REASONS FOR PERFORMING STUDY: Stress echocardiography could be a useful diagnostic test in horses suspected to suffer from exercise-induced myocardial dysfunction as a cause of exercise intolerance. OBJECTIVE: To evaluate the effect of treadmill exercise and pharmacological stress test on left ventricular echocardiographic parameters. METHODS: Echocardiography was performed in 2 groups of 5 healthy horses, either immediately after a near-maximal treadmill exercise (Group EXE) or during a pharmacological challenge (Group DOB) consisting of 35 microg/kg atropine, followed by incremental dobutamine infusion rates of 2-6 microg/kg bwt/min for 5 min duration each, which led to a cumulative dobutamine dose of 100 microg/kg. Left ventricular M-mode parameters were compared at rest and at heart rates of 80, 100, 110, 120 130, and 140 beats/min, within each group. RESULTS: In 2 horses of Group EXE, echocardiographic measurements were impossible at 140 and 130 beats/min, as their heart rates dropped too fast in the immediate post exercise period. In the remaining 3 horses image quality was not always satisfactory at heart rates of 130 and 140 beats/min. Systolic left ventricular parameters and fractional shortening measured at 130 and 140 beats/min were significantly different from values obtained at lower heart rates. Horses in Group DOB reached expected heart rates of 80 and 100 beats/min, after the administration of atropine and during a dobutamine infusion rate of 2 microg/kg bwt/min, respectively. Heart rates targets of 110, 120, 130, and 140 beats/min were reached at mean (+/- s.d.) dobutamine infusion rates of 2.8 +/- 0.4, 3.2 +/- 0.4, 4.0 +/- 0.7, 5 +/- 0.7 microg/kg bwt/min, respectively. Systolic left ventricular parameters and fractional shortening at heart rates of 110, 120, 130, and 140 beats/min, were significantly different from values obtained at lower heart rates. CONCLUSION: The pharmacological stress test induced changes in ventricular dimensions at heart rates of 80 to 140 beats/min. Using this test, high quality images can easily be obtained at heart rates of 140 beats/min. Conversely, in post exercise echocardiography, obtaining good quality images at heart rates of 130 and 140 beats/min is difficult, which limits use of the technique in routine clinical settings. POTENTIAL RELEVANCE: Further studies should demonstrate the potential of pharmacological stress test as a diagnostic tool in horses suffering from exercise-induced myocardial dysfunction. [less ▲]

The objective of the present study was to assess the validity of barometric whole-body plethysmography (BWBP), to establish reference values, and to standardise a bronchoprovocative test to investigate airway responsiveness using BWBP in healthy dogs. BWBP measurements were obtained from six healthy beagle dogs using different protocols: (1) during three consecutive periods (3.5 min each) in two morning and two evening sessions; (2) before and after administration of two protocols of sedation; (3) before and after nebulisation of saline and increasing concentrations of carbachol and histamine both in conscious dogs and in dogs under both protocols of sedation. Enhanced pause (PENH) was used as index of bronchoconstriction. Basal BWBP measurements were also obtained in 22 healthy dogs of different breeds, age and weight. No significant influence of either time spent in the chamber or daytime was found for most respiratory variables but a significant dog effect was detected for most variables. A significant body weight effect was found on tidal volume and peak flow values (P < 0.05). Response to carbachol was not reproducible and always associated with side effects. Nebulisation of histamine induced a significant increase in respiratory rate, peak expiratory flow, peak expiratory flow/peak inspiratory flow ratio and PENH (P < 0.05). The response was reproduced in each dog at different concentrations of histamine. Sedation with acepromazine + buprenorphine had little influence on basal measurements and did not change the results of histamine challenge. It was concluded that BWBP is a safe, non invasive and reliable technique of investigation of lung function in dogs which provides new opportunities to characterise respiratory status, to evaluate airway hyperresponsiveness and to assess therapeutic interventions. We constructed a mathematical model of the early response to Escherichia coli infection of the mammary gland and explored the roles and interactions between inflammatory cells and bacteria. The model incorporates 3 equations that describe the interactions among bacteria, milk somatic cells, and blood leukocyte densities. These 3 equations were fitted to cell densities observed during acute inflammatory responses in unvaccinated and vaccinated heifers inoculated with 10(4) or 10(6) cfu of E. coli. The rates computed for the cellular transit from the storage sites to the blood and from the blood to the milk were lower in cows receiving 10(4) cfu but increased at approximately 6 x 10(-6) and 30 x 10(-6) microL/cfu per h in nonvaccinated or vaccinated cows inoculated with 10(6) cfu, respectively. The cellular rates of bacterial killing were highest in unvaccinated cows ( approximately 400 x 10(-6) microL/cell per h) when compared with vaccinated cows (200 to 300 x 10(-6) microL/cell per h). A critical density of milk somatic cells at which bacteria density is constant was computed from the model at 2 x 10(6) cells/mL, and a one-way sensitivity analysis revealed that the changes in milk cellular densities were mostly sensitive to variations in the rate of bacterial killing and in the rate of production of inflammatory cells. A comparison of somatic cell count and antimicrobial susceptibility of subclinical mastitis pathogens in organic and conventional dairy herds.Bovine subclinical mastitis is the most important disease affecting dairy cows. The fluctuating increase in somatic cell count (SCC) that occurs causes major economic losses in dairy industry. This comparative study between conventional and organic dairy herds was conducted in the aim to better characterize which consequences might have different management practices on SCC but also on the frequency of pathogens isolated and their antimicrobial susceptibility. Four conventional and four organic herds, with bulk milk SCC >300x103cells/ml were selected, in which respectively 47 and 44 cows were investigated. Each quarter was sampled 3 times at 15 days interval for SCC, microbiological analysis and antimicrobial susceptibility. In both herd categories, major pathogens isolated were by order of importance Streptococcus uberis, Staphylococcus aureus and Streptococcus dysgalactiae with a great impact on SCC. Coagulase negative staphylococci were the most frequent minor germs and had a moderated but real impact on SCC. In certified organic dairy farms, the three most frequently isolated major pathogens were significantly more susceptible to antimicrobials in vitro. This study suggests that the limited use of antibiotics in organic dairy herds could explain, at least in part, the lower resistance obtained from analysed isolates. [less ▲]

Techniques of cost-effectiveness analyses were applied to determine whether or not it is economically efficient to measure the packed cell volume (PCV) on a colic horse before deciding on abdominal surgery. The effects of this decision of uncertainty on the estimated values of the parameters (probability of survival after surgery, surgery costs, PCV positive predictive value, and length of survival after surgery) were considered along with the monetary values of collecting additional information on those parameters. The effects of uncertainty on the incremental net benefits of each alternative were depicted by tornado diagrams, cost-effectiveness acceptability curves, and posterior probability distributions. The worth of additional information was computed as the expected values of perfect and sampling information. Given previously published results, the best PCV cut-off point to distinguish between survivors and nonsurvivors was at 44%. At this threshold, the most economically effective alternative is to measure PCV before surgery providing the owner is willing to pay less than €672 for each year the horse survives. Uncertainty on probability of survival after surgery largely influenced the decision whether or not to measure the PCV, but one should spend at most €381 in research to reduce this uncertainty. A study of postoperative survival of 500 colic horses would ensure an expected gain of €370 associated with a reduction in uncertainty. [less ▲]
