ABSTRACT: BACKGROUND: Recurrent airway obstruction (RAO, also known as equine heaves) is an inflammatory condition caused by exposure of susceptible horses to organic dusts in hay. The immunological processes responsible for the development and the persistence of airway inflammation are still largely unknown. Hypoxia-inducible factor (Hif) is mainly known as a major regulator of energy homeostasis and cellular adaptation to hypoxia. More recently however, Hif also emerged as an essential regulator of innate immune responses. Here, we aimed at investigating the potential involvement of Hif1-alpha in myeloid cells in horse with recurrent airway obstruction. RESULTS: In vitro, we observed that Hif is expressed in equine myeloid cells after hay dust stimulation and regulates genes such as tumor necrosis factor alpha (TNF-alpha), interleukin-8 (IL-8) and vascular endothelial growth factor A (VEGF-A). We further showed in vivo that airway challenge with hay dust upregulated Hif1-alpha mRNA expression in myeloid cells from the bronchoalveolar lavage fluid (BALF) of healthy and RAO-affected horses, with a more pronounced effect in cells from RAO-affected horses. Finally, Hif1-alpha mRNA expression in BALF cells from challenged horses correlated positively with lung dysfunction. CONCLUSION: Taken together, our results suggest an important role for Hif1-alpha in myeloid cells during hay dust-induced inflammation in horses with RAO. We therefore propose that future research aiming at functional inactivation of Hif1 in lung myeloid cells could open new therapeutic perspectives for RAO. [less ▲]

The clinical examination and the ancillary tests at the disposal from the veterinarian may be poor diagnostics in horses suffering from subclinical inflammatory airway disease or in clinical remission of recurrent airway obstruction (RAO). The objective of this study was to determine whether the methacholine bronchoprovocation test -which is daily used for diagnosis of asymptomatic human asthma- may be used as an applicable and repeatable method for the diagnosis of an increased susceptibility to bronchospasm in horses. The test, realised on 8 horses (6 with RAO history and 2 controls) presents a good feasibility and significant repeatability. Dust exposure increased the bronchoreactivity, while it did not affect other clinical and functional parameters or inflammatory markers. In the future, the bronchoprovocation test could therefore find its place in clinicat practice for the detection of asymptomatic horses which are susceptible to develop bronchospasm in poor environmental conditions. A total of 15 boars from 3 genetic groups were used for this study, including 5 Piétrain boars (purebred Piétrain), 5 ♀Duroc × ♂Piétrain (½ Piétrain) and 5 ♀(Piétrain × Duroc) × ♂Duroc (¼ Piétrain) in order to evaluate the growth performance and sperm quality of stress negative Piétrain boars and their hybrids. Testing period started at an average age of 60 days and ended at an average age of 225 days. The growth performances were weights at starting and finishing periods, average daily gain, backfat thickness, longissimus depth and lean content. The sperm quality was assessed using ejaculate volume, spermatozoon motility, sperm concentration, total number of spermatozoon in the ejaculate, rate of abnormal spermatozoon and pH of sperm. All these measurements were used to compare the genetic groups. The results showed that: The average daily gain of stress negative Piétrain boar purebred was not significantly different from Piétrain hybrids while the lean content was higher. The ejaculate volume and the spermatozoon motility of stress negative Piétrain purebred boars were higher than Piétrain hybrids but the sperm concentration and the total number of spermatozoon in the ejaculate were lower. Echocardiography has become a routine non invasive cardiac diagnostic tool in most species. Accurate measurement of cardiac dimensions requires reference values, which are poorly documented in goats. Goats are animals easy to handle with a body and heart size comparable to humans’. This makes goats an attractive candidate for the development of animal models for human cardiology research. The aim of this study was to test the repeatability and to establish the reference values of bi-dimensional (2D-), time-motion (M-) mode and pulsed wave (PW) Doppler echocardiographic variables in adult goats. Six healthy female adult Saanen goats were investigated three times by the same observer at one day interval using a standardized 2D-, M-mode and PW Doppler echocardiographic protocol. Calculation of the coefficient of variation for each variable measured within day and depending on the day allowed to evaluate their degree of variability. A single echocardiographic examination was performed in 6 other goats by the same observer, and the obtained values were added to these obtained on the third day of the 6 first goats. Then the observed mean, the standard deviation and the range of these measurements were calculated to establish the reference values of echocardiographic parameters in unsedated adult healthy female Saanen goats. Statistical analysis revealed a good inter-day repeatability of the 2D- and M-mode echocardiographic cardiac measurements, but PW Doppler parameters presented moderate to high variability, as documented in other species. Echocardiographic reference values obtained in healthy adult Saanen goats were similar to those reported in healthy adult sheep and in healthy adult humans. [less ▲]

REASONS FOR PERFORMING STUDY: There is a clinical impression that tetracaine hydrochloride (THCl) eyedrops is a suitable topical anaesthetic in horses. OBJECTIVE: To determine the duration of corneal anaesthesia following instillation of multiple doses and 2 concentrations of THCl in 10 healthy horses. METHODS: The corneal touch threshold (CTT) was determined, in both eyes, before (basal CTT) and after application of one drop of 0.5% THCl, 2 drops at a 1 min interval of 0.5% THCl or one drop of 1% THCl. CTT was measured in mm every 5 min until complete recovery of the basal CTT. Treatments were separated by an interval of at least one week. RESULTS: Corneal sensitivity was significantly reduced from baseline values for 30, 60 and 50 min after application of one drop of 0.5% THCl, 2 drops of 0.5% THCl and one drop of 1% THCl, respectively. Mean maximal anaesthetic effects, corresponding to a CTT of 0 mm, lasted 5.5, 16 and 15.25 min and maximal anaesthetic effect was present in 55, 90 and 80% of eyes, 5 min after application of one drop of 0.5% THCl, 2 drops of 0.5% THCl and one drop of 1% THCl, respectively. CONCLUSIONS: The application of a second drop or the use of more concentrated eyedrops significantly increases duration of both anaesthesia and maximal anaesthetic effect. POTENTIAL RELEVANCE: Duration of corneal anaesthesia following tetracaine instillation was established enabling a better use when performing diagnostic and therapeutic procedures. Comparison of tetracaine with other ocular anaesthetics needs to be published in the future. [less ▲]

REASONS FOR PERFORMING STUDY: Tetracaine hydrochloride (THCl) has been reported to cause irritation in dogs. In man, some topical anaesthetics have been shown to disrupt the tear film. Tear break-up time (TBUT) is a useful test allowing an assessment of the quality of the precorneal tear film. Only one TBUT value has been reported in horses with no information on the technique used. OBJECTIVES: To provide a method for performing the TBUT in horses and to report any side effects of a single application of THCl in clinically normal horses, particularly on the stability of the tear film. METHODS: In Study 1, one drop of 0.5 or 1% THCl was applied to one eye of 20 horses divided in 2 groups. Treated eyes were assessed for the development of side effects 2.5 and 5 min after treatment. In Study 2, the TBUT was measured in both eyes of 2 groups of 10 horses, before and 2.5 and 5 min after, instillation of one drop of either 0.5 or 1% THCl. RESULTS: No animals developed any ocular side effect after instillation. Basal TBUT was 8.3±1.3 s. TBUT decreased from baseline 5 and 2.5 min after application of one drop of 0.5% THCl and one drop of 1% THCl, respectively. CONCLUSIONS: A technique to measure the TBUT in healthy horses is described and normal range values that could be used as a reference were obtained. POTENTIAL RELEVANCE: THCl is well tolerated in horses but lowers the TBUT. [less ▲]

We report mapping of a quantitative trait locus (QTL) with a major effect on bovine stature to a approximately 780-kb interval using a Hidden Markov Model-based approach that simultaneously exploits linkage and linkage disequilibrium. We re-sequenced the interval in six sires with known QTL genotype and identified 13 clustered candidate quantitative trait nucleotides (QTNs) out of >9,572 discovered variants. We eliminated five candidate QTNs by studying the phenotypic effect of a recombinant haplotype identified in a breed diversity panel. We show that the QTL influences fetal expression of seven of the nine genes mapping to the approximately 780-kb interval. We further show that two of the eight candidate QTNs, mapping to the PLAG1-CHCHD7 intergenic region, influence bidirectional promoter strength and affect binding of nuclear factors. By performing expression QTL analyses, we identified a splice site variant in CHCHD7 and exploited this naturally occurring null allele to exclude CHCHD7 as single causative gene. [less ▲]

A total of 395 tail samples of stress negative Piétrain new born piglets from the pig farm of Dong Hiep Haiphong were used to determine halothane genotypes (CC, CT and TT) in order to study the effect of mating type (♂CC×♀CC, ♂CC×♀CT, ♂CT×♀CC, and ♂CT×♀CT) on halothane genotype frequencies. Effects of genotype on live weight (174 pigs of 2 month old, 96 pigs of 5.5 month old), back fat thickness, loin muscle thickness and lean percentage (117 pigs), average daily gain (89 pigs) were also studied. Results showed that the mating type affected halothane genotype frequencies in the progeny. For the mating type ♂CC×♀CT and ♂CT×♀CT, the genotype containing allele T was reduced in the next generation; in the rest two genotypes (♂CC×♀CC and ♂CT×♀CC) the halothane allele frequency in the offspring was consistent with the theoretical frequency. The halothane genotypes CC and CT did not affect live weights at 2.0 and 5.5 months of age, average daily gain, backfat thickness, loin muscle thickness and lean percentage. [less ▲]
