Animal cells are industrially cultivated inside stirred bioreactors to produce proteinic compounds. Due to the use of mild agitation conditions in order to limit mechanical constraints, the homogeneity of the culture medium can be far from perfect. This study has therefore two objectives. 1) the global characterization of the mixing via the mixing time 2) the local description of concentration fields. The mixing time is measured by conductimetry inside 20 L, 80 L, 600L tanks. The Grenville correlation is adjusted on these experimental measurements to improve the prediction of the mixing time during the scale-up of the process. The concentration fields are visualized by the Planar Laser Induced Fluorescence (P.L.I.F) technique in the 20 L tank. This part of the study is focused on the time evolution of the maximum value of the tracer concentration inside measurement planes and of the numerical distribution of theses concentration fields. Sludge liming is largely used in the wastewater industry. This operation, at first realized to stabilize sludge, has also proved to enhance dewatering and to improve the resulting cakes texture. Nevertheless, almost nothing is known about the impact of lime addition on sludge drying behavior. As drying can be considered as a necessary step after mechanical dewatering, it is crucial to evaluate the influence of the sludge pre-treatments on their drying kinetics. This study shows that both pre- and post-liming exert a positive effect on sludge convective drying behavior. The negative impact of sludge destructuring by pumping is also established. [less ▲]

Some structural and hydrodynamic properties of RCM-NCX-1116, a Ni-Cr metallic foam packing manufactured by Recemat International B.V (The Netherlands) are investigated using X-ray radiography and X-ray micro-tomography. Local values of porosity and pore diameters are measured on a foam sample 3D image obtained with a Skyscan-1172 high-resolution desk-top micro-CT system operated at 100 kV. Structural parameters computed on tomography images agree well with manufacturer data. Hydrodynamic tests are performed in a flat rectangular column packed with a sheet of metallic foam packing. Measurements are realized with a single-phase flow of liquid as well as with a counter-current flow of gas and liquid. Dynamic and static liquid holdup distributions and liquid radial spreading coefficient are measured on 2D radiographic images of the bed obtained with the high energy large scale ULG 420 kV scanner operated in the radiographic mode at 420 kV. Pressure drop measurements are successfully compared to experimental results reported in literature, as well as to the literature model predictions and to values computed using an in-house Lattice Boltzmann CFD simulation code. Packing simulations of generic, nonspherical pellets were performed and compared with experimental data sets obtained using X-ray Computerized tomography (CT). Two modified versions of what was previously a purely geometrical, digitally based packing algorithm were implemented. Both are aimed at incorporating the effects of particle interaction forces, one utilizing the distinct element method (DigiDEM) and the other an intermediate solution (collision-guided packing or DigiCGP). This article summarizes the models and the simulations performed using these two modified versions of DigiPac and, for model validation purposes, compares the predicted results with the corresponding X-ray tomographic scans of packed columns, in terms of bulk density, local packing density profiles, and pellet orientation distributions. For packed beds of relatively large and identical pellets, the simulation results indicate that particle-particle and particle-wall interactions cannot be ignored if realistic packing structures are to be obtained by simulation and even a simplistic treatment of these interactions can produce significantly more realistic packing structure than none at all. Experiments were conducted in a 0.25 m diameter bubble column to investigate the effect of animal cell culture medium composition on oxygen transfer rate. Air is used as the dispersed phase. The gas superficial velocity is varied between 0.4 and 2 cm/s (aeration rate ranging between 0.05 and 0.25 vvm) and the bubble column is thus operated in the homogenous regime. Aqueous solutions the composition (electrolyte, protein concentrations) of which mimics a mammalian cell culture medium) are used as the continuous phase. In particular, the effect on oxygen transfer rate of additive such as PVP, Pluronic F-68 which are usually used to protect cells against local hydrodynamic stresses induced by bubble coalescence and bursting is addressed. For each composition, the mass transfer coefficient is measured by the "gas in – gas out" method using a fast response dissolved oxygen probe. Its increase with gas velocity is measured. The liquid viscosity and surface tension are experimentally determined as a function of culture medium composition. Bubble size distribution is measured at different scales using three experimental techniques: gas disengagement technique, two dual optical probes and photography, which lead to results in good qualitative agreement one with each other. The integrated analysis of these data allows to decouple the effects of the different additive on coefficient kL and on the interfacial area, a. [less ▲]
