Economically, cheese yield (CY) is very important. Todate, empirical or theoretical formulae allow estimating the theoretical CY from milk fat and casein or protein content of milk. It would be interesting to predict CY during milk recording directly without the need to estimate milk components. Through the BlueSel project, 157 milk samples were collected in Wallonia from individual cows and analyzed using a mid-infrared (MIR) MilkoScanFT6000 spectrometer. Individual laboratory cheese yields (ILCY) were determined for each sample and expressed as g of dry coagulum/100 g of milk dry matter. An equation to predict ILCY from MIR was developed using partial least squared regression (Winisi III). A first derivative pre-treatment of spectra was used to correct the baseline drift. To improve the repeatability of the spectral data, a file which contained the spectra of samples analyzed on 5 spectrometers was used during the calibration. During calibration, 23 outliers were detected a nd removed from the calibration set. The ILCY mean of the final calibration set was 63.9% with a SD of 11.2%. The calibration (C) coefficient of determination (R²) was equal to 0.76 with a standard error (SE) of calibration of 5.5%. A full cross-validation (CV) was preformed to assess the robustness. R²cv was 0.72 with a SECV of 6.0%. The similarity between R²c and R²cv as well as between SEC and SECV permits to consider robustness of the developed equation as good. Even if it is planned to improve the equation with additional samples, this first equation will permit to study ILCY in the Walloon dairy cattle. [less ▲]

To develop a new type of microbiological Reference Materials (RMs), displaying long-term stability at room temperature. The purpose was to produce and validate two batches of RMs for the enumeration of Bacillus cereus and Clostridium perfringens. Methods and Results: The RMs were based on spores of B. cereus and Cl. perfringens, adsorbed on calcium carbonate pellets. Two batches of 1000 units were manufactured and validated in compliance with ISO guide 35. After verification of their homogeneity, the stability of the ‘RM-B. cereus’ and ‘RMCl. perfringens’ batches was proven during at least 36 and 9 months, respectively, at room temperature. The validation study was completed by international collaborative trial involving 12 laboratories, allowing the validation of the assigned values. Conclusions: The methodology developed in this work enabled to produce easy-to-handle and cost-effective RMs, displaying an unprecedented stability at room temperature, a good homogeneity and a precise and validated assigned value. Significance and Impact of the Study: This study revealed new paths for the development of stable microbiological RMs. Overcoming the intrinsic instability of the living cells makes it possible to produce valuable tools for the quality assurance of microbiology laboratories. Agar contact microbiological sampling techniques, based on a transfer of the microorganisms present on a surface to a culture medium, are widely used to assess and control surface cleanliness and to evaluate microbial contamination levels. The effectiveness of these techniques depends on many environmental parameters that influence the strength of attachment of the bacteria to the surface. In the present study, stainless steel and high density polyethylene surfaces were inoculated with known concentrations of Staphylococcus epidermidis. Following an experimental design, the surfaces were sampled with different types of replicate organism direct agar contact plates and Petrifilm; results indicated that recovery rates were influenced by the presence of egg white albumin or Tween 80 in the inoculum solutions or by the introduction of surfactants into the contact agar of the microbiological sampling techniques. The techniques yielded significantly different results, depending on sampling conditions, underlining the need for a standardization of laboratory experiments to allow relevant comparisons of such techniques. Milk coagulation has a direct effect on cheese yield. Several factors influence the milk coagulation kinetics. In addition to calcium and milk protein concentrations, titratable acidity influences all the phases of milk coagulation. The objective of this research was to study the feasibility of prediction of titratable acidity directly in bovine milk using mid-infrared spectrometry. In order to maximize the variability in the measurements of titratable acidity, milk samples were collected on basis of several criteria (e.g. breeds). The titratable acidity was recorded as Dornic degree. All samples were also analyzed by MIR spectrometry. Using partial least squares regressions and first derivative pretreatment of spectral data, a calibration equation was built to predict the Dornic degree in cow milk. First results were promising and showed the potentiality to this calibration. The calibration and cross-validation coefficients of determination were 92.25 and 89.88 %, respectively. Moreover, the ratio of standard error of prediction to standard deviation was 3.13 and permits us to consider the calibration equation as usable in most application such as scientific researches and the screening of the Walloon dairy herd particularly in order to improve the milk coagulation properties. Assessing the microbial contamination level of surfaces is critical in environments where high hygiene levels are required. This is typically the case in food and catering industries or in hospitals and medical appliances. On a routine base, RODAC plates and other techniques based on microbial transfer are generally used to quantify the microbial load of a surface. There are however still polemics on the limitations and performances of these techniques arising from the fact that the initial contamination level of a surface is never known and due to the difficulty of reproducing field conditions in laboratory environments.
