Background Finding epistatic interactions in large association studies like genome-wide association studies (GWAS) with the nowadays-available large volume of genomic data is a challenging and largely unsolved issue. Few previous studies could handle genome-wide data due to the intractable difficulties met in searching a combinatorial explosive search space and statistically evaluating epistatic interactions given a limited number of samples. Our work is a contribution to this field. We propose a novel approach combining K-Nearest Neighbors (KNN) and Multi Dimensional Reduction (MDR) methods for detecting gene-gene interactions as a possible alternative to existing algorithms, e especially in situations where the number of involved determinants is high. After describing the approach, a comparison of our method (KNN-MDR) to a set of the other most performing methods (i.e., MDR, BOOST, BHIT, MegaSNPHunter and AntEpiSeeker) is carried on to detect interactions using simulated data as well as real genome-wide data. Results Experimental results on both simulated data and real genome-wide data show that KNN-MDR has interesting properties in terms of accuracy and power, and that, in many cases, it significantly outperforms its recent competitors. Conclusions The presented methodology (KNN-MDR) is valuable in the context of loci and interactions mapping and can be seen as an interesting addition to the arsenal used in complex traits analyses. The Kabyle goat breed represents 10.52% of the total Algerian market (3.8 million heads). A survey of 69 farmers has been carried out in order to characterize it and its breeding in the District of Chemini. The questions focused on household agricultural activities, including breeding of goats, cattle, sheep, rabbits, chicken, turkeys, honeybees and the production of olive oil and figs. The goat morpho-biometric characterization was based on 18 corporal measurements. The Hierarchical Cluster Analysis (HCA) of the farm structures defined four groups of farms, variance between groups accounting for 55.7% of total variability. The average number of goats in groups 1 to 4 was 7.2±2.8, 11.1±3.5, 22.3±1.4 and 3.4±1.0, respectively. Group 3 (n=6), showed the greatest number; it consists of older farmers (67 years old or over) not practicing arboriculture. They also have the largest numbers of sheep (48.67), rabbits (50.83), chicken (48.33) and turkeys (42). Group 4 (n=9), with the lowest number of goats, was the group of young farmers (39 years old or less), more dedicated to cattle breeding (~52 heads) and the production of olive oil (~207 trees) and figs (~47 trees). The 18 morpho-biometric variables were significantly higher in males than in females (p<0.05). The Kabyle goat is small (Height at withers: male: 68.23±0.97cm and female: 65.41±0.55cm) with long hair (male: 12.24±0.51cm, and female: 9.51±0.29cm). Its ears are drooping, its convex profile has a slightly pronounced nasal split and its dress color ranges from dark brown to black. This adapted genetic resource should be key in the development of a local production, based on a strong commitment of farmers inside a breed association, for the production of specimens corresponding to a standard, to be determined collectively. Introduction: The aim of this study was to establish echocardiographic reference values for the equine species using allometric regression equations based on body weight (BW) and thoracic circumference (TC). Animals: A total of 239 horses or ponies were studied, including 65 warmbloods, 33 Standardbreds, 41 Thoroughbreds, 32 Arabian horses, 28 draft horses and 40 ponies aged from 1 day to 30 years, weighing from 18 to 890 kg, with no evidence of cardiac disease. Methods: For each horse or pony, a two-dimensional and M-mode echocardiography was performed. Within each breed, the relationships between BW or TC and echocardiographic dimensions were examined using power regression equations. Predictions and their 95% prediction intervals were calculated for the echocardiographic measurements. Results: Within each breed, all echocardiographic measurements showed a significant and positive relationship with a high coefficient of determination for the estimation of the regression equations using BW and TC as the main explanatory variables. Breed-specific power regression equations as well as the 95% prediction intervals were calculated for each echocardiographic measurement as a function of BW and TC. Conclusions: In the future, the body size-corrected and breed-specific echocardiographic reference values calculated in the present study could be used to discriminate between normal and abnormal values in a given animal. Respiratory viral infections represent the most common cause of allergic asthma exacerbations. Amplification of the type-2 immune response is strongly implicated in asthma exacerbation, but how virus infection boosts type-2 responses is poorly understood. We report a significant correlation between the release of host double-stranded DNA (dsDNA) following rhinovirus infection and the exacerbation of type-2 allergic inflammation in humans. In a mouse model of allergic airway hypersensitivity, we show that rhinovirus infection triggers dsDNA release associated with the formation of neutrophil extracellular traps (NETs), known as NETosis. We further demonstrate that inhibiting NETosis by blocking neutrophil elastase or by degrading NETs with DNase protects mice from type-2 immunopathology. Furthermore, the injection of mouse genomic DNA alone is sufficient to recapitulate many features of rhinovirus-induced type-2 immune responses and asthma pathology. Thus, NETosis and its associated extracellular dsDNA contribute to the pathogenesis and may represent potential therapeutic targets of rhinovirus-induced asthma exacerbations. [less ▲]

Gammaherpesviruses are important human and animal pathogens. Infection control has proven difficult because the key process of transmission is ill understood. Murid herpesvirus 4 (MuHV-4), a gammaherpesvirus of mice, is transmitted sexually. We show that this depends on the major virion envelope glycoprotein gp150. gp150 is redundant for host entry, and in vitro, it regulates rather than promotes cell binding. We show that gp150-deficient MuHV-4 reaches and replicates normally in the female genital tract after nasal infection but is poorly released from vaginal epithelial cells and fails to pass from the female to the male genital tract during sexual contact. Thus, we show that the regulation of virion binding is a key component of spontaneous gammaherpesvirus transmission.IMPORTANCE Gammaherpesviruses are responsible for many important diseases in both animals and humans. Some important aspects of their life cycle are still poorly understood. Key among these is viral transmission. Here we show that the major envelope glycoprotein of murid herpesvirus 4 functions not in entry or dissemination but in virion release to allow sexual transmission to new hosts. [less ▲]

Canine idiopathic pulmonary fibrosis (CIPF) is a progressive interstitial lung disease mainly affecting West Highland white terriers (WHWTs). Thoracic high-resolution computed tomographic (T-HRCT) findings for CIPF acquired under general anesthesia have been described previously. However, the use of general anesthesia may be contraindicated for some affected dogs. Sedation may allow improved speed and safety, but it is unknown whether sedation would yield similar results in identification and grading of CIPF lesions. The aim of this prospective, observational, method-comparison, case-control study was to compare findings from T-HRCT images acquired under sedation versus general anesthesia for WHWTs affected with CIPF (n=11) and age-matched controls (n=9), using the glossary of terms of the Fleischner Society and a scoring system. Ground-glass opacity (GGO) was identified in all affected WHWTs for both sedation and general anesthesia acquisitions, although the GGO extent varied significantly between the two acquisitions (P<0.001). Ground-glass opacity was the sole lesion observed in control dogs (n=6), but was less extensive compared with affected WHWTs. Identification and grading of a mosaic attenuation pattern differed significantly between acquisitions (P<0.001). Identification of lesions such as consolidations, nodules, parenchymal and subpleural bands, bronchial wall thickening, and bronchiectasis did not differ between acquisitions. The present study demonstrated that T-HRCT obtained under sedation may provide different information than T-HRCT obtained under general anesthesia for identification and grading of some CIPF lesions, but not all of them. These differences should be taken into consideration when general anesthesia is contraindicated and sedation is necessary for evaluating WHWTs with CIPF. [less ▲]

Both endotherms and ectotherms (e.g., fish) increase their body temperature to limit pathogen infection. Ectotherms do so by moving to warmer places, hence the term "behavioral fever." We studied the manifestation of behavioral fever in the common carp infected by cyprinid herpesvirus 3, a native carp pathogen. Carp maintained at 24 degrees C died from the infection, whereas those housed in multi-chamber tanks encompassing a 24 degrees C-32 degrees C gradient migrated transiently to the warmest compartment and survived as a consequence. Behavioral fever manifested only at advanced stages of infection. Consistent with this, expression of CyHV-3 ORF12, encoding a soluble decoy receptor for TNF-alpha, delayed the manifestation of behavioral fever and promoted CyHV-3 replication in the context of a temperature gradient. Injection of anti-TNF-alpha neutralizing antibodies suppressed behavioral fever, and decreased fish survival in response to infection. This study provides a unique example of how viruses have evolved to alter host behavior to increase fitness. [less ▲]

This study aimed to determine morphological characteristics, reproductive performance and egg quality of Dong Tao chicken, an indigenous chicken breed in Vietnam. This study was conducted on 10 households raising purebred Dong Tao chicken. A sample of 240 adult individuals was morphologycally characterized. Reproductive performances were assessed on 10 backyard chicken (1 cock and 5 hens), and egg quality was assessed on 60 eggs. The results showed that Dong Tao chickens have a variety of phenotypes, as indicated in their feather colors, tarsus colors and comb types. The size of Dong Tao chickens was large, the body weight and all body measurements of the cocks were greater than those of hens (P < 0.001). Their reproductive performances were low but egg weight, yolk weight and the maximum breakage strength were higher compared to other chickens breeds. The Dong Tao breed, in the present state of things, is endangered. It should improve its reproductive performance before considering the in- situ conservation. [less ▲]

Gammaherpesviruses are important pathogens in human and veterinary medicine. During co-evolution with their hosts, they developed many strategies allowing them to shed infectious particles in presence of immune response. Understanding these strategies is likely to be important to control infection. Interestingly, we recently observed that Murid herpesvirus 4 (MuHV-4), a gammaherpesvirus infecting laboratory mice, could be sexually transmitted between mice. This model offers therefore the opportunity to understand the mechanisms underlying natural transmission. Some of these mechanisms could rely on the glycoprotein 150 (gp150), which could limit virus neutralization and promote the release of infectious particles from cells. In this study, we tested therefore the importance of gp150 in the context of MuHV-4 sexual transmission. Briefly, female mice were infected with WT or gp150- strains expressing luciferase. They were imaged with an in vivo imaging system to follow infection. When lytic replication was observed in the genital tract, infected females were mated with naïve males to compare the capacity of transmission of the two strains. Our results show that, while the gp150- strain has no deficit in reaching and replicating in the female genital tract, it displays a major deficit of sexual transmission in comparison with WT virions. Interestingly, this deficit appears to reflect a deficit of virions release from vaginal epithelial cells. Altogether, our results show that, while gp150 is not required for efficient dissemination and maintenance of MuHV-4 within its host, it is essential for efficient transmission, by promoting the releasing of infectious particles from the mucosal cells. Thanks to the genotyping of ~580,000 SNPs on 95 chickens, this study aimed to assess the genetic diversity in four Vietnamese local breeds (Mia, Mong, Ho and Dong Tao). Inbreeding was examined through the detection of runs of ho- mozygosity (ROH; Curik et al., 2014). The ROHs are long stretches of homozygous genome that are likely to re ect autozygosity. Given that Ho chickens have a small e ective population size, more inbreeding and consequently longer ROHs are expected. Conversely, Mia chickens, with a larger e ective population size should show less consanguinity and shorter ROHs. As expected, Mia chickens led to less observed homozygosity and less ROHs than the three other breeds. Moreover, when ROHs were subdivided in dif- ferent lengths, the proportion of individuals having at least one R OH longer than 5Mb was signi cantly higher in Ho (63%) than in Mia chickens (33%). Given that very long ROHs are particularly associated with recent inbreeding, it seems that many Ho chickens have consanguineous parents. However, no signi cant di erence was observed between the fourbreed concerning the total length of long ROHs (> 5Mb). In conclusion, the analysis of ROHs reported less consanguinity in Mia than in Ho chickens. In each breed, some individuals are found more consanguineous than expected. OBJECTIVES: To describe the results of a prevention campaign in terms of participation and pet health status and to identify opportunities to improve preventive medicine in cats and dogs. METHODS:An awareness campaign was designed to highlight the role of veterinarians and emphasise the benefits of a veterinary visit. Owners were invited to make an appointment for a free pet health check in a voluntarily participating veterinary clinic. Observations recorded by the veterinarians were entered in a database and subsequently analysed using simple descriptive statistics. RESULTS:A total of 5305 completed health check forms were analysed. The percentages of overweight and obese dogs and cats were 34 and 36%, respectively; this was the most common finding, followed by dental calculus (31% in dogs, 21% in cats). In total 67% of cats did not undergo flea control and 59% were not vaccinated. CLINICAL SIGNIFICANCE: Opportunities for increased quality of care are numerous given the high percentage of intact, unvaccinated or non-permanently identified pets and the low level of worm and flea control. Animal health should benefit from preventive measures, and improved management can be undertaken after early detection of diseases. [less ▲]

Rectal temperature measurement (RTM) can promote stress and defensive behaviour in hospitalised cats. The aim of this study was to assess if axillary temperature measurement (ATM) could be a reliable and less stressful alternative for these animals. In this prospective study, paired rectal and axillary temperatures were measured in 42 cats, either by a veterinarian or a student. To assess the impact of these procedures on the cat's stress state, their heart rate was checked and a cat stress score (CSS) was defined and graded from 1 (relaxed) to 5 (terrified). A moderate correlation was found between RTM and ATM (r=0.52; P<0.0001). RTM was on average 0.9°C (1.6°F) higher than ATM (P<0.0001), although a wide variation was found in the difference between these two measurements (−2.1°C to 3.6°C (−3.8°F to 6.5°F)). ATM failed to identify hypothermia in 25 per cent of the cases and hyperthermia in 19 per cent of the cases but may be considered less stressful than RTM. Indeed, RTM induced a mildly greater increase in heart rate (+6 bpm; P=0.01) and in CSS (+0.2; P=0.001) than ATM. The results were not affected by operator type. In conclusion, RTM should remain the standard method to obtain accurate temperatures in cats. [less ▲]

Human noroviruses (HuNoV) are the leading cause of acute nonbacterial gastroenteritis in humans and can be transmitted either by person-to-person contact or by consumption of contaminated food. Knowledge of an efficient disinfection for both hands and food contact surfaces is helpful for the food sector and provides precious information for public health. The aim of this study was to evaluate the effect of seven disinfectants belonging to different groups of biocides (alcohol, halogen, oxidizing agents, quaternary ammonium compounds, aldehyde and biguanide) on infectious viral titre and on genomic copy number. Due to the absence of a cell culture system for HuNoV, two HuNoV surrogates such as murine norovirus (MNV) and feline calicivirus (FCV), were used and the tests were performed in suspension, on gloves and on stainless steel discs. When, as criteria of efficacy, a log reduction > 3 of the infectious viral titre on both surrogates and in the three tests is used, the most efficacious disinfectants in this study appear to be biocidal products B, C and D, representing the halogens, the oxidizing agents group and a mix of QAC, alcohol and aldehyde, respectively. In addition, these three disinfectants also elicited a significant effect on genomic copy number for both surrogate viruses and in all three tests. The results of this study demonstrate that a halogen compound, oxidizing agents and a mix of QAC, alcohol and aldehyde are advisable for HuNoV disinfection of either potentially contaminated surfaces or materials in contact with foodstuffs. The objectives of this study were to determine the morpho-biometric characteristics of Vietnamese indigenous Ho chicken breed and to evaluate its production, reproduction performance and egg quality. The study was carried out on 20 households raising purebred Ho chicken from September 2013 to April 2015 in Northern Vietnam. Morpho-biometric characterization was based on measurement of 128 adult individuals. The egg production was observed on 66 hens within 20 households. A total of 29 eggs were freshly collected from 8 hens at the 20th laying week for quality analysis. Ho chicken breed is a breed with a large dimension, a large diversity in the colors (black with gold hackle, black copper, wheat, tan and tricolor), a heavy weight 3.78 and 2.64 for adult males and females, respectively), a low egg production (12.73 eggs/clutch) and a relatively low rate of embryo eggs (72.81%). A research to improve the egg production and reproduction should be undertaken to obtain a better performance, economic efficiency while maintaining this genetic heritage and sustaining the biodiversity of the avian breeds in Vietnam.
