Cellulose is a linear biopolymer composed of glucose residues linked by β 1-4 glucosidics bonds. These characteristics enable cellulose molecules to adopt an extended rod-like configuration. In the microfibrils, the multiple hydroxyl groups on the glucose residues hydrogen bond with each other, holding the chains firmly together and contributing to their high tensile strength. This study has shown the possibility to produce cellulose microfibrils by applying shearing treatment. Homogenisation and microfluidisation treatments permit to obtain microfibrils with 5 to 20 μm length and a 20 to 50 nm diameter. This study also shows that a tridimensional network, typical of a gel, appears in the treated samples [less ▲]

Two different procedures, alcohol precipitation with washing (APW) and a 10 kD MWCO membrane ultrafiltration with diafiltration (UF-DF), for pectin recovering and purification from crude aqueous extracts were investigated. The results showed that the yield, purity, chemical, and physicochemical features of isolated pectins depended upon the type of procedure used. The APW technique gave a higher pectin yield, and the isolated pectin contained more neutral sugars, more proteins, and more ash but less galacturonic acids than the 10 kD membrane UF-DF one. The weight-average molar mass of the APP was lower and its emulsifying properties slightly better than those of the UFPR10. Molar mass distribution of APP or UFPR10 showed two distinct pectin fractions peaks, which were separated in high-MW and low-MW pectin fractions using a 50kD MWCO membrane. The high-MW fractions exhibited poor emulsifying and emulsion stabilizing abilities whereas the low-MW fractions displayed a much higher emulsifying ability than the initial pectin fractions. Therefore, it was inferred that the ability of beet pectin to be an effective emulsifier and/or emulsion stabilizer was greatly dependent on its weight-average molar mass. (c) 2006 Elsevier Ltd. The surface chemical composition of nine strains of Bacillus subtilis was determined by X-ray photoelectron spectroscopy. Regressions between elemental concentrations and concentrations associated with different components of C1s, N1s, and O1s peaks provided a more precise validation of the procedure used for peak decomposition and allowed the assignment of the peak components to be completed or strengthened. The component of the O1s peak appearing around 531.2 eV was shown to contain a contribution of oxygen from phosphate groups (P=O, P–O−), the other contribution being due to oxygen involved in amide functions. The surface negative charge may be fully attributed to phosphate groups, despite the observation of two types of zeta potential vs pH curves. The strains exhibiting a sharp variation of the zeta potential (range of −35 to −55 mV) between pH 2 and 4.7 were characterized by a high phosphate surface concentration and by an excess (about 25%) of phosphate with respect to the sum of potassium, an exchangeable cation, and protonated nitrogen, attributed to protein or to alanine involved in teichoic acids. Mixed monolayers of the surface-active lipopeptide surfactin-C-15 and various lipids differing by their chain length (DMPC, DPPC, DSPC) and polar headgroup (DPPC, DPPE, DPPS) were investigated by atomic force microscopy (AFM) in combination with molecular modeling (Hypermatrix procedure) and surface pressure-area isotherms. In the presence of surfactin, AFM topographic images showed phase separation for each surfactin-phospholipid system except for surfactin-DMPC, which was in good agreement with compression isotherms. On the basis of domain shape and line tension theory, we conclude that the miscibility between surfactin and phospholipids is higher for shorter chain lengths (DMPC > DPPC > DSPC) and that the polar headgroup of phospholipids influences the miscibility of surfactin in the order DPPC > DPPE > DPPS. Molecular modeling data show that mixing surfactin and DPPC has a destabilizing effect on DPPC monolayer while it has a stabilizing effect towards DPPE and DPPS molecular interactions. Our results provide valuable information on the activity mechanism of surfactin and may be useful for the design of surfactin delivery systems. (c) 2007 Elsevier B.V. All rights reserved. [less ▲]
