As the inbred mouse strain SJL/J displays increased resistance to several pathogens and as its immune system shows multiple specificities, it is tempting to infer a causal link between these observations. The first question that comes to mind is whether adaptive immunity plays a role, and a way to answer this question is to see if the resistance phenotype persists when adaptive immunity is depressed. Although it has long been known that irradiation causes repression of leukopoiesis in mice, the technical data available in the literature are of no help in the case of strain SJL/J, because it displays exceptional radioresistance. Here we show that exposure of SJL/J to ∼9Gy, an intensity corresponding to the lethal dose 50 for the species Mus musculus, leads to serious but reversible alteration of leukopoiesis. This conclusion stems from an examination of the effects, 1-11 days post-exposure, of whole-body gamma-ray irradiation on leukocyte populations in the thymus and peripheral blood of young adult females. Immunodepression was most severe 4 days post-exposure. As in other strains, leukocyte populations displayed differential radiosensitivity, B (CD19(+)) cells being most sensitive, T (CD4(+)/CD8(+)) cells moderately sensitive, and natural killer (NK1.1(+)) cells most resistant. Surprisingly, however, the helper/inducer T lymphocytes proved more resistant than the cytotoxic/suppressor T lymphocytes, contrarily to what is observed in other strains. The procedure described will make it possible to refute or establish reliably the existence of causal links between SJL-specific phenotypic traits and immune aberrations and to elucidate further the respective roles of innate and acquired immunity in determining the resistance of this strain to an array of viral diseases. We herein describe the positional identification of a 2-bp deletion in the open reading frame of the MRC2 receptor causing the recessive Crooked Tail Syndrome in cattle. The resulting frame-shift reveals a premature stop codon that causes nonsense-mediated decay of the mutant messenger RNA, and the virtual absence of functional Endo180 protein in affected animals. Cases exhibit skeletal anomalies thought to result from impaired extracellular matrix remodeling during ossification, and as of yet unexplained muscular symptoms. We demonstrate that carrier status is very significantly associated with desired characteristics in the general population, including enhanced muscular development, and that the resulting heterozygote advantage caused a selective sweep which explains the unexpectedly high frequency (25%) of carriers in the Belgian Blue Cattle Breed. This experiment aims to compare in vivo breast morphometric and ultrasound measurements, as well as live body weight to predict breast meat weight in slow-growing chickens. Traits included Thoracic Circumference (TC), Chest Width (CW), Keel Length (KL) and angle (KA), Live Weight (LW), thickness of muscle determined by sonography (TM) and Breast Meat Weight (BMW). Birds were then slaughtered and total breast muscles (Pectoralis major and Pectoralis minor) were dissected and weighed. A linear model including sex effect and the different predictor measurements, as covariates, were adjusted to the data. Homogeneity test of slopes between sexes showed no difference. Means of the traits were 115.58g (±19.72) for BMW, 1031 g (±163.44) for LW, 68.65o (±6.89) for KA, 26.81 cm (±1.57) for TC, 10.40 cm (±0.62) for KL, 4.67 cm (±0.47) for CW and 11.52 mm (±1.11) for TM. All traits were highly correlated to BMW: TC (0.85), LW (0.84), KL (0.81) and TM (0.79), except for KA (0.28) and CW (0.19). Finally, TC, LW, KL and TM appear to be valuable indicators for estimating BMW in slow-growing chickens but KA and CW remain poor predictors [less ▲]

Enhancing the use of traditional chicken breeds is necessary to insure long-term conservation of global genetic diversity in poultry. A logical start in this process is to evaluate performance. The objective of this study was to estimate reproductive traits in the Ardennaise chicken breed under traditional and modern management systems. Reproductive performance indicated lighting programs [Natural (NAT) or Artificial (ART) day length] were highly significant for egg weight, hatchability (p<0.001), early and mid-term embryonic mortality (p<0.01). Feeding treatments [ad libitum (AL) or mild Feed-restriction (FR)] varied greatly with egg weight, hatchability, fertility (p<0.001) and late embryonic mortality (p<0.05). Most precocious age at first egg (23 weeks), longest duration of laying period (>27 weeks), utmost maximum production peak (56.67%), largest egg number (70.13 eggs/hen) and number of chicks hatched (24.10 chicks/hen) were performed under ART-AL. Heaviest egg weight (53.21g), highest fertility (70.20%), greatest hatchability (55.58%), highest liveability during rearing (95.74%) and lay (100%), lowest early (3.51%) and mid-term embryonic mortality (1.17%) were achieved under NAT-AL. To summarize, most of traits studied in the Ardennaise chicken were moderate when compared to those expected by broiler breeders. [less ▲]

Degenerative osteoarthropathy resulting in a reduced active lifespan was observed in Ardenner horses. In the context of joint biology, insulin-like growth factor I (IGF-I) is a potential candidate to affect the anabolism of cartilage matrix molecules. A group of 30 Ardenner horses reared under standardized conditions from weaning were evaluated periodically from 15 to 28 months of age to detect the early manifestations of the disease. At the end of this period, horses were classified in two pathological groups related to the degree of interphalangeal degenerative osteoarthropathy based on clinical and radiographic evaluations: healthy (46.7%) and pathological (53.3%) horses. Seven sequential blood samples were taken from each horse (during the evaluation period) to study the variation of IGF-I plasma concentration. We tested the variations of the IGF-I plasma concentration during growth, and the effect of sex and of pathological classes. Significant variations were observed during the research period, with a maximum value corresponding to spring and a minimum in autumn. A significant reduction of the IGF-I plasma concentration was also observed in the pathological horses (433.5 +/- 19.5 ng/ml) compared to the healthy horses (493.9 +/- 18.2 ng/ml). An alteration in the level of this growth factor could induce a disregulation of the mechanisms involved in the local control of joint and bone tissue development. [less ▲]

The bovine species is highly susceptible to respiratory dis-eases. A previous experiment has shown that Belgian Blue and White calves have lower spiro-metric variables than Holstein calves. The first aim of this study was to measure the spirometric variables (peak inspiratory flow, peak expiratory flow, maximal tidal volume, maximal ventilation in 15 seconds and ventilatory reserve) of 18 crossed Belgian Blue x Holstein calves and of 32 Dual purpose calves by the lobelin test and to compare them with 19 Holstein calves and 22 Belgian Blue and White calves. The second aim was to test the correlation between some mor-phometrical parameters (chest girth, thoracic width, thoracic length, thoracic height, thoracic section and costal angle) and those spirometric variables. The spirometric variables of the crossed calves and of the Dual purpose calves are situated between the ones of the Belgian Blue and White and of the Holstein. There is a positive sig-nificant correlation between these parameters and the spirometric variables. The higher cor-relation is obtained with the chest girth. All these parameters are easier and quicker measura-ble, and at lower cost than the spirometric variables [less ▲]

The effects of weight loss on hormonal and biochemical blood parameters were measured monthly [carnitine, creatinine, urea, free T4 (fT4), total T4 (TT4), plasma alkaline phosphatases (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), potassium and total proteins] or bimonthly [cholesterol, triglycerides, non-esterified fatty acids (NEFA), insulin-like growth factor I (IGF-I), glucose, insulin] in eight obese Beagles dogs fed either a high protein dry diet, DP (crude protein 47.5%, on dry matter basis) or a commercial high fibre diet, HF (crude protein 23.8%, crude fibre 23.3%). The dogs were allotted to two groups according to sex and body weight (BW) and they were respectively fed with the DP or the control HF diet during 12-26 weeks, until they reach their optimal BW. The plasma basal triglycerides and cholesterol concentrations were decreased by the two diets but the difference was only significant for the DP diet. The plasma mean NEFA concentration increased regularly over the period with the HF diet, without significant difference between the two diets. No effect of diet or weight loss was observed on plasma carnitine, urea, creatinine, ALP, AST, ALT, potassium, TT4, FT4, IGF-I, glucose and insulin. Weight loss induced a decrease in fT4 plasma concentration (p < 0.001). The high protein diet allowed a safe weight loss. [less ▲]

Real-time ultrasound data collection, consisting specifically of longissimus thoracis muscle echogenicity, was carried out on 80 Pietrain pigs (42 gilts and 34 barrows) using the Pie Medical scanner 200 equipped with an ASP-18 probe and 3.5 MHz to predict intramuscular fat. Two ribs thickness (12th and 13th ribs) was excised from the longissimus thoracis muscle for subsequent ether extract value (EE) determination. The percentage of white pixels in the ultrasound image were related to EE percentage. The EE percentages were 1.44, 1.37 and 1.15 % respectively for the homozygous stress-negative Pietrain (CC), heterozygous stress-negative Pietrain (CT) and homozygous stress-positive Pietrain (TT) individuals. The percentage of white pixels in the longissimus thoracis images were 9.98, 8.75 and 7.79 % respectively for CC, CT and TT genotypes. The barrows had an higher intramuscular fat and white pixel percentage than the gilts. The determination coefficient (R 2) of the EE prediction model originating from the white pixels percentage was 0.35 with a root mean squared error of 0.26 %. These performance could be potentially improved using the calibration phantom of the ASP-18 probe, before taking images. [less ▲]
