations. Sera was collected from pregnant cows (n=4) during the whole duration of gestation. Samples were tested by both RIA and ELISA. Samples giving high PAG concentrations were serially diluted in order to fit with optical density (OD) of the ELISA standard curve. Intra-and inter-assay CV ranged from 5.5 to 9.1%. Accuracy ranged from 92.9 to 105.5%. No cross reaction was observed with almost all tested compounds excepting lactic acid at supra-physiological levels (1 mg/ml). Dilutions of bovine maternal sera were parallel to standard curve. Profiles obtained by ELISA in pregnant cows were very similar to those obtained by RIA. In conclusion, a new PAG-ELISA is available for measurement of PAG concentrations in cattle. This new test was validated in vitro and can be used for pregnancy diagnosis and follow-up of gestation in cattle. [less ▲]

In the present study we report the use of a new PAG-ELISA for routine pregnancy diagnosis in cattle. A total of 2777 samples were collected from pregnant or non-pregnant cows and assayed by both classical PAG RIA-497 method and newly developed PAG-ELISA (Ref. code E.G.7, CER, Maloie). Pregnancy status was determined on the basis of RIA results. Samples were assumed to be issued from pregnant cows when PAG concentrations were higher than 0.8 ng/mL. PAG concentrations <0.8 ng/mL were considered negative (non-pregnant). PAG concentrations between 0.8 and 1.0 ng/mL were considered doubtful. With regard to ELISA, concentrations below 0.6 were considered negative. Concentrations between 0.6 and 0.8 ng/mL were considered doubtful and finally concentrations under 0.8 ng/mL were assumed as positive. Sensitivity(Se), specificity(Sp) and predictive values (PV) of pregnancy diagnosis were determined. A total of 143(5.2%), 137(4.9%) and 38(1.4%) samples gave doubtful results by RIA, ELISA or both systems, respectively. These samples (318/2777=11.5%) were not used for estimation of Se, Sp, PPV and NPV because clinical confirmation of pregnancy diagnosis could not be established by an additional method (rectal palpation, ultrasound or calving). Taking into account the remaining samples (2459), Se, Sp, PPV and NPV obtained by ELISA were respectively 98.9%, 89.9%, 98.7% and 96.4%. In conclusion, PAG-ELISA can be successfully used for pregnancy diagnosis in cattle. In the near future, further investigations are to be conducted in order to compare results obtained by ELISA test with those obtained by rectal palpation and ultrasound in field conditions. [less ▲]

The concentrations of pregnancy-associated glycoproteins (PAG) were determined in buffalo cows (Bubalus bubalis) using three different radioimmunoassay (RIA) systems (RIA-497, RIA-706, and RIA-708). Samples (10 mL) were collected by jugular venipuncture from Week 0 until Week 28 of pregnancy (9 females), and from parturition until Week 10 postpartum (13 females). During pregnancy, PAG molecules were detectable at Week 6 using the three aforementioned RIA systems (3.9 ± 1.3 ng/mL, 9.7 ± 1.3 ng/mL, and 9.9 ± 0.7 ng/mL for RIA-497, RIA-706, and RIA-708, respectively). These concentrations increased gradually until Week 28, reaching values of 39.6 ± 4.0 ng/mL (RIA-497), 50.5 ± 11.9 ng/mL (RIA-706), and 68.2 ± 20.8 ng/mL (RIA-708). PAG concentrations determined by RIA-497, RIA-706, and RIA-708 were strongly correlated throughout the entire gestation period, RIA-708 giving the higher concentrations. At parturition, the mean concentrations ranged from 34.9 ± 4.0 (RIA-497) to 84.7 ± 10.6 ng/mL (RIA-708). Thereafter, the concentrations decreased steadily, reaching very low levels (< 1.0 ng/mL) at Week 8 postpartum. In conclusion, PAG concentrations measured by the above three RIA systems showed a profile similar to those previously described in bovine species, with higher concentrations being detected by RIA-706 and RIA-708. However, the patterns of PAG concentrations, studied using the three aforementioned PAG-RIA systems, differed around parturition, with very low levels being observed in the female buffaloes [less ▲]

The present study describes the isolation and characterization of new pregnancy-associated glycoprotein molecules (PAG) from midpregnancy and late-pregnancy placentas in the water buffalo (Bubalus bubalis). After extraction, the homogenates are subjected to acid and ammonium sulfate precipitations followed by DEAE chromatography. Subsequently, the water buffalo PAG (wbPAG) from these solutions are enriched by Vicia villosa agarose (VVA) affinity chromatography. As determined by western blotting with anti-PAG sera, the apparent molecular masses of the immunoreactive bands from the VVA peaks range from 59.5 to 75.8kDa and from 57.8 to 73.3kDa in the midpregnancy and late-pregnancy placentas, respectively. Amino-terminal microsequencing of the immunoreactive proteins has allowed the identification of three distinct wbPAG sequences, which have been deposited in the SwissProt database: RGSXLTIHPLRNIRDFFYVG (acc. no. P85048), RGSXLTILPLRNIID (acc. no. P85049), and RGSXLTHLPLRNI (acc. no. P85050). Their comparison to previously identified proteins has shown that two of them are new because they have not been described before. Our results confirm the suitability of VVA chromatography for the enrichment of the multiple PAG molecules expressed in buffalo placenta. [less ▲]
